Nanocrystalline titanium dioxide (nanoTiO2) has been reported to generate reactive oxygen species (ROS) under UV illumination. In our studies, changes in mechanical properties of human skin fibroblasts, exposed to the oxidative stress induced in the presence of nanoTiO2 and UV light, were studied using atomic force microscopy (AFM). The exposure of cells to the action of ROS was performed at low TiO2 concentration (4 microg/mL) and under illumination with low-intensity UVA (8 and 20 mW/cm2) or UVC (0.1 mW/ cm2). AFM measurements of the cell stiffness were carried out immediately after exposure of cells to the oxidative stress. The data suggest that under illumination with low-intensity UVA nanoTiO2 generates ROS, which, in turn, damage cellular and subcellular structures. This process was detected by AFM as a marked drop in the cellular stiffness of ca. 30-75%, which occurred rapidly, in the time frame of 1 min. The photo-oxidative stress inducing the decrease of cell stiffness was cancelled in the presence of a well-established antioxidant, beta-carotene. Inductively coupled plasma-mass spectrometry (ICP-MS) method EPA 200.8 is gradually finding acceptance as an alternative to uranium analysis. A comparison of the ICP-MS with the accepted radiochemical method EPA 908.0 has been carried out based on data from laboratory control standards, national proficiency test samples, and environmental and drinking water samples from the State of Utah. The method detection limit (MDL) for ICP-MS was determined to be 0.017 microg/L or (0.011 pCi/L), and the minimum reporting limit (MRL) was 0.17 microg/L (MDL x 10) or (0.11 pCi/L). The minimum reporting limit for radiochemical 908.0 method is 1 pCi/L. Our spiked matrix recoveries, spiked blank samples, and reference materials deviate only a few percentage from the listed true values. Results demonstrate that the ICP-MS is a superior analytical tool for the determination of uranium in drinking and environmental waters at concentrations required by the United States Environmental Protection Agency. 相似文献

In this work, the response surface methodology was used to determine the optimum level of input variables, gluten deformation index (mm) and the level of grape seed flour (%) addition in wheat flour that improve falling number index and Mixolab dough rheological parameters. The full factorial model was the best compatible model which fitted to each response. A replacement dose of 4.08% in wheat flour with gluten deformation index value of 8.27 mm was found optimal. Methyl-xanthines and adrenergic stimulants, such as caffeine and synephrine, are commonly added to food supplements due to their stimulating and thermogenic effects. In addition, the abusive consumption of food supplements with ergogenic and aesthetic purposes has been observed worldwide. This work describes the study of caffeine, p-synephrine, hordenine, octopamine, tyramine, ephedrine and salicin as stimulants in dietary supplements marketed in Brazil for weight loss and physical fitness claims. A total of 94 different products were acquired from 30 Brazilian websites. Thus, the sampling of marketed supplements was performed in virtual commerce (e-commerce) with claims of weight loss, appetite reduction, fat burning and metabolism acceleration. The developed analytical method involved the separation of the stimulants by HPLC with diode array detection (HPLC-DAD) by using a gradient elution of flow rate (0.7–2.5 ml min?1) and mobile phase composition (0.1% H3PO4/methanol). The validated method was applied to the study of 46 dietary supplements. Caffeine, p-synephrine and ephedrine were found to be present as stimulants in 52% of the studied samples marketed as encapsulated or bulk forms. Caffeine was found to be present in concentrations that represent doses from 25.0 to 1476.7 mg day–1. Synephrine was found in concentrations that represent doses from 59.1 to 127.0 mg day–1. Ephedrine was found to be associated with caffeine in one formulation at a concentration representing a 26.1 mg day–1 dosage. 相似文献

The production and characterization of nanocomposites based on whey protein concentrate (WPC) and montmorilonite (MMT) incorporated with lycopene as a functional substance is presented and discussed as an alternative biomaterial for potential uses in foodstuff applications. A full factorial design with varying levels of MMT (0% and 2% in w/w) and lycopene (0%, 6%, and 12% in w/w) was used. Color, light transmission, film transparency, moisture, density, solubility, water vapor permeability, and antioxidant activity of the resulting materials were evaluated. Results indicated that lycopene and MMT nanoparticles were successfully included in WPC films using the casting/evaporation method. Inclusion of 2% w/w of MMT in the polymeric matrix significantly improved barrier property against water vapor. Lycopene, besides its good red coloring ability, provided to the films antioxidant activity and UV‐vis light protection. Five anthocyanins were detected in the sweet cherry extract as follows: cyanidin 3-rutinoside, cyanidin 3-glucoside, peonidin 3-rutinoside, peonidin 3-glucoside, and pelargonidin 3-rutinoside, whereas the cyanidin 3-rutinoside was found to be in the highest amount. The effect of thermal treatment on the degradation of the polyphenolic compounds in sweet cherry extract was investigated in the range of 70–120 °C by means of fluorescence spectroscopy and spectrophotometric techniques. The fluorescence spectra were dominated by emission bands with maximum ranging from 356 nm at 25 °C to 350 nm at 110 °C. The heating of sweet cherry extract resulted in structural changes that led to a significant decrease in fluorescence intensity when increasing temperature. Degradation rate constants were estimated using a fractional conversion kinetic model. The activation energy values revealed a higher-temperature dependence of antioxidant activity, followed by anthocyanins, total polyphenols, and total flavonoids. 相似文献

The extracts (0.5% [w/w]) from Caesalpinia spinosa or tara (CS) and Caesalpinia decapetala (CD) leaves were evaluated for their ability to inhibit lipid oxidation using oil‐in‐water (O/W) emulsions as the model system. Liquid chromatography (LC) combined with mass spectrometry (MS), using a hybrid quadrupole time‐of‐flight (QTOF) mass spectrometer, was employed for (1) the identification of potential antioxidant species in the extracts and (2) to follow their evolution through aging of the emulsions. CS extracts, containing esters and conjugated forms of gallic acid (GA), turned out to be more effective than CD, rich in flavonoid species, in stabilizing the O/W emulsions. After 3 wk at 33 °C, peroxide values of emulsions stabilized with CS and CD extracts were 6.7 and 18.2 meq hydroperoxides/kg, respectively. Responses measured for the oligomers and esterified forms of GA in CS containing emulsions decreased with time; however, the response for the free form of GA kept increasing. 相似文献
