Engorged females and 1- to 3-d-old eggs of the lone star tick, Amblyomma americanum (L), were exposed to 9 treatments of pyriproxyfen (4, 8, and 16 micrograms/cm2) in glass vials at exposure periods of 7 d, 14 d, and continuous at each dosage level. Treatment of newly engorged females did not affect the number of females ovipositing, but the number of eggs oviposited decreased as dosage and exposure time increased. Complete inhibition of egg hatch occurred at all treatment levels except that of the lowest dosage and exposure time where 99.9% inhibition was observed. Egg masses from treated females contained eggs that turned a dark amber color and imploded, retained normal shape and color without visible evidence of embryogenesis, or had developed embryos that were unable to emerge. Treatments of 1-to 3-d-old eggs were effective in reducing hatch and larval survivorship to 25 d after emergence only at the continuous exposures of dosages of 4, 8, and 16 micrograms/cm2. Larvae emerging from eggs treated at the lower dosage rates of 0.2 and 0.02 microgram/cm2 were as successful in feeding on chickens and subsequently molting as acetone treated control ticks.  相似文献

A new member of the tumor necrosis factor (TNF) cytokine family, designated Apo-2 ligand (Apo-21) [1] or TRAIL [2], has been shown recently to induce apoptosis in various tumor cell lines; however, its biological role is unknown. Here, we show that Apo-21, activated apoptosis in T-cell-enriched cultures of peripheral blood lymphocytes stimulated by interleukin-2 (IL-2), but not in unstimulated cells. This finding suggests that, like Fas/Apo-1 ligand and TNF [3-5], Apo-2L may play a role in regulating post-stimulation apoptosis of mature lymphocytes. Studies on the mechanism of Apo-2L action demonstrated marked membrane blebbing, a hallmark of apoptosis, within a few minutes of the addition of Apo-2L to tumor cells. Ectopic expression of a dominant negative mutant of FADD, a cytoplasmic protein that mediates death signalling by Fas/Apo-1 and by TNF receptor type 1 (TNFR1) [6-9], inhibited the induction of apoptosis by anti-Fas/Apo-1 antibody, but had little effect on Apo-2L function. In contrast, expression of CrmA, a cowpox virus-derived inhibitor of the Ced-2-like proteases ICE [10] and CPP32/Yama [11,12], blocked the induction of apoptosis by either Apo-2L or anti-Fas/Apo-1 antibody. These results suggest that Apo-2L activates a rapid, FADD-independent pathway to trigger a cell-death programme that requires the function of cysteine proteases such as ICE or CPP32/Yama.  相似文献

In the past, the diagnosis of "growing skull fracture" or "diastatic fracture" has included a subset of injuries better referred to as "cranial burst fracture." Cranial burst fracture, typically associated with severe injury in infants less than 1 year of age, is a closed, widely diastatic skull fracture accompanied by acute cerebral extrusion outside the calvarium. We treated 11 such infants at the LeBonheur Children's Medical Center and 2 at the Children's National Medical Center from January 1986 through December 1994. Infants ranged in age from 1 to 17 months, with an average age of 5.7 months. All presented with marked scalp swelling and a Glasgow Coma Scale score of 10 or less. Twelve had a history consistent with severe injury (motor vehicle accident, 7, abuse 5). The cause of injury in one patient remains unproven. Surgery (reduction of herniated cerebral tissue, repair of large dural laceration, and cranioplasty) was usually performed within 10 days of injury, a time period long enough to assure hemodynamic stability and resolution of acute cerebral swelling, yet sufficiently brief to avoid the chronic changes (scarring, parasitization of scalp vessels by damaged cortex) associated with a "growing skull fracture." Prompt repair of cranial burst fracture may prevent ongoing brain injury such as has been neuropathologically demonstrated in patients with "growing skull fracture." Magnetic resonance imaging establishes the diagnosis of cranial burst fracture in equivocal cases, rendering unnecessary a "waiting period" to see if scalp swelling resolves. Our experience, together with information in the neuropathological and neurosurgical literature, suggests that cranial burst fracture is associated with severe trauma, requires expeditious treatment, and has been underdiagnosed in the past, leading to "growing skull fracture," a condition requiring more extensive surgery.  相似文献

-1We address the problem of visual domain adaptation for transferring object models from one dataset or visual domain to another. We introduce a unified flexible model for both supervised and semi-supervised learning that allows us to learn transformations between domains. Additionally, we present two instantiations of the model, one for general feature adaptation/alignment, and one specifically designed for classification. First, we show how to extend metric learning methods for domain adaptation, allowing for learning metrics independent of the domain shift and the final classifier used. Furthermore, we go beyond classical metric learning by extending the method to asymmetric, category independent transformations. Our framework can adapt features even when the target domain does not have any labeled examples for some categories, and when the target and source features have different dimensions. Finally, we develop a joint learning framework for adaptive classifiers, which outperforms competing methods in terms of multi-class accuracy and scalability. We demonstrate the ability of our approach to adapt object recognition models under a variety of situations, such as differing imaging conditions, feature types, and codebooks. The experiments show its strong performance compared to previous approaches and its applicability to large-scale scenarios.  相似文献

