We describe a simple, effective approach to the creation of autologous tissue-engineered cartilage in the shape of a human nipple by injecting a reverse thermosensitive polymer seeded with autologous chondrocytes in an immunocompetent porcine animal model. A biodegradable, biocompatible copolymer of polyethylene oxide and polypropylene oxide (Pluronic F-127), which exists as a liquid below 4 degrees C and polymerizes to a thick gel when it is exposed to physiologic temperatures (body temperatures), was used as a vehicle for chondrocyte delivery and as a scaffold to guide growth. Autologous chondrocytes isolated from porcine auricular elastic cartilage and suspended in 30% (weight/volume) Pluronic F-127 were injected on the ventral surface of the pigs from which the cells had been isolated. A circumferential subdermal suture was used to support the contour of the implant and assist in its projection in the form of a human nipple. After 3 weeks, the skin over and surrounding the implant was tattooed to create the appearance of a human nipple-areolar complex. As controls, an equal number of injections were made using either cells alone (not suspended in hydrogel), or hydrogel alone. After 10 weeks, all specimens were excised and examined both grossly and histologically. Before harvesting, visual inspection of the tattooed chondrocyte-Pluronic F-127 hydrogel implant sites revealed that they closely resembled a human female nipple-areolar complex. Nodules were similar in size, shape, and texture to a human nipple at each injection site. Glistening opalescent tissue was surgically isolated from each implant site. Hematoxylin and eosin, safranine o, trichrome blue, and Verhoeff's stains of the experimental implants showed nodules with the characteristic histologic signs of elastic cartilage. Control injections of copolymer hydrogel alone exhibited no evidence of cartilage formation. Control injections of chondrocytes alone showed evidence of dissociated microscopic nodules of elastic cartilage.  相似文献

We compare kernel estimators, single and multi-layered perceptrons and radial-basis functions for the problems of classification of handwritten digits and speech phonemes. By taking two different applications and employing many techniques, we report here a two-dimensional study whereby a domain-independent assessment of these learning methods can be possible. We consider a feed-forward network with one hidden layer. As examples of the local methods, we use kernel estimators like k-nearest neighbour (k-nn), Parzen windows, generalised k-nn, and Grow and Learn (Condensed Nearest Neighbour). We have also considered fuzzy k-nn due to its similarity. As distributed networks, we use linear perceptron, pairwise separating linear perceptron and multi-layer perceptrons with sigmoidal hidden units. We also tested the radial-basis function network, which is a combination of local and distributed networks. Four criteria are taken for comparison: correct classification of the test set; network size; learning time; and the operational complexity. We found that perceptrons, when the architecture is suitable, generalise better than local, memory-based kernel estimators, but require a longer training and more precise computation. Local networks are simple, leant very quickly and acceptably, but use more memory.  相似文献

The distribution of NADPH-diaphorase (ND) activity was histochemically investigated in the brain of the frog Rana perezi. This technique provides a highly selective labeling of neurons and tracts. In the telencephalon, labeled cells are present in the olfactory bulb, pallial regions, septal area, nucleus of the diagonal band, striatum, and amygdala. Positive neurons surround the preoptic and infundibular recesses of the third ventricle. The magnocellular and suprachiasmatic hypothalamic nuclei contain stained cells. Numerous neurons are present in the anterior, lateral anterior, central, and lateral posteroventral thalamic nuclei. Positive terminal fields are organized in the same thalamic areas but most conspicuously in the visual recipient plexus of Bellonci, corpus geniculatum of the thalamus, and the superficial ventral thalamic nucleus. Labeled fibers and cell groups are observed in the pretectal area, the mesencephalic optic tectum, and the torus semicircularis. The nuclei of the mesencephalic tegmentum contain abundant labeled cells and a conspicuous cell population is localized medial and caudal to the isthmic nucleus. Numerous cells in the rhombencephalon are distributed in the octaval area, raphe nucleus, reticular nuclei, sensory trigeminal nuclei, nucleus of the solitary tract, and, at the obex levels, the dorsal column nucleus. Positive fibers are abundant in the superior olivary nucleus, the descending trigeminal, and the solitary tracts. In the spinal cord, a large population of intensely labeled neurons is present in all fields of the gray matter throughout its rostrocaudal extent. Several sensory pathways were heavily stained including part of the olfactory, visual, auditory, and somatosensory pathways. The distribution of ND-positive cells did not correspond to any single known neurotransmitter or neuroactive molecule system. In particular, abundant codistribution of ND and catecholamines is found in the anuran brain. Double labeling techniques have revealed restricted colocalization in the same neurons and only in the posterior tubercle and locus coeruleus. If ND is in amphibians a selective marker for neurons containing nitric oxide synthase, as generally proposed, with this method the neurons that may synthesize nitric oxide would be identified. This study provides evidence that nitric oxide may be involved in novel tasks, primarily related to forebrain functions, that are already present in amphibians.  相似文献

Cholera returned to South America in January 1991, after almost a century of absence. The hygienic status of the countries affected, aggravated by economic and political difficulties, allowed the disease to spread rapidly. In Peru, fishery products were incriminated from the outset, although without conclusive evidence. However, epidemiological and laboratory findings in other countries have confirmed the transmission of the disease by these products. The authors discuss the effects of the recent cholera epidemic on the trade and consumption of fishery products in countries of South America. The actual risk of cholera transmission by food and in particular by the consumption of fishery products is discussed, and a basis for conducting an accurate evaluation of these risks is proposed. Finally, the authors summarise the measures recommended to prevent the transmission of cholera by fishery products.  相似文献

