Recent experimental research on spill plume entrainment has developed a range of empirically-based formulae for smoke management design. These formulae form the spill plume entrainment model in B-RISK, a new fire risk zone model. This article describes the performance of B-RISK in predicting spill plume entrainment. Selected experimental data from the series of reduced-scale experiments used to form the new design formulae have been used for model validation, along with other full-scale experimental data from ‘hot smoke tests’ conducted to assess the performance of installed smoke management systems. B-RISK provides predictions of the plume clear-layer height that generally agree with experimental results within the range of experimental error. This gives confidence in its use to predict spill plume entrainment for smoke management design purposes.  相似文献

An ultralow volume fraction collection system referred to as nano fraction analysis chip technology (nanoFACT) is reported. The system collects 25-2500-nL fractions from 75-microm nanoLC columns into pipet tips at a user-defined, timed interval, typically one fraction every 15-120 s. Following collection, the fractions in the tip dry down naturally on their own in such a way as to create a concentrated band at the very end of the interior of the pipet tip. The fractions are then reconstituted directly in the pipet tips in approximately 250 nL of solvent prior to analysis. Because the chromatography and reconstitution solvent are independent, the reconstitution solvent can be selected to maximize ionization efficiency without compromising chromatography. In the infusion analysis of the nanoLC fractions, a low-flow electrospray chip is used which consists of 400 nozzles, each with an inner diameter of 2.5 microm and yielding flow rates of approximately 20 nL/min. Therefore, when reconstituted in 250 nL, each nanoLC fraction can be analyzed for over 10 min. This increase in analysis time allows for signal averaging, resulting in higher data quality, collision energy optimization, slower scanning techniques to be used, such as neutral loss and precursor ion scanning, higher resolution scans on FTMS instruments, and improved peptide quantitation. Furthermore, the nanoLC fractions could be archived in the pipet tips for analysis at a later date. Here, the advantages of nanoFACT are shown for phosphorylation analysis using bovine fetuin and glycosylation analysis using bovine ribonuclease B (RNase B). In the phosphorylation analysis, a comparison between conventional nanoLC and a nanoFACT analysis was performed. An MS/MS spectrum of a triply phosphorylated peptide, 313-HTFSGVApSVEpSpSSGEAFHVGK-333 could only be obtained using nanoFACT, not with nanoLC. Furthermore, spectral quality for the nanoFACT analysis was significantly improved over nanoLC. This was determined by comparing the number of diagnostic ions between the nanoFACT and nanoLC spectra, and it was found that the nanoFACT spectra contained a 19% or greater number of diagnostic ions for nonphosphorylated peptides and 55% or greater for phosphorylated peptides. For the glycosylation analysis, the glycosylation site of RNase B was fully characterized using 100 fmol of tryptic digest on a three-dimensional ion trap mass spectrometer.  相似文献

Two experiments tested whether differences in problem-solving strategies influence the ability of people to monitor their problem-solving effectiveness as measured by confidence judgments. On multiple choice problems, people tend to use either a constructive matching strategy, whereby they attempt to solve a problem before looking at the response options, or a response elimination strategy, whereby they work backward from response options trying to find one that fits as a solution. Constructive matching gives rise to different cues that may enhance confidence monitoring. Experiment 1 showed that spontaneous constructive matching in nonverbal spatial reasoning problems was associated with better confidence calibration and resolution than response elimination. We manipulated strategy in Experiment 2 by requiring constructive matching and found improved monitoring. The effect of sterols on the osmotic stability of mitochondrial and plasma membranes of yeast wild-types and mutants that are defective in ergosterol biosynthesis has been studied. Incorporation of the nonfungal sterol, cholesterol, into yeast membranes reduces membrane elasticity which is observed as an increased susceptibility to osmotic lysis. However, the wild-type and nystatin-resistant strains which were examined indicate that qualitative alterations in endogenously generated sterols do not affect resistance to swelling. Although these strains exhibit differences in membrane fluidity, which is influenced by the sterol accumulated by the organisms, the membrane stretching capacity shows no distinct dependence on sterol structure or bilayer fluidity.  相似文献

E. coli is an indicator of recent fecal contamination of freshwater beaches around the Great Lakes region. Elevated concentrations indicate that a fecal contamination has occurred, and that the risk for contact with fecal pathogenic organisms is heightened. The green algae, Cladophora, harbors populations of E. coli and potentially allows for prolonged survival and even replication of the bacterium in the lake environment. If presence of Cladophora mats on beaches is associated with persistence of E. coli in beach water, then E. coli would be a useful indicator organism only if pathogens also were able to survive and persist in the algae. This study utilized lab microcosms to study the persistence of E. coli, and of the fecal pathogens, Salmonella and Shigella, in lake water with and without the presence of Cladophora. E. coli was able to persist for extended periods in the presence of Cladophora (attached to algal mats for 45 days). Salmonella and Shigella, however, were unable to persist for this time period while in the presence of Cladophora (Salmonella attached to Cladophora was detectable for 10 days and Shigella was detectable for only 2 days). These data imply that E. coli is able to survive in the presence of Cladophora for greater times than are the fecal pathogens and that E. coli may not be an appropriate indicator organism for beaches with accumulations of algal material.  相似文献

Objective: Intervention effectiveness can potentially be affected by membership in different demographic subgroups (race, ethnicity, gender, age, and education level) or smoking behavior variables (time to first cigarette, longest previous quit attempt, number of attempts in the past year, number of cigarettes, and stage of change). Previous research on these 2 sets of variables has produced mixed results. Design: This secondary data analysis combined data from 5 effectiveness trials (a random-digit-dial sample [N = 1,358], members of an HMO [N = 207], parents of students recruited for a school-based study [N = 347], patients from an insurance provider list [N = 535], and employees [N = 175]) in which smokers were all proactively recruited from a defined population and all received the same expert system intervention. The intervention produced a consistent 22% to 26% point prevalence cessation rate across the 5 studies. Main Outcome Measures: The main outcome measures were 24-hr point prevalence, 7-day point prevalence, 30-day prolonged abstinence, and 6-month prolonged abstinence. Results: There were no significant differences in outcome across gender, race, and ethnicity subgroups. There were significant differences and small effect sizes for age and education subgroups. There were significant differences and large effect sizes for all 5 smoking behavior variables. Discussion: Demographic variables are static variables, whereas the smoking variables are more dynamic, that is, open to change. Given the dynamic nature of the smoking variables and the large effect sizes, interventions tailored on the smoking variables should be more successful. The relative influence of adolescents' closest friends and their friendship group on their cigarette smoking and alcohol use was investigated in a short-term, longitudinal study of 1,028 students in the 6th, 8th and 10th grades in 2 school systems. The amount of influence over the school year was modest in magnitude and came from the closest friend for initiation of cigarette and alcohol use. Only the friendship group use predicted transition into current cigarette use, whereas only the close friend use predicted transition into current alcohol use. Both group and close friends independently contributed to the prediction of adolescents' drinking to intoxication. No difference in the amount of influence, was found between stable and unstable close friendships or friendship groups; neither grade nor gender of the adolescents related to the amount of influence.
