Die Wasserrahmenrichtlinie regelt den Schutz von Grund- und Oberflächengewässern und fordert eine nachhaltige Wassernutzung. Sie legt einen Zeitplan fest, um bis zum Jahr 2015 für alle Gewässer einen guten Zustand zu erreichen. Der erste Schritt ist eine Bestandsaufnahme der Gewässer. Für das Grundwasser besteht diese aus einer erstmaligen Beschreibung, einer weitergehenden Beschreibung und der Prüfung der Auswirkungen menschlicher Tätigkeit auf das Grundwasser. Die Wasserrahmenrichtlinie wird unabhängig von administrativen Grenzen innerhalb von hydrologischen Einzugsgebieten umgesetzt. Im deutschen Teil des Flusseinzugsgebietes der Elbe wurden 5 Koordinierungsräume gegründet. Der Koordinierungsraum Mulde-Elbe-Schwarze Elster (MES) wird vorgestellt. Beispielhaft für die angewandten Methoden wird die Beurteilung diffuser Stoffeinträge in das Grundwasser bei der erstmaligen Beschreibung erläutert. Die Ergebnisse der Bestandsaufnahme Grundwasser im Koordinierungsraum MES werden präsentiert. Von 54 Grundwasserkörpern ist für 25 Körper aufgrund der Belastungen durch Punktquellen und diffuse Quellen, wegen des mengenmäßigen Zustands oder sonstiger anthropogener Einwirkungen die Zielerreichung nach der Wasserrahmenrichtlinie unklar/unwahrscheinlich.  相似文献

We show how to change optically the distance between two protein-linked gold nanoparticles by Raman-induced motion of the linker protein. Rayleigh scattering spectroscopy of the coupled-particle plasmon allows us to compare the inter-nanoparticle distance of individual protein-linked gold nanoparticle dimers before and after surface-enhanced Raman scattering (SERS). We find that low-intensity (50 microW/microm2) laser light in resonance with the nanoparticle-dimer plasmon provokes a change of the inter-nanoparticle distance on the order of 0.5 nm whenever SERS from the proteins connecting the nanoparticles can be observed.  相似文献

The first application of nanocrystal (NC)-encoded microbeads to clinical proteomics is demonstrated by multiplexed detection of circulating autoantibodies, markers of systemic sclerosis. Two-color complexes, consisting of NC-encoded, antigen-covered beads, anti-antigen antibody or clinical serum samples, and dye-tagged detecting antibodies, were observed using flow cytometry assays and on the surface of single beads. The results of flow cytometry assays correlated with the ELISA technique and provided clear discrimination between the sera samples of healthy donors and patients with autoimmune disease. Microbead fluorescence signals exhibited narrow distribution regardless of their surface antigen staining, without the need of any fluorescence compensation-a parameter determining the limit of sensitivity of flow cytometry assays. In single bead measurements, less than 30 dye-labeled antibodies interacting with the topoI-specific antibodies at the surface of a bead have been detected by the emission of dye excited through the FRET from NCs. In this format, the antibody-bead interaction reaction turns specifically the fluorescence signal from dye label off and on, additionally increasing autoantibody detection sensitivity.  相似文献

Nonlinear optical materials are essential in areas such as nanophotonics, optical information processing, and biomedical imaging. However, nanomaterials employed for these diverse applications to date are efficient only for one type of nonlinear optical activity. Herein, the first multimodal nonlinear optically active class of nanomaterials based on lanthanide-doped lithium niobate nanoparticles, which simultaneously exhibit unprecedentedly efficient second and third harmonic generation, as well as up-conversion photoluminescence, is reported. These dielectric nanoparticles retain their high nonlinear optical conversion efficiency both as powder and as aqueous colloidal solution. The high stability also allows for the fabrication of optically active biocompatible micron-sized fibers and polymer-based 3D-printable objects, as well as for fingerprint detection. Finally, the first 8-bit coding platform purely based on multimodal nonlinear optical activity originating from different parametric and nonparametric processes is demonstrated, showcasing the technological potential of these materials for both anti-counterfeiting and advanced optical information processing.  相似文献

We present a method that performs a rigid 2D/3D image registration efficiently on the Graphical Processing Unit (GPU). As one main contribution of this paper, we propose an efficient method for generating realistic DRRs that are visually similar to x-ray images. Therefore, we model some of the electronic post-processes of current x-ray C-arm-systems. As another main contribution, the GPU is used to compute eight intensity-based similarity measures between the DRR and the x-ray image in parallel. A combination of these eight similarity measures is used as a new similarity measure for the optimization. We evaluated the performance and the precision of our 2D/3D image registration algorithm using two phantom models. Compared to a CPU + GPU algorithm, which calculates the similarity measures on the CPU, our GPU algorithm is between three and six times faster. In contrast to single similarity measures, our new similarity measure achieved precise and robust registration results for both phantom models.  相似文献