Protein extrusion has frustrated earlier predictions regarding its impact in the development of food products. The main reason for this disappointing performance has been its failure to yield fabricated food products with textural quality close enough to that of natural products at competitive prices. Texturized soya protein by extrusion is presently the only commercial success in this area, being incorporated into several convenience products, increasing their protein content and quality and conferring them some desirable sensory properties. Technological and scientific gaps in the extrusion texturization are still to be bridged if this technique is to be applied for upgrading unconventional protein. The precise mechanisms responsible for protein texturization through extrusion are still unclear. Proteins show a very wide range of extrusion behavior that is probably related to large differences in their association properties. New peptide bonds, formed by free amino and carboxylic groups of the protein, were postulated as being responsible for the cross-linking that takes place in protein extrusion. However, disulfide bonds and electrostatic and hydrophobic interactions are regarded presently as the texturization mechanism in this process. The recently suggested suspension (or filled "melt") model for biopolymer extrusion offered a new framework for testing extrusion of novel proteins. According to this view, the large differences between the association properties of proteins produce different types of aggregates. Some of them can be insoluble under extrusion conditions and act as a dispersed phase within the melt phase. The extrusion performance of a protein will thus depend on the amount of insoluble aggregate produced inside the extruder and on protein-protein interactions that occur after the superheated molten mass leaves it.  相似文献

Transmission performance of a CMUT element in terms of output pressure and displacements was evaluated. A SIMULINK model of single CMUT element based on mechanical model of MEMS capacitor was used and the analyses were performed under different ac and dc conditions. 2.6 µm thick Si, Poly-Si and Si3Ni4 membranes with a radius 60 µm were used to obtain results for underwater imaging application. Relation between membrane stress and outputs of CMUT was also investigated using SIMULINK model for commonly used CMUT membrane made of Si3Ni4 and polysilicon membrane under different electrical driving parameters. It was observed that different ac signal inputs (sine, square and sawtooth) showed different effects on CMUTs pressure and displacement characteristics. Our results indicated that the maximum output pressure and displacement were obtained in a square waveform. In addition, although stress on membrane increases the displacement and pressure of CMUT membrane made of Poly-Si, quality factor inversely proportional to stress on membrane. Membrane stress has adverse effect on Si3Ni4 membrane transmission outputs. The used model in this study might enable to determine optimum driving electrical inputs and stress on membrane to control CMUT outputs in terms of output pressure, displacement, quality factor and bandwitdh.

The ultimate performance of a solid state device is limited by the restricted number of crystalline substrates that are available for epitaxial growth. As a result, only a small fraction of semiconductors are usable. This study describes a novel concept for a tunable compliant substrate for epitaxy, based on a graphene–porous silicon nanocomposite, which extends the range of available lattice constants for epitaxial semiconductor alloys. The presence of graphene and its effect on the strain of the porous layer lattice parameter are discussed in detail and new remarkable properties are demonstrated. These include thermal stability up to 900 °C, lattice tuning up to 0.9 % mismatch, and compliance under stress for virtual substrate thicknesses of several micrometers. A theoretical model is proposed to define the compliant substrate design rules. These advances lay the foundation for the fabrication of a compliant substrate that could unlock the lattice constant restrictions for defect‐free new epitaxial semiconductor alloys and devices.  相似文献

This paper is a tutorial on how to model hybrid systems as hybrid programs in differential dynamic logic and how to prove complex properties about these complex hybrid systems in KeYmaera, an automatic and interactive formal verification tool for hybrid systems. Hybrid systems can model highly nontrivial controllers of physical plants, whose behaviors are often safety critical such as trains, cars, airplanes, or medical devices. Formal methods can help design systems that work correctly. This paper illustrates how KeYmaera can be used to systematically model, validate, and verify hybrid systems. We develop tutorial examples that illustrate challenges arising in many real-world systems. In the context of this tutorial, we identify the impact that modeling decisions have on the suitability of the model for verification purposes. We show how the interactive features of KeYmaera can help users understand their system designs better and prove complex properties for which the automatic prover of KeYmaera still takes an impractical amount of time. We hope this paper is a helpful resource for designers of embedded and cyber–physical systems and that it illustrates how to master common practical challenges in hybrid systems verification.  相似文献

Dried lung defatted with several solvents was texturized by extrusion cooking. The emulsifying capacity, water absorption capacity, and protein solubility, at different pH and MgCl(2) concentrations, of this material were determined prior to the after extrusion, as well as the texture profile analysis of the rehydrated extrudates. Solubility profile and emulsifying capacity of the lung proteins varied in relation to the solvent used in their defatting while the water absorption capacity was not affected. The extrusion of these materials promoted a higher solubilization of the protein fraction. The texture parameters of the extrudates studied and the solvent used to defat this material are related, indicating that the lipid-protein complex, left after solvent extraction, may play an important role in the texturization processof lung proteins.  相似文献

This paper investigates the impacts of fuzzy genetic (FG), a new fuzzy logic model with genetic algorithm, artificial neural networks (ANN) and general linear model (GLM) approaches on abrasive wear of concrete. For this purpose, experimental studies were made to investigate the influence on wear of the following input parameters: hematite, cement, compressive strength and different loads on the experiments. In these models, 60 data sets were used. For training set, 48 data (80 %) were randomly selected and the residual data (12 data, 20 %) were test set. Model results were compared with experimental results. In this paper, main model performance criterion was root mean square errors. Also, sum of squared error and determination coefficient statistics were used as comparing criteria for the evaluation of models’ performances. Comparison results indicate that FG models are superior to ANN and GLM models in modeling of influence hematite, cement, compressive strength and loads on wear of concrete.  相似文献
