Solutions of partial differential equations could exhibit a multiscale behavior. Standard discretization techniques are constraints to mesh up to the finest scale to predict accurately the response of the system. The proposed methodology is based on the standard proper generalized decomposition rationale; thus, the PDE is transformed into a nonlinear system that iterates between microscale and macroscale states, where the time coordinate could be viewed as a 2D time, representing the microtime and macrotime scales. The macroscale effects are taken into account because of an FEM-based macrodiscretization, whereas the microscale effects are handled with unidimensional parent spaces that are replicated throughout the domain. The proposed methodology can be seen as an alternative route to circumvent prohibitive meshes arising from the necessity of capturing fine-scale behaviors.  相似文献

We report an experimental investigation of the electrokinetic properties and size variations of four F-specific bacteriophages of the types MS2, GA, Qbeta and SP (21-30 nm in diameter) over a broad range of pH values (1.5-7.5) and NaNO3 electrolyte concentrations (1-100 mM). The results obtained by dynamic light scattering show that the aggregation of SP and GA particles takes place over the whole range of pH and ionic strength conditions examined. For MS2 phages, the aggregation of MS2 particles is not observed for pH higher than the isoelectric point (pI) and large ionic strengths for which interparticular repulsive electrostatic interactions are however expected to be sufficiently screened. Aggregation of the MS2 phages, dispersed in 1 and 100 mM electrolyte concentration, occurs at pH 4, which basically corresponds to the pI as determined by electrophoresis measurements. The Qbeta particles suspended in solutions of low electrolyte concentrations aggregate at low pH values (pI approximately 3) and, unlike MS2, at large ionic strengths over the whole range of pH conditions considered in this study. These elements allow the determination of the hydrophobic sequence for the four phages SP approximately GA>Qbeta>MS2. Close inspection of the electrokinetic results reveals small to significant variations of the pI values-depending on the phage considered-with respect to the concentration of indifferent NaNO3 electrolyte. This indicates that features other than chemical and electrostatic in nature play a key role in determining the pI and more generally the electrophoretic mobility mu of viral particles. A qualitative interpretation is given and is based on the consideration of inner electro-osmotic flow within the isolated or aggregated particles. The impact of the flow properties within the particles is further in agreement with recent theoretical formalism developed for the electrokinetics of soft multiplayer particles, the phages analyzed here being some illustrative examples. The determination and qualitative interpretation of the surface properties of the viral particles as reported in the current study are commented within the context of water treatment especially concerning viral removal by membrane filtration processes.  相似文献

A line of mice deficient in vitamin D binding protein (DBP) was generated by targeted mutagenesis to establish a model for analysis of DBP's biological functions in vitamin D metabolism and action. On vitamin D-replete diets, DBP-/- mice had low levels of total serum vitamin D metabolites but were otherwise normal. When maintained on vitamin D-deficient diets for a brief period, the DBP-/-, but not DBP+/+, mice developed secondary hyperparathyroidism and the accompanying bone changes associated with vitamin D deficiency. DBP markedly prolonged the serum half-life of 25(OH)D and less dramatically prolonged the half-life of vitamin D by slowing its hepatic uptake and increasing the efficiency of its conversion to 25(OH)D in the liver. After an overload of vitamin D, DBP-/- mice were unexpectedly less susceptible to hypercalcemia and its toxic effects. Peak steady-state mRNA levels of the vitamin D-dependent calbindin-D9K gene were induced by 1,25(OH)2D more rapidly in the DBP-/- mice. Thus, the role of DBP is to maintain stable serum stores of vitamin D metabolites and modulate the rates of its bioavailability, activation, and end-organ responsiveness. These properties may have evolved to stabilize and maintain serum levels of vitamin D in environments with variable vitamin D availability.  相似文献

