Traditionally, the amount of infective chlamydiae in a given sample is determined by inoculating dilution series into cell cultures and physically counting chlamydial inclusions. This approach is time consuming, tedious, and error prone, mainly when dealing with high titers. Therefore, this paper describes a largely automated technique that was developed to standardize the determination of chlamydial load in vitro. Cells are fixed at 36 h post-inoculation and bacteria visualized using standard immunological detection methods. Consequently, for 81 microscopic fields, an image is recorded at the interpolated focal plane. These images are then automatically processed using an ImageJ plugin and the obtained results are imported into Excel to determine the number of inclusion forming units per mL in the sample. The main advantage of this technique is that no or minimal sample dilution is required, thus minimizing dilution errors. In addition, this technique was employed during the early, middle and late growth stages of the chlamydial developmental cycle and results correlated well (P < 0.01) with 16S rRNA values from previous experiments, thereby proving its suitability to follow chlamydial growth in vitro. The method described is highly suitable for high throughput titration of cell culture inoculated samples and assessment of possible antichlamydial effects of novel compounds throughout the chlamydial growth cycle. The effect of metal exposure on the accumulation and cytosolic speciation of metals in livers of wild populations of European eel with special emphasis on metallothioneins (MT) was studied. Four sampling sites in Flanders showing different degrees of heavy metal contamination were selected for this purpose. An on-line isotope dilution method in combination with size exclusion (SE) high pressure liquid chromatography (HPLC) coupled to Inductively Coupled Plasma time-of-flight Mass Spectrometry (ICP-TOFMS) was used to study the cytosolic speciation of the metals. The distribution of the metals Cd, Cu, Ni, Pb and Zn among cytosolic fractions displayed strong differences. The cytosolic concentration of Cd, Ni and Pb increased proportionally with the total liver levels. However, the cytosolic concentrations of Cu and Zn only increased above a certain liver tissue threshold level. Cd, Cu and Zn, but not Pb and Ni, were largely associated with the MT pool in correspondence with the environmental exposure and liver tissue concentrations. Most of the Pb and Ni and a considerable fraction of Cu and Zn, but not Cd, were associated to High Molecular Weight (HMW) fractions. The relative importance of the Cu and Zn in the HMW fraction decreased with increasing contamination levels while the MT pool became progressively more important. The close relationship between the cytosolic metal load and the total MT levels or the metals bound on the MT pool indicates that the metals, rather than other stress factors, are the major factor determining MT induction.  相似文献

In recent research, the use of ICT applications in real time construction projects have been documented and analysed extensively. However, there is a need for identifying and analysing in-depth the mechanisms influencing the use of interorganisational ICT applications and for solutions to eliminate potential barriers to the successful use of ICT. A model is developed providing insights in these mechanisms. Based on this model and expert interviews in the United States construction industry directions for solutions to barriers to the successful use of interorganisational ICT — document management applications, workflow management applications, and product modelling applications — in construction projects are formulated. These solutions focus on stimulating the personal motivation to use ICT, the external motivation to use this technology, and facilitating conditions in terms of knowledge and skills and acting opportunities to use ICT.  相似文献

The interplay between information and communication technology (ICT) and the competitiveness of construction firms is considered. More specifically, the question is whether firms that invest in information and communication technology have a production cost advantage. The economics literature hypothesizes that ICT brings about a production cost advantage, as ICT brings flexibility and improves the planning, organization and control of work. To test this proposition for the construction industry, a production cost function allowing for the inclusion of ICT is formulated. Using statistical nearest-neighbour matching methods it is possible to identify the effect of ICT on production costs thereby controlling for economic moderators. Data from a sample of Dutch construction firms reveal that those firms that installed ICT capital do have a production cost advantage. The results indicate further that firms need a minimum level of ICT capital to fully benefit from its production cost advantage.  相似文献

This paper reviews long-term development of biofuels in Sweden and the Netherlands. In particular this paper explores the social dynamics of ‘niche protection’. The Swedish and Dutch cases are analyzed by means of the Strategic Niche Management (SNM) perspective extended with insights from political science. Our main argument is that the development of biofuels and the way this development is protected relies on a variety of actor strategies and (local and global) discourses. This case therefore suggests that policy making for biofuels is a complex and non-linear process that can only partly be managed by policy actors.  相似文献

