A hybrid Subset Simulation approach is proposed for reliability estimation for general dynamical systems subject to stochastic excitation. This new stochastic simulation approach combines the advantages of the two previously proposed Subset Simulation methods, Subset Simulation with Markov Chain Monte Carlo (MCMC) algorithm and Subset Simulation with splitting. The new method employs the MCMC algorithm before reaching an intermediate failure level and splitting after reaching the level to exploit the causality of dynamical systems. The statistical properties of the failure probability estimators are derived. Two examples are presented to demonstrate the effectiveness of the new approach and to compare with the previous two Subset Simulation methods. The results show that the new method is robust to the choice of proposal distribution for the MCMC algorithm and to the intermediate failure events selected for Subset Simulation.  相似文献

The author examined a public goods (PG) dilemma with environmental uncertainty in a sequential protocol of play paradigm. Using a 5-person game, Experiment 1 examined provision point (PP) uncertainty and found that a certain PP requiring 3 contributors produced the PG more frequently than an uncertain PP of the same expectation that varied uniformly among 2, 3, and 4 contributors. Fixing PP at 3, Experiment 2 showed that PG provision rate was higher with a certain group size of 5 persons than an uncertain group size of the same expectation that varied uniformly among 4, 5. and 6 persons. Further analyses showed that a fear of wasting one's contribution and pessimistic beliefs of environmental uncertainty could have undermined contribution. This paper presents a two-stage structural health monitoring methodology and applies it to the Phase I benchmark study sponsored by the IASC-ASCE Task Group on Structural Health Monitoring. In the first stage, modal parameters are identified using measured structural response from the undamaged system and then from the (possibly) damaged system. In the second stage, these data are used to update a parametrized structural model of the system using Bayesian system identification. The approach allows one to obtain not only estimates of the stiffness parameters but also the probability that damage in any substructure exceeds any specified threshold expressed in terms of a fractional stiffness loss. It successfully identifies the location and severity of damage in all cases of the benchmark problem.  相似文献

Reports an error in "Prospective Relations Between Bulimic Pathology, Depression, and Substance Abuse: Unpacking Comorbidity in Adolescent Girls" by Eric Stice, Emily M. Burton and Heather Shaw (Journal of Consulting and Clinical Psychology, 2004[Feb], Vol 72[1], 62-71). The findings of Leon, Fulkerson, Perry, Keel, and Klump ("Three to four year prospective evaluation of personality and behavioral risk factors for later disordered eating in adolescent girls and boys," Journal of Youth and Adolescence, 1999, Vol. 28, No. 2, pp. 181-196), cited on page 62, were incorrectly reported. Leon et al. (1999) found that the latent variable of negative affect/attitudes determined at study entrance significantly correlated with final year eating disorder risk score when conducted with the full sample and when removing Time 1 high eating disorder risk subjects. This latent variable consisted of the GBI Depression, MPQ Negative Emotionality, EDI Ineffectiveness, EDI Interoceptive Awareness, and EDI Body Dissatisfaction scales. A subsequent univariate analysis of Time 1 Negative Emotionality scale score on Time 3 eating disorder risk, adjusting for initial eating disorder risk score, showed a nonsignificant effect size (Stice, E. [2002]. Risk and maintenance factors for eating pathology: A meta-analytic review. Psychological Bulletin, 128, 825-848). To elucidate the processes that contribute to the comorbidity between bulimic pathology, depression, and substance abuse, the authors tested the temporal relations between these disturbances with prospective data from adolescent girls (N = 496). Multivariate analyses indicated that depressive symptoms predicted onset of bulimic pathology but not of substance abuse, bulimic symptoms predicted onset of depression but not of substance abuse, and substance abuse symptoms predicted onset of depression but not of bulimic pathology. Results suggest that the comorbidity arises because certain disorders are risk factors for the other disorders. Findings also provide support for select etiologic theories and further establish the clinical significance of these conditions by showing that they increase risk for onset of other psychiatric disturbances. Important asymmetries between self-perception and social perception arise from the simple fact that other people's actions, judgments, and priorities sometimes differ from one's own. This leads people not only to make more dispositional inferences about others than about themselves (E. E. Jones & R. E. Nisbett, 1972) but also to see others as more susceptible to a host of cognitive and motivational biases. Although this blind spot regarding one's own biases may serve familiar self-enhancement motives, it is also a product of the phenomenological stance of naive realism. It is exacerbated, furthermore, by people's tendency to attach greater credence to their own introspections about potential influences on judgment and behavior than they attach to similar introspections by others. The authors review evidence, new and old, of this asymmetry and its underlying causes and discuss its relation to other psychological phenomena and to interpersonal and intergroup conflict. Speech Recognition is frequently cited as a potential remedy to distraction resulting from drivers' operation of in-vehicle devices. This position typically assumes that the introduction of speech recognition will result in reduced cognitive workload and improved driving performance. Past research neither fully supports nor fully discounts this assumption. However, it is difficult to compare many of these studies, due to differences in device operation tasks, the pacing of those tasks, speech recognition system performance, and system interface designs. In an effort to directly address the effect of voice recognition on driver distraction, the present authors developed a capability to manipulate the performance characteristics of a speech recognition system through a Wizard of Oz speech recognition system and installed this system in a simulated driving environment. The sensitivity of the simulated driving environment and speech recognition accuracy manipulation were evaluated in an initial study comparing driver cognitive workload and driving performance during self-paced simulated operation of a personal digital assistant (PDA) during no PDA use, manual control of the PDA, and speech control of the PDA. In the Speech PDA condition, speech recognition accuracy was varied between drivers. Analysis of drivers' emergency braking response times and rated cognitive workload revealed significantly lower cognitive demand and better performance in the No PDA condition when compared to the Manual PDA condition. The Speech PDA condition resulted in response times and rated cognitive workload levels that were between the No PDA and Manual PDA conditions, but not significantly different from either of these conditions. Further analysis of emergency braking performance revealed a non-significant trend towards better performance in conjunction with higher speech recognition accuracy levels. The potential for reducing driver distraction through the careful development and evaluation of speech recognition systems is discussed.  相似文献