The model enzyme β‐galactosidase was entrapped in chitosan gel beads and tested for hydrolytic activity and its potential for application in a packed‐bed reactor. The chitosan beads had an enzyme entrapment efficiency of 59% and retained 56% of the enzyme activity of the free enzyme. The Michaelis constant (Km) was 0.0086 and 0.011 μmol/mL for the free and immobilized enzymes, respectively. The maximum velocity of the reaction (Vmax) was 285.7 and 55.25 μmol mL?1 min?1 for the free and immobilized enzymes, respectively. In pH stability tests, the immobilized enzyme exhibited a greater range of pH stability and shifted to include a more acidic pH optimum, compared to that of the free enzyme. A 2.54 × 16.51‐cm tubular reactor was constructed to hold 300 mL of chitosan‐immobilized enzyme. A full‐factorial test design was implemented to test the effect of substrate flow (20 and 100 mL/min), concentration (0.0015 and 0.003M), and repeated use of the test bed on efficiency of the system. Parameters were analyzed using repeated‐measures analysis of variance. Flow (p < 0.05) and concentration (p < 0.05) significantly affected substrate conversion, as did the interaction progressing from Run 1 to Run 2 on a bed (p < 0.05). Reactor stability tests indicated that the packed‐bed reactor continued to convert substrate for more than 12 h with a minimal reduction in conversion efficiency. © 2003 Wiley Periodicals, Inc. Organic semiconductors can be employed as the active layer in accumulation mode organic electrochemical transistors (OECTs), where redox stability in aqueous electrolytes is important for long‐term recordings of biological events. It is observed that alkoxy‐benzo[1,2‐b:4,5‐b′]dithiophene (BDT) copolymers can be extremely unstable when they are oxidized in aqueous solutions. The redox stability of these copolymers can be improved by molecular design of the copolymer where it is observed that the electron rich comonomer 3,3′‐dimethoxy‐2,2′‐bithiophene (MeOT2) lowers the oxidation potential and also stabilizes positive charges through delocalization and resonance effects. For copolymers where the comonomers do not have the same ability to stabilize positive charges, irreversible redox reactions are observed with the formation of quinone structures, being detrimental to performance of the materials in OECTs. Charge distribution along the copolymer from density functional theory calculations is seen to be an important factor in the stability of the charged copolymer. As a result of the stabilizing effect of the comonomer, a highly stable OECT performance is observed with transconductances in the mS range. The analysis of the decomposition pathway also raises questions about the general stability of the alkoxy‐BDT unit, which is heavily used in donor–acceptor copolymers in the field of photovoltaics.  相似文献

We create polymer liquid crystals (PLCs) on a computer and subject them to constant-force tensile deformations. Molecular dynamics simulation procedure is used and graphical models and animations of crack formation and propagation as a function of time are generated. Special attention is given to realistic rigid LC island geometry and the island spatial distribution in the material. Internal fracture processes can now be easily studied. As a result of the numerous animations recorded, clear patterns in PLC crack initiation and propagation emerge. With the help of the animations, the structure of PLCs may be understood without having to resort to costly laboratory experimentation. A goal of secondary organic aerosol (SOA) experiments performed in smog chambers is to determine the condensation of SOA onto suspended particles. Complicating the calculation of the condensation rate are uncertainties in particle wall-loss rates. Wall-loss rates generally depend on particle size, turbulence in the bag, the size and shape of the bag, and particle charge. In analyzing smog-chamber data, some or all of the following assumptions are commonly made regarding the first-order wall-loss rate constant: (a) that it is constant during an experiment; (b) that it is constant between experiments; and (c) that it is not a strong function of particle size for the relatively narrow size distributions in smog chamber experiments. Each of these assumptions may not be justified in some circumstances. We present the development and evaluation of the Aerosol Parameter Estimation (APE) model. APE is an inverse model that solves the aerosol general dynamic equation to determine best estimates for the size-dependent condensation rate and size-dependent wall-loss rate as a function of time. Size distribution measurements from a Scanning Mobility Particle Sizer (SMPS) provide time boundary conditions that constrain the general dynamic equation. The APE model is tested using data from a smog chamber experiment with dry ammonium sulfate particles in which no condensation occurred. Finally, we assess the variability in predicted SOA production between different wall-loss correction methods for relatively-fast-chemistry limonene-ozonolysis experiments and relatively-slow-chemistry toluene-oxidation experiments. In the fast limonene experiments, wall-loss correction methods agree within 10% for SOA production, and in the slow toluene experiments, wall-loss correction methods disagree up to a factor of 2.  相似文献
