Intravenously injected nanoparticles can be delivered to skeletal muscle through capillary pores created by the activation of microbubbles with ultrasound; however, strategies that utilize coinjections of free microbubbles and nanoparticles are limited by nanoparticle dilution in the bloodstream. Here, improvement in the delivery of fluorescently labeled ≈150 nm poly(lactic-co-glycolic acid) nanoparticles to skeletal muscle is attempted by covalently linking them to albumin-shelled microbubbles in a composite agent formulation. Studies are performed using an experimental model of peripheral arterial disease, wherein the right and left femoral arteries of BalbC mice are surgically ligated. Four days after arterial ligation, composite agents, coinjected microbubbles and nanoparticles, or nanoparticles alone are administered intravenously and 1 MHz pulsed ultrasound was applied to the left hindlimb. Nanoparticle delivery was assessed at 0, 1, 4, and 24 h post-treatment by fluorescence-mediated tomography. Within the coinjection group, both microbubbles and ultrasound are found to be required for nanoparticle delivery to skeletal muscle. Within the composite agent group, nanoparticle delivery is found to be enhanced 8- to 18-fold over 'no ultrasound' controls, depending on the time of measurement. A maximum of 7.2% of the initial nanoparticle dose per gram of tissue was delivered at 1 hr in the composite agent group, which was significantly greater than in the coinjection group (3.6%). It is concluded that covalently linking 150 nm-diameter poly(lactic-co-glycolic acid) nanoparticles to microbubbles before intravenous injection does improve their delivery to skeletal muscle.  相似文献

Mobility-selected fractal and non-fractal soot particles (mobility diameters d m = 135 to 310 nm) were produced at three controlled fuel equivalence ratios (φ = 2.1, 3.5, and 4.5) by an ethylene/oxygen flame. Oleic acid (liquid) and anthracene (solid) coatings were alternately applied to the particles and removed. Simultaneous measurements with an Aerodyne aerosol mass spectrometer and a scanning mobility particle sizer yielded the particle mass, volume, density, composition, dynamic shape factor, fractal dimension, surface area, and the size and number of the primary spherules forming the fractal aggregate. For a given φ, the diameters of the primary spherules are approximately the same, independent of d m (15 nm, 35 nm, and 55 nm for φ = 2.1, 3.5, and 4.5, respectively). As the coating thickness on a particle increases, the dynamic shape factor decreases but d m remains constant until the particle reaches a spherical (for oleic acid) or non-fractal but irregular (for anthracene) shape. Under some conditions, liquid oleic acid coating causes the internal BC framework to rearrange into a more compact configuration. The surface area of fractal particles is up to 2.4 times greater than that of a sphere with the same d m . Using the surface area determinations, the time for a fractal particle to obtain a monolayer of coating material is compared to that of spheres. If it is assumed that the fractal particle is a sphere with the same d m as the fractal particle, the monolayer coating time is underestimated by a factor of up to 1.7.  相似文献

This study presents a distributed H-infinity control method for uncertain platoons with dimensionally and structurally unknown interaction topologies provided that the associated topological eigenvalues are bounded by a predesigned range.With an inverse model to compensate for nonlinear powertrain dynamics, vehicles in a platoon are modeled by third-order uncertain systems with bounded disturbances. On the basis of the eigenvalue decomposition of topological matrices, we convert the platoon system to a norm-bounded uncertain part and a diagonally structured certain part by applying linear transformation. We then use a common Lyapunov method to design a distributed H-infinity controller. Numerically, two linear matrix inequalities corresponding to the minimum and maximum eigenvalues should be solved. The resulting controller can tolerate interaction topologies with eigenvalues located in a certain range. The proposed method can also ensure robustness performance and disturbance attenuation ability for the closed-loop platoon system. Hardware-in-the-loop tests are performed to validate the effectiveness of our method.  相似文献

The potential short-term (10(2) years) biogeochemical and ecological effects of diverting stream water (pH 4.9-6.7) into a limed, flooded tailings impoundment (pH 8-12) were studied by combining geochemical and biological data. In the long-term perspective, the successional development of lakes was used as a natural analogue. Based on the vertical distribution of temperature and total dissolved solids (TDS < 0.22 microm), the impoundment can be characterised as a continuous/discontinuous cold polymictic lake, with holomictic summer circulation. A re-inoculation study indicated that the growth of autotrophic, aerobic bacteria (presumably Acidithiobacillus ferrooxidans) presently is inhibited by the high pH in the impoundment. In a short-term perspective, termination of liming and diversion of stream water into the impoundment will result in a complex interplay between physical, biogeochemical and ecological effects. A reduced vertical mixing of the approximately 2-m-deep water column, dissolution of calcite and gypsum (compounds of a sludge formed in the impoundment) and an enhanced microbiological activity are major expected effects. The dissolution of calcite may act as a pH buffer and result in metal remobilisation from the sludge. Excluding autochthonous organic matter produced in the impoundment, streamwater input of suspended matter and formation of settling flocculants are expected to result in a sediment accumulation rate of approximately 1.5 mg cm(-2) year(-1) (1.6-3.3 cm/10(2) years). Settling allochthonous organic C (0.15-0.30 mg C cm(-2) year(-1)) may serve as an oxygen barrier and as a reservoir of organic compounds capable of driving redox reactions. In a long-term perspective, a hydroseral development into a wetland/peatland can be expected, with a bog lake, poor fen or flat bog as final stage. This development presupposes a decreasing pH when liming is terminated and stream water is diverted into the impoundment. It also assumes that the impoundment will be similar to an acidified lake, and that the succession is driven by Sphagnum colonizing the impoundment. If the hydrological conditions/water level is affected (e.g., by climatic changes or a dam failure), a terrestrialization culminating in coniferous forest on peat soil may occur.  相似文献

