Aqueous interfaces are of paramount importance in the study of biological systems as well as in the biomedical sciences. To study these interfaces at the nanometer level it is of interest to develop methods that allow their observation with cryogenic transmission electron microscopy (cryo-TEM). Prevention of dehydration to preserve the "native" state during sample preparation prior to vitrification is often one of the most important parameters to control in cryo-TEM experiments. For the preparation of these types of samples, we felt the need for an extended workspace with temperature and humidity control; a 'glove-box' that seamlessly connects to the vitrification instrument, the Vitrobot. In this paper we describe the use of the glove-box in the 2D and 3D cryo-TEM study of DNA adsorption and calcium carbonate mineralization to Langmuir films. The data presented illustrates the necessity of a humidity-controlled environment to preserve the original "native" state of the monolayer system. 相似文献

A new LAPS (light-addressable potentiometric sensor) set-up will be introduced, in which the light sources are miniaturised by the utilisation of a VCSEL (vertical-cavity surface-emitting laser) array to increase the measurement spot density. An FPGA (field-programmable gate array) is used to generate modulation signals for individual illumination of each measurement spot of the LAPS. The new set-up can operate a large number of measurement spots simultaneously by reading out the sum photocurrent and separate the signals of the individual measurement spots by an FFT analysis. The frequency, amplitude and offset of the modulation signal can be configured for each measurement spot by software. The new system can be combined with a positioning stage allowing the parallel read out of a single line of measurement spots and a scan perpendicular to that line in a similar manner, like for an optical scanner set-up. First measurements demonstrate the functionality of the new LAPS set-up as a chemical imaging system. 相似文献

The Resources‐Events‐Agents (REA) model is a semantic data model for the development of enterprise information systems. Although this model has been proposed as a benchmark for enterprise information modelling, only few studies have attempted to empirically validate the claimed benefits of REA modelling. Moreover, these studies focused on the evaluation of REA‐based system implementations rather than directly assessing the REA‐modelled conceptual schemas that these systems are based on. This paper presents a laboratory experiment that measured the user understanding of diagrammatic conceptual schemas developed using the REA model. The theoretical foundation for the hypotheses are cognitive theories that explain pattern recognition phenomena and the resulting reduction in cognitive effort for understanding conceptual schemas. The results of the experiment indicate a more accurate understanding of the business processes and policies modelled when users recognize the REA model’s core pattern of enterprise information in the diagram. The implication for modelling practice is that the use of the REA model improves the requirements engineering process by facilitating the user validation of conceptual schemas produced by analysts, and thus helps ensuring the quality of the enterprise information system that is developed or implemented. 相似文献

AbstractAttachment of microorganisms to food contact surfaces and the subsequent formation of biofilms may cause equipment damage, food spoilage and even diseases. Mixed-species biofilms are ubiquitous in the food industry and they generally exhibit higher resistance to disinfectants and antimicrobials compared to single-species biofilms. The physiology and metabolic activity of microorganisms in mixed-species biofilms are however rather complicated to study, and despite targeted research efforts, the potential role of mixed-species biofilms in food industry is still rather unexplored. In this review, we summarize recent studies in the context of bacterial social interactions in mixed-species biofilms, resistance to disinfectants, detection methods, and potential novel strategies to control the formation of mixed-species biofilms for enhanced food safety and food quality. 相似文献

Polymer nanocarriers are used as transport modules in the design of the next generation of drug delivery technology. However, the applicability of nanocarrier‐based technology depends strongly on our ability to precisely control and reproduce their synthesis on a large scale because their properties and performances are strongly dependent on their size and shape. Fundamental studies and practical applications of polymer nanocarriers are hampered by the difficulty of using the current methods to produce monodispersed nanocarriers in large quantities and with high reproducibility. Here, a versatile and scalable approach is reported for the large‐scale synthesis of polymer nanocarriers from water‐in‐oil miniemulsions. This method uses microfluidization to perform a controlled emulsification and is proven to be effective to prepare nanocarriers of different biopolymers (polysaccharides, lignin, proteins) up to 43 g min?1 with reproducible size and distribution. 相似文献

A series of ruthenium complexes with and without TiO2 anchoring carboxylic acid groups have been synthesised and characterised using nuclear magnetic resonance (NMR), UV–vis and luminescence. These complexes were adsorbed on thin films of the wide band-gap semiconductor anatase and were tested as photosensitisers under standard conditions. I/V characteristics of such devices revealed superior performance of the non-symmetric complexes [4′-(4-bromophenyl)-[2,2′; 6′,2″] terpyridine]Ru(II) [4′-(4-bromophenyl)-[2,2′; 6′,2″] terpyridine-4,4″-dicarboxylic acid] and [2,2′; 6′,2″] terpyridine Ru(II) [2,2′; 6′,2″] terpyridine-4′-carboxylic acid with a maximum output power 0.016 mW cm−2 under illumination at 100 mW cm−2 AM1.5 and efficiencies 3 times higher than the symmetric complexes. 相似文献

The relation between texture and microstructure and the macroscopic solidification direction has been investigated for an austenitic 316L stainless steel weld. The fusion zone has been characterised by EBSD. Texture development has been simulated by thermal Finite Element Analysis coupled with a simple model of preferential <100>‐oriented solidification following the solidification direction. The thermal part is validated by means of thermocouple measurements at several locations on the specimen. The predicted texture is in very good agreement with the measured one. 相似文献

Duocarmycins are highly cytotoxic natural products that have potential for development into anticancer agents. Herein we describe proposed but previously unidentified NH analogues of the DNA‐alkylating subunit and characterise these by solvolysis studies, NMR and computational modelling. These compounds are shown to be the exclusive intermediates in the solvolysis of their seco precursors and to possess very similar structural features to the widely studied O‐based analogues, apart from an unusually high basicity. The measured pKa of 10.5 implies that the NH compounds are fully protonated under physiological conditions. Remarkably, their extremely high reactivity (calculated hydrolysis rate 108 times higher for protonated NH compared to the neutral O analogue) is still compatible with potent cytotoxicity, provided the active species is formed in the presence of cells. These surprising findings are of relevance to the design of duocarmycin‐based tumour‐selective therapies. 相似文献

In this article barycenters of the places of publication of monographs, edited books and book chapters are used to represent the internationalization of research in the Social Sciences and Humanities (SSH) as practiced at universities in Flanders (Belgium). Our findings indicate that, in terms of places of publication, the distance between peer reviewed and non-peer reviewed SSH book literature is growing. Whereas peer reviewed books are increasingly published abroad and in English, non-peer reviewed book literature remains firmly domestic and published in the Dutch language. This divergence is more the case for the Social Sciences than for the Humanities. For Law we have found a pattern along the lines of the Social Sciences. We discuss these findings in view of the two main readerships of SSH publications: international academia on the one hand, and a mostly domestic intelligentsia on the other. 相似文献

A simple and efficient method for the encapsulation of polymer and organic photovoltaic prototypes is presented. The method employs device preparation on glass substrates with subsequent sealing using glass fiber reinforced thermosetting epoxy (prepreg) against a back plate. The method allows for transporting oxygen and water sensitive devices outside a glove box environment after sealing and enables sharing of devices between research groups such that efficiency and stability can be evaluated in different laboratories. 相似文献