The use of fluorescent probes that allow visualization of leukocyte-endothelial cell (EC) interactions has greatly informed our understanding of leukocyte recruitment. However, effects of these agents on the biological functions of leukocytes are poorly described, leading to concerns about the interpretation of such data. Here we used two flow-based neutrophil adhesion assays to compare the effects of phase contrast illumination (PCI) with high intensity illumination (HII) used for fluorescent microscopy, in the presence or absence of five commonly used fluorochromes. Isolated neutrophils were either (1) perfused across P-selectin to establish a population of rolling cells, which were subsequently activated with fMLP; or (2) perfused across EC activated with TNF-alpha. In the absence of fluorescent dyes, HII did not affect levels of leukocyte adhesion; however, subsequent neutrophil behavior was dramatically altered when compared with cells under PCI, for example, dramatically reducing their migration velocities. In the presence of fluorescent dyes, the effects of HII were exacerbated, although the precise nature of the biological effects of these probes was agent specific. Thus, for the first time, our experiments describe the effects of fluorescent microscopy on the separate stages of the neutrophil recruitment process and reveal a previously unsuspected effect of HII on neutrophil migration.  相似文献

In plastic injection moulding process, cooling channel design is an essential factor that affects the quality of the moulded parts and the productivity of the process. Non-uniform cooling or long cooling cycle time would result if a poorly designed cooling channel is adopted. Due to limitations of traditional machining processes, the cooling channel is usually formed from straight-line drilled holes and only simple shapes are allowed, regardless of the shape complexity of the part being moulded. With the advent of rapid tooling technology, cooling channels in complex shapes can now be possible. However, there are not many design methodologies for supporting this type of cooling channel. In this paper, a methodology called visibility-based cooling channel generation is proposed for automatic preliminary cooling channel design for rapid tooling. The cooling process between a mould surface and a cooling channel is considered analogous to whether they can be visible from each other. Without loss of generality, the mould surface is approximated by a polyhedral terrain and is normally offset. A number of point light sources together that can illuminate the whole polyhedral terrain are assigned to suitable terrain offset vertices. A cooling channel is then generated by connecting all the assigned light sources. When comparing the conventional verification and redesign methods by melt flow analysis, computer-aided design and, a better design of cooling channel for its mould surface results in a short time independent of the experience of mould engineer.  相似文献

Cold Atmospheric Plasma (CAP) is an emerging technology with great potential for biomedical applications such as sterilizing equipment and antitumor strategies. CAP has also been shown to improve skin wound healing in vivo, but the biological mechanisms involved are not well known. Our study assessed a possible effect of a direct helium jet CAP treatment on keratinocytes, in both the immortalized N/TERT-1 human cell line and primary keratinocytes obtained from human skin samples. The cells were covered with 200 µL of phosphate buffered saline and exposed to the helium plasma jet for 10–120 s. In our experimental conditions, micromolar concentrations of hydrogen peroxide, nitrite and nitrate were produced. We showed that long-time CAP treatments (≥60 s) were cytotoxic, reduced keratinocyte migration, upregulated the expression of heat shock protein 27 (HSP27) and induced oxidative cell stress. In contrast, short-term CAP treatments (<60 s) were not cytotoxic, did not affect keratinocyte proliferation and differentiation, and did not induce any changes in mitochondria, but they did accelerate wound closure in vitro by improving keratinocyte migration. In conclusion, these results suggest that helium-based CAP treatments improve wound healing by stimulating keratinocyte migration. The study confirms that CAP could be a novel therapeutic method to treat recalcitrant wounds.  相似文献

A disintegrin and metalloproteinase (ADAM) family proteins, acting as sheddases, are important factors in a number of pathologies, including cancer, and have been suggested as promising therapeutic targets. The study presented focuses on the involvement of ADAM10 and ADAM17 in retinoblastoma (RB), the most common malignant intraocular childhood tumor. A significant correlation between ADAM17 expression levels and RB laterality and RB staging was observed. Levels of ADAM10 or ADAM17 regulating miRNAs miR-145, -152, and -365 were significantly downregulated in RB cell lines, and reduced miR levels with simultaneously upregulated ADAM10 and ADAM17 expression were found in RB patients. The involvement of both ADAMs analyzed in ectodomain shedding of the neuronal cell adhesion molecule L1 (L1CAM), shown to induce pro-tumorigenic effects in RB, was confirmed. Lentiviral ADAM10 and ADAM17 single or ADAM10/17 double knockdown (KD) induced caspase-dependent apoptosis and reduced cell viability, proliferation, growth, and colony formation capacity of RB cells. Moreover, differential phosphorylation of the serine/threonine kinase AKT was observed following ADAM17 KD in RB cells. Chicken chorioallantoic membrane (CAM) assays revealed that ADAM17 and ADAM10/17 depletion decreases the tumorigenic and migration potential of RB cells in vivo. Thus, ADAMs are potential novel targets for future therapeutic RB approaches.  相似文献