Extraction of As, Cd, Cu, Mn, Ni and Ti from two white as well as from one brown rice samples was studied by inductively coupled plasma sector field mass spectrometry (ICP-SF-MS). Samples were subjected to cold and hot water extraction (rice mass to deionised water volume ratio = 1:6, and 1:3, respectively). The discarded liquids were freeze-dried and digested by microwave-assisted digestion. About 50% of the content of the investigated elements could be extracted from the white rice. In case of the brown rice, the boiling water contained As, Cd, Ni and Ti in significant percentages. Elemental distributions in the rice grains determined by synchrotron radiation confocal micro-X-ray fluorescence analysis revealed that a surface layer having a thickness of about 80 μm is the richest region in elements. Ti was detectable only in this so-called skin region. Good correlation was observed for the extraction of As, Mn, Ni and Ti by the above-mentioned two techniques. Thus, in regions affected by heavy metal and other toxic element contamination, those rice dishes would be preferred whose preparation should need abundant amounts of water for washing and cooking, which later should be discarded.  相似文献

The aim of this review is to give a comprehensive overview of the current knowledge on plant metabolites of mycotoxins, also called masked mycotoxins. Mycotoxins are secondary fungal metabolites, toxic to human and animals. Toxigenic fungi often grow on edible plants, thus contaminating food and feed. Plants, as living organisms, can alter the chemical structure of mycotoxins as part of their defence against xenobiotics. The extractable conjugated or non‐extractable bound mycotoxins formed remain present in the plant tissue but are currently neither routinely screened for in food nor regulated by legislation, thus they may be considered masked. Fusarium mycotoxins (deoxynivalenol, zearalenone, fumonisins, nivalenol, fusarenon‐X, T‐2 toxin, HT‐2 toxin, fusaric acid) are prone to metabolisation or binding by plants, but transformation of other mycotoxins by plants (ochratoxin A, patulin, destruxins) has also been described. Toxicological data are scarce, but several studies highlight the potential threat to consumer safety from these substances. In particular, the possible hydrolysis of masked mycotoxins back to their toxic parents during mammalian digestion raises concerns. Dedicated chapters of this article address plant metabolism as well as the occurrence of masked mycotoxins in food, analytical aspects for their determination, toxicology and their impact on stakeholders.  相似文献

Conventional husbandry systems for pork production are scrutinized by members of the general public as well as the scientific community. As a response, alternative forms of pig production, such as outdoor housing, organic farming and environmental enrichment are gaining interest. The question arises whether these production systems are indeed able to improve the welfare and health status of the animals, and whether these production systems alter production characteristics and meat or carcass traits. Measures of poor welfare have been described, but evaluating overall welfare is difficult. Certain parameters of alternative housing will improve welfare in some ways but, simultaneously, other welfare problems are inflated, and the weighting of each of these problems is very subjective. Alternative housing systems allow pigs to display species‐specific behaviour and decrease the occurrence of abnormal behaviours by acting on several parameters: indoor versus outdoor housing, floor space/density, floor type, and provision of bedding or other types of environmental enrichment. Evaluating alternative housing systems should be done by looking at all the welfare‐improving factors and the cost of alleviating welfare‐decreasing problems in a given production system. Data in the literature on growth, meat and carcass traits in alternative production systems, are inconsistent, indicating that other factors can play an important role. However, as equal, or in some cases even better, performance can be attained in certain production systems that meet concerns of animal welfare scientists and members of the general public, alternative production forms may be considered preferable. One of the main challenges in food contact materials research is to prove that the presence of non-intentionally added substances (NIAS) is not a safety issue. Migration extracts may contain many unknown substances present at low concentrations. It is difficult and time-consuming to identify all these potential NIAS and concurrently to assess their health risk upon exposure, whereas the health relevance at low exposure levels might not even be an issue. This paper describes a scientifically based, but pragmatic safety assessment approach for unknown substances present at low exposure levels in food contact matrices. This complex mixture safety assessment strategy (CoMSAS) enables one to distinguish toxicologically relevant from toxicologically less relevant substances, when related to their respective levels of exposure, and allows one to focus on the substances of potential health concern. In particular, substances for which exposure will be below certain thresholds may be considered not of health relevance in case specific classes of substances are excluded. This can reduce the amount of work needed for identification, characterisation and evaluation of unknown substances at low concentration. The CoMSAS approach is presented in this paper using a safety assessment of unknown NIAS that may migrate from three carton samples.  相似文献

Foodborne illness continues as a considerable threat to public health. Despite improved hygiene management systems and increased regulation, pathogenic bacteria still contaminate food, causing sporadic cases of illness and disease outbreaks worldwide. For many centuries, microbial antagonism has been used in food processing to improve food safety. An understanding of the mode of action of this microbial antagonism has been gained in recent years and potential applications in food and feed safety are now being explored. This review focuses on the potential opportunities presented, and the limitations, of using microbial antagonism as a biocontrol mechanism to reduce contamination along the food chain; including animal feed as its first link. © 2014 Society of Chemical Industry  相似文献
