Friedreich’s ataxia (FRDA) is a hereditary neurodegenerative disease characterized by a reduced synthesis of the mitochondrial iron chaperon protein frataxin as a result of a large GAA triplet-repeat expansion within the first intron of the frataxin gene. Despite neurodegeneration being the prominent feature of this pathology involving both the central and the peripheral nervous system, information on the impact of frataxin deficiency in neurons is scant. Here, we describe a neuronal model displaying some major biochemical and morphological features of FRDA. By silencing the mouse NSC34 motor neurons for the frataxin gene with shRNA lentiviral vectors, we generated two cell lines with 40% and 70% residual amounts of frataxin, respectively. Frataxin-deficient cells showed a specific inhibition of mitochondrial Complex I (CI) activity already at 70% residual frataxin levels, whereas the glutathione imbalance progressively increased after silencing. These biochemical defects were associated with the inhibition of cell proliferation and morphological changes at the axonal compartment, both depending on the frataxin amount. Interestingly, at 70% residual frataxin levels, the in vivo treatment with the reduced glutathione revealed a partial rescue of cell proliferation. Thus, NSC34 frataxin silenced cells could be a suitable model to study the effect of frataxin deficiency in neurons and highlight glutathione as a potential beneficial therapeutic target for FRDA. 相似文献

In this article we present the principle methods for volumerendering of non-regular grids as used in the Finite Element Method or the Method of Control Volumes. Besides linear element types, the methods discussed here also cover the handling of commonly used non-linear element types but little considered in the Computer Graphics community. The very basic operation in the whole rendering process, namely the interpolation of the geometry and function values is fully based on the element type specific shape functions, ensuring a consistent mathematical treatment of the object in the sense of the underlying numerical simulation. We discuss the implications of this approach and propose optimizations justified by general assumptions on the qualify of the simulation model. 相似文献

Allocating production batches to subcontractors arises frequently in industry. When subcontractors operate different equipment, batch assignment is a complex decision that must take into account both throughput and quality of finished goods. In this paper, a Mixed Integer Linear Goal Programming Model, where productivity uncertainty is taken into account through Fuzzy Set Theory, is developed. Our study was motivated by a real-world application arising in an Italian textile company. Computational results show that this method outperforms the hand-made solutions put to use by the management so far. 相似文献

Miscibility and thermal and mechanical properties of poly(butylene terephthalate) (PBT) blends with poly(ε‐caprolactone) (PCL) were analyzed as a function of the molecular mass of PCL. It was found that the components are miscible when oligomeric PCL is blended with PBT, probably due to favorable interactions between ? OH end groups of poly(ε‐caprolactone) and ester groups of PBT. In the blends containing high molar mass PCL, the concentration of hydroxyl end groups is lower, allowing only partial miscibility of the components. An artificial fruit (AF) was used to test for social learning in pig-tailed macaques (Macaca nemestrina) and adult humans (Homo sapiens). A monkey demonstrator opened the AF, showing alternative methods to 2 groups of cage mates. Video films of the monkey demonstrations were presented to adult humans. Compared with chimpanzees and children, the macaques watched the demonstrations significantly less and in a much more sporadic manner. They also produced only very weak and transitory evidence of social learning. In contrast, the adult humans performed as one might expect of optimum imitators, even producing evidence of components of a "ratchet effect." In this paper we present a novel process based on SU-8 technology for the fabrication of double clamped radio frequency (RF) micro-electro-mechanical system (MEMS) capacitive shunt switches in coplanar configuration. The key element of the exploited process is the MicroChem SU-8 2002 negative photoresist. The polymeric material is widely used in MEMS device processes because of its excellent thermal and chemical stability. In this paper, SU-8 polymer has been utilized in a double way to get suspended structures as double clamped beams: (1) SU-8 for the lateral supports, and (2) as a sacrificial layer for the release of the suspended membrane. Preliminary RF tests on the manufactured switches have been done, and the measured electrical performances are in good agreement with the performed simulations. 相似文献

The influence of multilayer carton scraps (MC) on crystallization kinetics of high-density polyethylene (HDPE) is detailed in this contribution. HDPE/MC composites were prepared by melt mixing, and a maleated linear low density polyethylene (MAPE) was added as compatibilizer. The crystallization kinetics of HDPE/MC/MAPE was analyzed as function of composition both in isothermal and nonisothermal conditions. The multilayer carton scraps appear to promote the onset of crystallization of HDPE, acting as efficient nucleating agent. The presence of MAPE as compatibilizer slightly reduces the nucleating efficiency of MC: the compatibilizer induces a delay in the onset of crystallization, caused by the need to exclude the bulky pendant groups of maleated linear low-density polyethylene from the crystals, as HDPE and MAPE chains cocrystallize. © 2012 Wiley Periodicals, Inc. In this paper a low-vacuum scanning electron microscope (SEM) coupled with an energy-dispersive X-ray spectrometer (EDX) was used to investigate the alteration processes that occur on silica–soda–lime glass exposed to soil materials and dated from XI to second half of XIII sec. The chemical data were collected for altered glass gel and fresh glass. In order to study the influence of chemical composition on weathering process, 16 glasses have been selected on the basis of the chemical characterization and on the basis of the different corrosion processes present on the fragments. Six selected samples had been produced with the use of natron as fluxer and 10 samples with the use of plant ash as fluxer. The analysed pieces come from Siponto excavations (Foggia, Italy) and they include feet and rims of chalices, fragments of lamps and of globular bottles. 相似文献

The authors carried out 2 experiments designed to cast light on the locus of redundancy gain in simple visual reaction time by using a stop-signal paradigm. In Experiment 1, the authors found that single visual stimuli were more easily inhibited than double visual stimuli by an acoustic stop signal. This result is in keeping with the idea that redundancy gain occurs prior to the ballistic stage of the stop-signal task. In Experiment 2, the authors found that the response to an acoustic go signal was more easily inhibited by a double than by a single visual stop signal. This result provides conclusive evidence for a redundancy gain in the stop process—in a process that does not involve a motor response but rather its inhibition.