Hepatitis C virus (HCV) is the major cause of non-A, non-B hepatitis worldwide. Current treatments are not curative for most infected individuals, and there is an urgent need for both novel therapeutic agents and small-animal models which can be used to evaluate candidate drugs. A small-animal model of HCV gene expression was developed with recombinant vaccinia virus vectors. VHCV-IRES (internal ribosome entry site) is a recombinant vaccinia viral vector containing the HCV 5' nontranslated region (5'-NTR) and a portion of the HCV core coding region fused to the firefly luciferase gene. Intraperitoneal injection of VHCV-IRES produced high levels of luciferase activity in the livers of BALB/c mice. Antisense oligonucleotides complementary to the HCV 5'-NTR and translation initiation codon regions were then evaluated for their effects on the expression of these target HCV sequences in BALB/c mice infected with the vaccinia virus vector. Treatment of VHCV-IRES-infected mice with 20-base phosphorothioate oligonucleotides complementary to the sequence surrounding the HCV initiation codon (nucleotides 330 to 349) specifically reduced luciferase expression in the livers in a dose-dependent manner. Inhibition of HCV reporter gene expression in this small-animal model suggests that antisense oligonucleotides may provide a novel therapy for treatment of chronic HCV infection.  相似文献

Two studies were performed to examine the influence of fasting and food intake on the absorption and retention of nickel added to drinking water and to determine if nickel sensitization played any role in this regard. First, eight nonallergic male volunteers fasted overnight before being given nickel in drinking water (12 micrograms Ni/kg) and, at different time intervals, standardized 1400-kJ portions of scrambled eggs. When nickel was ingested in water 30 min or 1 h prior to the meal, peak nickel concentrations in serum occurred 1 h after the water intake, and the peak was 13-fold higher than the one seen 1 h after simultaneous intake of nickel-containing water and scrambled eggs. In the latter case, a smaller, delayed peak occurred 3 h after the meal. Median urinary nickel excretion half-times varied between 19.9 and 26.7 h. Within 3 days, the amount of nickel excreted corresponded to 2.5% of the nickel ingested when it was mixed into the scrambled eggs. Increasing amounts were excreted as the interval between the water and the meal increased, with 25.8% of the administered dose being excreted when the eggs were served 4 h prior to the nickel-containing drinking water. In the second experiment, a stable nickel isotope, 61Ni, was given in drinking water to 20 nickel-sensitized women and 20 age-matched controls, both groups having vesicular hand eczema of the pompholyx type. Nine of 20 nickel allergic eczema patients experienced aggravation of hand eczema after nickel administration, and three also developed a maculopapular exanthema. No exacerbation was seen in the control group. The course of nickel absorption and excretion in the allergic groups did not differ and was similar to the pattern seen in the first study, although the absorption in the women was less. A sex-related difference in gastric emptying rates may play a role. Thus, food intake and gastric emptying are of substantial significance for the bioavailability of nickel from aqueous solutions.  相似文献

SPICA, a Japanese led mission, is part of the JAXA future science program and is planned for launch in 2018. SPICA will perform imaging and spectroscopic observations in the mid- and far-IR waveband, and is developing instrumentation spanning the 5–400 μm range. The SPICA payload features several candidate instruments, some of them requiring temperature down to 50 mK. This is currently the case for SAFARI, a core instrument developed by a European-based consortium, and BLISS proposed by CALTECH/JPL in the US.SPICA’s distinctive feature is to actively cool its telescope to below 6 K. In addition, SPICA is a liquid cryogen free satellite and all the cooling will be provided by radiative cooling (L2 orbit) down to 30 K and by mechanical coolers for lower temperatures. The satellite will launch warm and slowly equilibrate to its operating temperatures once in orbit. This warm launch approach makes it possible to eliminate a large liquid cryogen tank and to use the mass saved to launch a large diameter telescope (3.2 m). This 4 K cooled telescope significantly reduces its own thermal radiation, offering superior sensitivity in the infrared region.The cryogenic system that enables this warm launch/cooled telescope concept is a key issue of the mission. This cryogenic chain features a number of cooling stages comprising passive radiators, Stirling coolers and several Joule Thomson loops, offering cooling powers at typically 20, 4.5, 2.5 and 1.7 K. The SAFARI and BLISS detectors require cooling to temperatures as low as 50 mK. The instrument coolers will be operated from these heat sinks. They are composed of a small demagnetization refrigerator (ADR) pre cooled by either a single or a double sorption cooler, respectively for SAFARI and BLISS. The BLISS cooler maintains continuous cooling at 300 mK and thus suppresses the thermal equilibrium time constant of the large focal plane.These hybrid architectures allow designing low weight coolers able to reach 50 mK. Because the sorption cooler has extremely low mass for a sub-Kelvin cooler, it allows the stringent mass budget to be met. These concepts are discussed in this paper.  相似文献