Semiconductor nanocrystals (SCNCs) made of CdSe, CdTe, and InP are used to photosensitize needlelike C(60) crystals. The photocurrent is increased by up to 3 orders of magnitude as compared with C(60) crystals without SCNCs. The photocurrent spectrum can be tuned precisely by the SCNC size and material, rendering the SCNC-functionalized C(60) crystals an excellent material for spectrally tuneable photodetectors. We explain the increased photocurrent as a result of photoexcited electrons transferring from the SCNCs to the C(60) crystals and causing photoconductivity, while the complementary holes remain trapped in the SCNCs.  相似文献

Binucleate trophoblast giant cells (BNC) in the bovine placenta produce glycoproteins, which are delivered into the mother after fusion of BNC with uterine epithelial cells. During most time of pregnancy, BNC produce pregnancy-associated glycoproteins (PAGs) and prolactin-related protein-I (PRP-I) with asparagine-linked lactosamine-type glycans terminating with N-acetyl-galactosamine. We show by lectin histochemistry that terminal N-acetyl-galactosamine (detected by Dolichos biflorus agglutinin, DBA) in placentomal BNC is greatly reduced prior to parturition, while lactosamine-type N-glycans (detected by Phaseolus vulgaris leucoagglutinin, PHA-L) remain unaltered. The change in DBA-staining showed no statistically significant differences between placentomes of cows with and without retention of fetal membranes. Western blots revealed that, at parturition the apparent molecular mass of PAGs and PRP-I is 1-2 kDa lower than in late pregnancy. These changes are due to alterations of asparagine-linked glycans, since the molecular weight of the peptide backbones after enzymatical release of asparagine-linked glycans is identical at late pregnancy and parturition. Lectin western blots showed a reduction of terminal N-acetyl-galactosamine on PAGs at parturition. A lectin sandwich-ELISAwas used to differentiate DBA- and PHA-L-binding PAGs in sera of pregnant and non-pregnant cows. The values for DBA-binding PAGs at parturition were not significantly different from non-pregnancy, while the values for PHA-L-binding PAGs were significantly higher at parturition. The peripartal changes of PAG- and PRP-I-glycosylation could alter functional properties of these proteins and might therefore be considered for functional studies. The differentiation of PAG glycoforms in maternal serum could be valuable for a further optimization of PAG-based pregnancy diagnosis in cattle.  相似文献

Blue emitting CaWO4 and greenish blue emitting ZnWO4 nanoparticles are synthesized via microemulsion techniques applying a cationic (CTAB) as well as a non-ionic surfactant (TritonX-100). The influence of the surfactant on particle size and shape is studied. Scanning electron microscopy and dynamic light scattering confirm the presence of uniform and non-agglomerated nanoparticles, 60–80 nm in diameter. Photoluminescence confirms [WO4]2−-related broad-band emission with its maximum at 440 nm (CaWO4) and 420 nm (ZnWO4). The highest quantum yield (QY) is observed for nanoscaled CaWO4 with a value of 23–25%. Doping of CaWO4 and ZnWO4 with Tb3+ and Eu3+ was performed and in the case of CaWO4:Tb and CaWO4:Eu and results in the emission of green and red light, again with comparably high QYs (17–19%).  相似文献

Designing nanoscale objects with the potential to perform externally controlled motion in biological environments is one of the most sought-after objectives in nanotechnology. Different types of chemically and physically powered motors have been prepared at the macro- and microscale. However, the preparation of nanoscale objects with a complex morphology, and the potential for light-driven motion has remained elusive to date. Here, we go a step forward by designing a nanoscale hybrid with a propeller-resembling shape, which can be controlled by focused light under biological conditions. Our hybrid, hereafter "Au@DNA-origami", consists of a spherical gold nanoparticle with self-assembled, biocompatible, two-dimensional (2D) DNA sheets on its surface. As a first step toward the potential utilization of these nanoscale objects as light-driven assemblies in biological environments, we show that they can be optically trapped, and hence translated and deposited on-demand, and that under realistic trapping conditions the thermally induced dehybridization of the DNA sheets can be avoided.  相似文献