Thirty-six strains of aerobic endospore-forming bacteria confirmed by polyphasic taxonomic methods to belong to Bacillus amyloliquefaciens, Bacillus cereus, Bacillus licheniformis, Bacillus megaterium, Bacillus subtilis (including Bacillus niger and Bacillus globigii), Bacillus sphaericus, and Brevi laterosporus were grown axenically on nutrient agar, and vegetative and sporulated biomasses were analyzed by Curie-point pyrolysis mass spectrometry (PyMS) and diffuse reflectance-absorbance Fourier-transform infrared spectroscopy (FT-IR). Chemometric methods based on rule induction and genetic programming were used to determine the physiological state (vegetative cells or spores) correctly, and these methods produced mathematical rules which could be simply interpreted in biochemical terms. For PyMS it was found that m/z 105 was characteristic and is a pyridine ketonium ion (C6H3ON+) obtained from the pyrolysis of dipicolinic acid (pyridine-2,6-dicarboxylic acid; DPA), a substance found in spores but not in vegetative cells; this was confirmed using pyrolysis-gas chromatography/mass spectrometry. In addition, a pyridine ring vibration at 1447-1439 cm-1 from DPA was found to be highly characteristic of spores in FT-IR analysis. Thus, although the original data sets recorded hundreds of spectral variables from whole cells simultaneously, a simple biomarker can be used for the rapid and unequivocal detection of spores of these organisms.  相似文献

This study compares the optical coefficients of size-selected soot particles measured at a wavelength of 870 nm with those predicted by three theories, namely, Rayleigh-Debye-Gans (RDG) approximation, volume-equivalent Mie theory, and integral equation formulation for scattering (IEFS). Soot particles, produced by a premixed ethene flame, were size-selected using two differential mobility analyzers in series, and their scattering and absorption coefficients were measured with nephelometry and photoacoustic spectroscopy. Scanning electron microscopy and image processing techniques were used for the parameterization of the structural properties of the fractal-like soot aggregates. The aggregate structural parameters were used to evaluate the predictions of the optical coefficients based on the three light-scattering and absorption theories. Our results show that the RDG approximation agrees within 10% with the experimental results and the exact electromagnetic calculations of the IEFS theory. Volume-equivalent Mie theory overpredicts the experimental scattering coefficient by a factor of approximately 3.2. The optical coefficients predicted by the RDG approximation showed pronounced sensitivity to changes in monomer mean diameter, the count median diameter of the aggregates, and the geometric standard deviation of the aggregate number size distribution.  相似文献

The preparation of a complete and concise project document is a critical issue facing professionals, developers, and contractors working in the building industry. The lack of an accurate, fast, and effective method for estimating the bill of quantities creates problems during the design, tender, and construction supervision of construction projects for designers and contractors practicing in Saudi Arabia. The capability of a computer-aided drafting (CAD) based system to interface with local data, either from the designers and contractors, or from manufacturers and commercial libraries, that allows for the estimation of the building bill of quantities in a more rigorous way is also lacking. The availability of an accurate and automated method to produce the bill of quantities from such libraries creates a benchmark in professional construction practice. This paper presents a computer program developed by the writer. This program is a third-party achievement that automates the aforementioned task and complements the standard AutoCAD system, which is not capable of extracting the project data pertinent to performing an automated bill of quantities. The program is written in C-language and has the capability to interface with other databases possessed by the designer or contractor. The AutoCAD has been taken as the base software because this is a cost-effective CAD software. Though Archicad, Design + available softwares can perform the automated quantity survey, it should be remembered that these packages are very expensive and used by fewer people compared with AutoCAD, which is used very widely.  相似文献

This paper describes Menv, an environment for the development of interactive three-dimensional modelling and animation systems. Menv supports both interactive and procedural techniques. Systems built upon Menv are implemented as a set of independent, co-operating tools operating upon a shared workspace.