CEA/SBT is currently developing a 2.5 K-50 mK cooling solution composed of a small demagnetization refrigerator (ADR) precooled by a sorption cooler, equivalent to the high temperature stage of a two-stage ADR system. Thanks to the use of this dual technology, a low weight cooler able to reach 50 mK with a heat sink up to 2.5 K can be designed. Because the sorption cooler is probably the lightest solution to produce sub-Kelvin temperatures, these developments allow us to propose a solution to face the drastic reduction in the mass budget of space missions like SPICA or IXO. The European Space Agency (ESA) is funding the development of an engineering model able to produce 1 μW net heat lift at 50 mK. It is sized so that the sorption cooler provides an additional 10 μW at 300 mK. The ESA main requirements are an autonomy of more than 24 h and a recycling time smaller than 8 h. We present the design of the system able to meet these requirements as well as the expected performances and preliminary measurements.  相似文献

The aim of this study was to analyze the impact of the water content of wood on the concentrations of volatile compounds which can be extracted after heat treatments. Head Space-Solid Phase Micro Extraction Gas Chromatography coupled to Mass Spectrometry (HS-SPME GC–MS) has been used to compare the concentrations of six aroma compounds (vanillin, furfural, eugenol, guaïacol and cis- and trans-whisky lactones) in hydroalcoholic extracts of heated oak wood samples either previously soaked in hot water or not. Except for eugenol, concentrations of extracted aromas appeared to be lower in soaked woods than in dry woods for temperatures up to 200 °C. If a delaying effect of water could explain such overall lower extracted concentrations from soaked woods, a PCA analysis revealed that for the longer duration (25 min of heat treatment), the adsorbed water could promote a higher impact of furfural, eugenol and both whisky lactones on the composition of hydroalcoholic extracts, suggesting that alternative mechanisms of thermal modifications of the wood macromolecular network could exist at high temperatures in presence of adsorbed water.  相似文献

ABSTRACT

Straight- and multigrade fluids were evaluated in a hydraulic dynamometer that incorporated a pressure-compensated axial piston pump and a fixed displacement axial piston motor. Pump, motor, pressure compensator, and directional control valve internal flow losses were determined under various conditions of pressure, speed, and temperature. Fluid samples were collected before and at various times during the dynamometer experiments, and viscosity measurements were performed to probe for correlations between viscosity, operating time, and system leakage flow losses. The low shear rate viscosities of the multigrade fluids decreased linearly throughout the duration of testing due to polymer degradation. However, system flow losses did not exhibit a statistically significant increase as the multigrade fluids sheared down. The fluids were also characterized by their permanent viscosity loss produced in sonic shear and tapered bearing tests and by their temporary shear thinning measured in an ultra-high-shear viscometer at several temperatures. The effects of these viscous properties were analyzed using an empirical model to identify which measures of viscosity were most correlated with flow loss. The results suggested that the relative contributions of temporary and permanent viscosity loss change as the fluid is used. Further, analysis of torque loss and input power revealed that input power and losses are more useful indicators of the effect of fluids on hydraulic system performance than pump efficiency.  相似文献

Silverpoint drawings from the Renaissance are among the most precious and rarest treasures of graphical art. Our research group is particularly interested in the analysis of silverpoint drawings by Albrecht Dürer (1471-1528). A very sensitive and non-destructive analytical method, either spatially resolved synchrotron-radiation induced X-ray fluorescence (SY-XRF) or proton-induced X-ray emission (PIXE), is needed to determine the chemical composition of the very faint silver marks on such drawings. Dürer drawings from the collection of the Albertina, Vienna, were analyzed to amend existing data on Dürer drawings. For this purpose an external-beam PIXE setup was installed at the Vienna Environmental Research Accelerator (VERA). It allows to analyze a spot of ∼0.15 mm on the object in air with 3 MeV protons, and to detect the emitted X-rays that are characteristic for the chemical composition with very good sensitivity and without harming the precious objects. After successful measurements on artificial test samples, four original silverpoint drawings were investigated: two portraits from Albrecht Dürer’s very early period (self-portrait and portrait of his father) and two drawings from Dürer’s sketch book of his travel to the Netherlands 1520/21.  相似文献
