Immunocytochemistry was used to investigate the localization of F1 ATPase in mitochondria of cryosections of adult mouse heart muscle cells. The initial aldehyde fixation was the only denaturation step for antigens. The fine structure was preserved with contrast enhancement as the sections were maintained hydrated, with the advantage that the entire procedure is completed in one working day. The reaction was highly specific, and entire mitochondria were labeled with the Protein A-gold complex. A new analytical technique, electron spectroscopic imaging (ESI), contributed to a better visualization of the localization of the F1 factor.  相似文献

Na‐ion cointercalation in the graphite host structure in a glyme‐based electrolyte represents a new possibility for using carbon‐based materials (CMs) as anodes for Na‐ion storage. However, local microstructures and nanoscale morphological features in CMs affect their electrochemical performances; they require intensive studies to achieve high levels of Na‐ion storage performances. Here, pyrolytic carbon nanosheets (PCNs) composed of multitudinous graphitic nanocrystals are prepared from renewable bioresources by heating. In particular, PCN‐2800 prepared by heating at 2800 °C has a distinctive sp2 carbon bonding nature, crystalline domain size of ≈44.2 Å, and high electrical conductivity of ≈320 S cm?1, presenting significantly high rate capability at 600 C (60 A g?1) and stable cycling behaviors over 40 000 cycles as an anode for Na‐ion storage. The results of this study show the unusual graphitization behaviors of a char‐type carbon precursor and exceptionally high rate and cycling performances of the resulting graphitic material, PCN‐2800, even surpassing those of supercapacitors.  相似文献

Filtration experiments in batch concentration mode (with recycling of the retentate stream) of the cork processing wastewater were performed in laboratory filtration membrane equipment, by using four commercial membranes: two UF membranes with MWCO of 20,000 and 5000 Da, and two NF membranes with an approximate MWCO of 150-300 Da. The filtration experiments of the selected wastewater were performed by modifying the most important operating variables: transmembrane pressure, tangential velocity, temperature, and the nature and MWCO of the membranes. The evolution of the cumulative permeate volumes and permeate fluxes with processing time were analysed, and it was established that the steady-state permeate flux was reached for a volume retention factor of 2. The effect of the mentioned operating conditions on this steady-state permeate flux was discussed. The effectiveness of the filtration treatments was determined by the evaluation of the rejection coefficients for several parameters, which measure the global pollutant content of the effluent: COD, absorbance at 254 nm, tannic content, color, and ellagic acid. Finally, the resistances in series model was used for the evaluation of the resistances to the permeate flux, and it was concluded that the contribution to the total resistance of the fouling resistance (combined external plus internal) was higher than the inherent resistance of the clean membrane.  相似文献

Gallic acid (3,4,5-trihydroxybenzoic acid) is a major pollutant present in the wastewater generated in the boiling cork process, as well as in other wastewaters from food manufacturing industries. Its decay in aqueous solutions has been studied by the action of several oxidation systems: monochromatic UV radiation alone and combined with hydrogen peroxide, Fenton's reagent and the combination Fenton's reagent with UV radiation (photo-Fenton system). The influence of the pH is discussed and the quantum yields are determined in the UV radiation system. Also, the influence of operating variables (initial concentrations of H2O2 and Fe(II), and pH) is established in the Fenton's reaction. The apparent pseudo-first-order rate constants are evaluated in all the experiments conducted in order to compare the efficiency of each one of the processes. Increases in the degradation levels of gallic acid are obtained in the combined processes in relation to the single UV radiation system, due to reactions of the very reactive OH*. These improvements are determined in every process by calculating the partial contribution to the overall decomposition rate of the radical pathways. For the oxidant concentrations applied, the most effective process in removing gallic acid was found to be the photo-Fenton system. The rate constant for the reaction of gallic acid with OH was also determined by means of a competition kinetics model, being its value 11.0+/-0.1 x 10(9)l mol(-1)s(-1).  相似文献

BACKGROUND: In recent years there has been a change in the opportunistic diseases incidence in HIV-infected patients. The change could be related to the use antiviral therapy and chemoprophylaxis strategies. The aim of the present study was to evaluate if medical intervention is able to modificate the clinical presentation of AIDS and the CD4+ lymphocyte counts at time of AIDS diagnosis. PATIENTS AND METHODS: The first AIDS-defining condition and the CD4+ count at time of AIDS diagnosis were analyzed in 95 HIV-infected patients who developed an AIDS-defining disease since April 1989 until March 1996, retrospectively reviewed. Patients who had been previously followed at an AIDS Unit were compared with those who had not. RESULTS: The frequency of Pneumocystis carinii pneumonia as the first AIDS-defining condition was lower in medically followed patients. Among this group, AIDS cases who received chemoprophylaxis with isoniazide showed a decrease in the rate of tuberculosis. No differences were found in CD4+ lymphocyte counts between both groups. CONCLUSIONS: As a result of medical intervention significant changes have occurred in the spectrum of initial AIDS-defining conditions in relation to medical intervention; a decrease in the frequency of Pneumocystis carinii pneumonia and tuberculosis have been found; however, the CD4+ lymphocyte counts at time of AIDS diagnosis are not modificated by medical intervention.  相似文献

Opioid drugs play important roles in the clinical management of pain, as well as in the development and treatment of drug abuse. The mu opioid receptor is the primary site of action for the most commonly used opioids, including morphine, heroin, fentanyl, and methadone. By sequencing DNA from 113 former heroin addicts in methadone maintenance and 39 individuals with no history of drug or alcohol abuse or dependence, we have identified five different single-nucleotide polymorphisms (SNPs) in the coding region of the mu opioid receptor gene. The most prevalent SNP is a nucleotide substitution at position 118 (A118G), predicting an amino acid change at a putative N-glycosylation site. This SNP displays an allelic frequency of approximately 10% in our study population. Significant differences in allele distribution were observed among ethnic groups studied. The variant receptor resulting from the A118G SNP did not show altered binding affinities for most opioid peptides and alkaloids tested. However, the A118G variant receptor binds beta-endorphin, an endogenous opioid that activates the mu opioid receptor, approximately three times more tightly than the most common allelic form of the receptor. Furthermore, beta-endorphin is approximately three times more potent at the A118G variant receptor than at the most common allelic form in agonist-induced activation of G protein-coupled potassium channels. These results show that SNPs in the mu opioid receptor gene can alter binding and signal transduction in the resulting receptor and may have implications for normal physiology, therapeutics, and vulnerability to develop or protection from diverse diseases including the addictive diseases.  相似文献
