This paper details the results of a survey of UK potable water supply organisations, which was undertaken in 1998 on behalf of UK Water Industry Research. An extrapolation of the data indicated that, in 1997, about 131 000 tDS of water-treatment works sludge were produced in the UK. The relevant methods and associated costs are reported. A number of companies are using, or developing, novel waste-management methods for water-treatment works sludges, such as incorporation into bricks and spreading on agricultural land, and these are discussed.  相似文献

A pilot-plant contact digester treating an urban waste which approximated the BOD strength of domestic sewage was operated and the effect of loading and temperature, in the range 20° – 27°C, on performance was evaluated. Acclimatization of the digester to full loading is described and the upper limit of loading was determined. The problem of separation of sludge is also discussed.  相似文献

Managing risks in design intensive multi-disciplinary capital projects presents unique problems for decision makers within the project environment. Existing risk management tools, tend to employ axiomatic software and as such do not provide any intelligent risk management capability or lead to better informed decision making processes. ConSERV is being developed as a comprehensive project management methodology able to extract project specific risk issues throughout the project life cycle and apply experiential knowledge by using a knowledge-based system to assist in the management of them. A visual representation of the project risks is obtained by employing a more practical non probabilistic approach. ConSERV will provide project managers with on line decision support, at key stages of the project to assist in the management and control of the project specific risks identified.  相似文献

The conformational properties of proteins can be probed with hydrogen/deuterium exchange mass spectrometry (HXMS). In order to maintain the deuterium label during LC/MS analyses, chromatographic separation must be done rapidly (usually in under 8-10 min) and at 0 degrees C. Traditional RP-HPLC with approximately 3-mum particles has shown generally poor chromatographic performance under these conditions and thereby has been prohibitive for HXMS analyses of larger proteins and many protein complexes. Ultraperformance liquid chromatography (UPLC) employs particles smaller than 2 mum in diameter to achieve superior resolution, speed, and sensitivity as compared to HPLC. UPLC has previously been shown to be compatible with the fast separation and low temperature requirements of HXMS. Here we present construction and validation of a custom UPLC system for HXMS. The system is based on the Waters nanoACQUITY platform and contains a Peltier-cooled module that houses the injection and switching valves, online pepsin digestion column, and C-18 analytical separation column. Single proteins in excess of 95 kDa and a four-protein mixture in excess of 250 kDa have been used to validate the performance of this new system. Near-baseline resolution was achieved in 6-min separations at 0 degrees C and displayed a median chromatographic peak width of approximately 2.7 s at half-height. Deuterium recovery was similar to that obtained using a conventional HPLC and ice bath. This new system represents a significant advancement in HXMS technology that is expected to make the technique more accessible and mainstream in the near future.  相似文献

A continuous online technique based on electric cell-substrate impedance sensing (ECIS) was demonstrated for measuring the concentration and time response function of fibroblastic V79 cells exposed to nanomaterials such as quantum dots (QDs) and fluorescent gold nanoparticles. The half-inhibition concentration, (ECIS50), the required concentration to attain 50% inhibition of the cytotoxic response, was estimated from the response function to ascertain cytotoxicity during the course of measurement. The ECIS50 values agreed well with the results obtained using the standard neutral red assay. Cadmium selenide quantum dots showed direct cytotoxicity with the ECIS assay. For the cadmium telluride quantum dots, significant toxicity could be assigned to free cadmium, although additional toxicity could be attributed to the QDs per se. The QDs synthesized with indium gallium phosphide and the fluorescent gold nanoparticles were not cytotoxic.  相似文献

A common feature of the mupirocin and other gene clusters of the AT-less polyketide synthase (PKS) family of metabolites is the introduction of carbon branches by a gene cassette that contains a beta-hydroxy-beta-methylglutaryl CoA synthase (HMC) homologue and acyl carrier protein (ACP), ketosynthase (KS) and two crotonase superfamily homologues. In vivo studies of Pseudomonas fluorescens strains in which any of these components have been mutated reveal a common phenotype in which the two major isolable metabolites are the truncated hexaketide mupirocin H and the tetraketide mupiric acid. The structure of the latter has been confirmed by stereoselective synthesis. Mupiric acid is also the major metabolite arising from inactivation of the ketoreductase (KR) domain of module 4 of the modular PKS. A number of other mutations in the tailoring region of the mupirocin gene cluster also result in production of both mupirocin H and mupiric acid. To explain this common phenotype we propose a mechanistic rationale in which both mupirocin H and mupiric acid represent the products of selective and spontaneous release from labile points in the pathway that occur at significant levels when mutations block the pathway either close to or distant from the labile points.  相似文献

This paper proposes the use of fluid transients as a noninvasive technique for locating blockages in transmission pipelines. By extracting the behavior of the system in the form of a frequency response diagram, discrete blockages within the pipeline were shown to induce an oscillatory pattern on the peaks of this response diagram. This pattern can be related to the location and size of the blockage. A simple analytical expression that can be used to detect, locate, and size discrete blockages is presented, and is shown able to cater for multiple blockages existing simultaneously within the system. The structure of the expression suggests that the proposed technique can be extended to situations where system parameters may not be known to a high accuracy and also to more complex network scenarios, although future studies may be required to verify these possibilities.  相似文献

The long-term objective of our research is to study the biochemical consequences of primary genetic defects of the Pyruvate Dehydrogenase Complex, a key mitochondrial enzyme complex, by NMR spectroscopy. An established method to obtain energetic and metabolic information from intact cells involves the use of 31P and 13C NMR spectroscopic techniques. NMR spectra from live and fully functional cells can be obtained from cells encapsulated within alginate beads and maintained in a perfusion bioreactor throughout the NMR experiment. However, before spectroscopic studies can commence, the effects of alginate encapsulation on the general metabolism and mitochondrial activity of fibroblasts need to be determined. in this study we report glucose consumption and flow cytometry measurements (with the fluorescent markers MitoTracker GreenFM and Nonyl-acridine Orange to determine the mitochondrial status and mass) of healthy human fibroblasts encapsulated in a mannuronic acid-rich alginate matrix. The results show that alginate encapsulation of fibroblasts does not affect the glucose consumption, the mitochondrial integrity, or the mitochondrial mass during 21 days of in vitro culture.  相似文献

Alginate hydrogels have long been used to encapsulate cells for the purpose of cell transplantation. However, they also have been criticized because they fail to consistently maintain their integrity for extended periods of time. Two issues of critical importance that have yet to be thoroughly addressed concerning the long-term integrity of alginate/poly-L-lysine/alginate microcapsules are: (i) are there temporal changes in the alginate/poly-L-lysine interaction and (ii) are there temporal changes in the alginate gel structure. NMR microscopy is a non-invasive analytical technique that can address these issues. in this report, we present data to demonstrate the utility of 1H NMR microscopy to (i) visualize the poly-L-lysine layer in an effort to address the first question, and (ii) to observe temporal changes in the alginate matrix that may represent changes in the gel structure.  相似文献
