The effect of progressive acetylation upon the conformation of the 11S globulin legumin from faba bean has been studied using chemical analysis, UV, fluorescence and CD spectroscopy, viscometry and analytical ultracentrifugation. The modification did not induce complete dissociation of the oligomeric protein. Only 30% of the protein was found to be a dissociated 3S subunit after excessive acetylation, whereas 70% was a dimeric legumin aggregate with a molecular mass of about 700 kDa. The aggregation of the highly modified legumin in high‐ionic‐strength buffer solution leads to soluble higher legumin oligomers. The acetylation resulted in a moderate molecular expansion of legumin due to a changed tertiary structure, whereas the far‐UV circular dichroism spectra did not provide definitive evidence of a decrease in domain‐stabilizing β‐sheet conformations in their secondary structure. © 2000 Society of Chemical Industry 相似文献

The article shows how rough sets can be applied to improve the classification ability of a hybrid pattern recognition system. The system presented here consists of a feature extractor based on a computer‐generated hologram (CGH) playing the role of a ring‐wedge detector. Features extracted by it are shift, rotation, and scale invariant. Although they can be optimized, no method has been proposed in the literature. This article presents an original method of optimizing the feature extraction abilities of a CGH. The method uses rough set theory (RST) to measure the amount of essential information contained in the feature vector. This measure is used to define an objective function in the optimization process. Since RST‐based factors are not differentiable, we use a nongradient approach for a search in the space of possible solutions. Finally, RST is used to determine decision rules for the classification of feature vectors. The alternative method of classification based on neural networks is also discussed. The whole method is illustrated by a system recognizing the class of speckle pattern images indicating the class of distortion of optical fibers. © 2001 John Wiley & Sons, Inc. 相似文献

As more floating farms are being developed, the wake interaction between multiple floating wind turbines (FWTs) is becoming increasingly relevant. FWTs have long natural periods in certain degrees of freedom, and the large‐scale movement of the wake, known as wake meandering, occurs at very low frequencies. In this study, we use FAST.Farm to simulate a two‐turbine case with three different FWT concepts: a semisubmersible (semi), a spar, and a tension leg platform (TLP), separated by eight rotor diameters in the wind direction. Since wake meandering varies depending on the environmental conditions, three different wind speeds (for all three concepts) as well as two different turbulence levels (for the semi) are considered. For the below‐rated wind speed, when wake meandering was most extreme, yaw motion standard deviations for the downstream semi were approximately 40% greater in high turbulence and over 100% greater in low turbulence when compared with the upstream semi. The low yaw natural frequency (0.01 Hz) of the semi was excited by meandering, while quasi‐static responses resulted in approximately 20% increases in yaw motion standard deviations for the spar and TLP. Differences in fatigue loading between the upstream and downstream turbines for the mooring line tension and tower base fore‐aft bending moment mostly depended on the velocity deficit and were not directly affected by meandering. However, wake meandering did affect fatigue loading related to the tower top yaw moment and the blade root out‐of‐plane moment. 相似文献

Membrane processes are able to separate liquid mixtures as well as mixtures of gases or vapours according to the sizes, solubilities, rates of diffusion or charges of the components. A separation is also possible using membranes in which the transport is coupled to a specific chemical reaction between the permeand and the membrane (facilitated or carrier mediated transport). This paper discusses aspects related to carrier‐mediated separation processes such as the transport mechanisms, the choice of appropriate carriers, the incorporation of carriers in the polymeric membrane material, the coating of porous support membranes with thin selective layers of the carrier‐polymer and, finally, the choice of appropriate parameters for the separation process. The separation of gas mixtures by carrier‐membranes appears very promising from a technical point of view. Experimental membrane samples often show excellent separation factors, but they have not met the permeability requirements for an economic use so far. 相似文献

Tepotinib is a novel tyrosine kinase inhibitor recently approved for the treatment of non-small cell lung cancer (NSCLC). In this study, we evaluated the tepotinib’s potential to perpetrate pharmacokinetic drug interactions and modulate multidrug resistance (MDR). Accumulation studies showed that tepotinib potently inhibits ABCB1 and ABCG2 efflux transporters, which was confirmed by molecular docking. In addition, tepotinib inhibited several recombinant cytochrome P450 (CYP) isoforms with varying potency. In subsequent drug combination experiments, tepotinib synergistically reversed daunorubicin and mitoxantrone resistance in cells with ABCB1 and ABCG2 overexpression, respectively. Remarkably, MDR-modulatory properties were confirmed in ex vivo explants derived from NSCLC patients. Furthermore, we demonstrated that anticancer effect of tepotinib is not influenced by the presence of ABC transporters associated with MDR, although monolayer transport assays designated it as ABCB1 substrate. Finally, tested drug was observed to have negligible effect on the expression of clinically relevant drug efflux transporters and CYP enzymes. In conclusion, our findings provide complex overview on the tepotinib’s drug interaction profile and suggest a promising novel therapeutic strategy for future clinical investigations. 相似文献

To protect the gill capillaries from high systolic pulse pressure, the fish heart contains a compliant non-contractile chamber called the bulbus arteriosus which is part of the outflow tract (OFT) which extends from the ventricle to the ventral aorta. Thermal acclimation alters the form and function of the fish atria and ventricle to ensure appropriate cardiac output at different temperatures, but its impact on the OFT is unknown. Here we used ex vivo pressure–volume curves to demonstrate remodelling of passive stiffness in the rainbow trout (Oncorhynchus mykiss) bulbus arteriosus following more than eight weeks of thermal acclimation to 5, 10 and 18°C. We then combined novel, non-biased Fourier transform infrared spectroscopy with classic histological staining to show that changes in compliance were achieved by changes in tissue collagen-to-elastin ratio. In situ gelatin zymography and SDS-PAGE zymography revealed that collagen remodelling was underpinned, at least in part, by changes in activity and abundance of collagen degrading matrix metalloproteinases. Collectively, we provide the first indication of bulbus arteriosus thermal remodelling in a fish and suggest this remodelling ensures optimal blood flow and blood pressure in the OFT during temperature change. 相似文献

The most important steps and decisions in the use of CFD are presented from the perspective of the process engineer.
