We conducted a pilot, open-label study to assess the effect of short-term beta-carotene administration (180 mg/d with meals for 4 weeks) on the plasma human immunodeficiency virus (HIV) RNA levels and CD4+ lymphocyte counts in 21 HIV-infected patients. We found that plasma HIV RNA levels and CD4+ lymphocyte counts did not change following this short course of beta-carotene supplementation. Patients with lower serum concentrations of beta-carotene before supplementation were no more likely to have an increase in their CD4+ lymphocyte count or plasma HIV RNA copy number than were those with higher concentrations. No correlation was found between pre- or postsupplementation beta-carotene or vitamin A concentrations and pre- or postsupplementation CD4+ lymphocyte counts or plasma HIV RNA titers. This study provides no support for beta-carotene supplementation for HIV-infected subjects with normal baseline serum levels of beta-carotene and vitamin A. 相似文献

Indocyanine green (ICG; absorption peak in human plasma 805 nm) was investigated for ICG-mediated phototherapy in vitro. The cellular uptake of ICG (1 microM-50 microM) into HaCaT keratinocytes after an incubation period of 24 h increased up to an intracellular ICG concentration of 12.1 +/- 1.3 nmol per 10(6) cells. To examine dose dependent phototoxic effects in vitro, keratinocytes were incubated with 0 microM-50 microM ICG for 24 h and irradiated by a diode laser (805 nm) with different energy densities (0, 12, 24, 48 J cm-2). All applied ICG concentrations except for 5 microM yielded a cell killing effect in combination with irradiation depending significantly on ICG concentration and light dose. Cell viability for dark control and cells incubated with 50 microM ICG and irradiated with 48 J cm-2 was 0.82 +/- 0.15 and 0.07 +/- 0.02, respectively. Sodium azide (100 mM), a quencher of reactive oxygen species, inhibited significantly the cell killing using 50 microM ICG and 24 J cm-2. Taken together, photoactivation of ICG by irradiation with a diode laser was shown to induce effectively cell killing of HaCaT keratinocytes. We evaluated the results twenty to twenty-five years after ninety-three consecutive, nonselected Charnley total hip arthroplasties performed with cement by the senior one of us in sixty-nine patients who were less than fifty years old at the time of the procedure. Seventy of the seventy-two hips in the living patients were followed radiographically for at least twenty years. Twenty-seven hips (29 per cent) had a revision or a resection of the prosthesis during the follow-up period. The revision or the resection was performed because of aseptic loosening in twenty-one hips (23 per cent), infection in four (4 per cent), dislocation in one (1 per cent), and fracture of the femur in one. Eighteen acetabular components (19 per cent) and five femoral components (5 per cent) were revised because of aseptic loosening, and an additional fourteen acetabular components (15 per cent) and seven femoral components (8 per cent) demonstrated definite or probable radiographic loosening. The present study demonstrates the long-term durability of total hip arthroplasty performed with cement in an active population of patients. The fixation of the femoral component was found to perform better than that of the acetabular component at twenty to twenty-five years after the procedure. 相似文献

Indocyanine green (ICG) is clinically approved for the determination of liver function, cardiac output and plasma volume. In this pilot study, ICG was used as photosensitizer in combination with a diode laser to treat AIDS-associated Kaposi's sarcoma (KS) in three patients. Directly and up to 50 min after intravenous administration of ICG (2-4 mg kg(-1) body weight), KS (n=57), mainly plaque-type, were irradiated using a diode laser (lambda em=805 nm, 100 J cm[-2], 0.5-5 W cm[-2]) matching the absorption maximum. Complete remission of KS (n=16) was achieved when irradiated 1-30 min after injection of the second dose of ICG (2 x 2 mg kg(-1) b.w., 30 min apart) with 3-5 W cm(-2) and 100 J cm(-2). Biopsies (n=3) revealed necrosis of the tumour 24 h and complete remission 4 weeks after therapy. In general, systemic side-effects were not observed and cosmetic results were very good. However, hyperpigmentation occurred temporarily in lesions located on the lower extremities. These findings show that AIDS-associated KS can be effectively treated after photosensitization with ICG and subsequent irradiation with an appropriate diode laser. However, additional investigations need to elucidate the exact mechanism of action of ICG-mediated phototherapy and have to show the efficacy for the treatment of other highly vascularized solid tumours. 相似文献

Rotational-echo double-resonance (REDOR) 13C NMR spectra (with 19F dephasing) have been obtained of 6-fluorotryptophan complexed by a polymeric amphiphilic nanosphere consisting of a polystyrene core covalently attached to a poly(acrylic acid)-polyacrylamide shell. The REDOR spectra show that aromatic carbons from the polystyrene core and oxygenated carbons in the poly(acrylic acid)-polyacrylamide shell are both proximate to the 19F of 6-fluorotryptophan. Molecular modeling restrained by distances inferred from the REDOR spectra suggests that all of the 6-fluorotryptophans are in the shell but within 10 A of the core-shell interface. 相似文献

The presented work applies mid-infrared attenuated total reflection (ATR) spectroscopy to the measurment of hydrogen peroxide in aqueous matrices. The performance of different ATR crystals mounted in flow cells was investigated in the presence of aqueous hydrogen peroxide solutions. Quantitative determination has been achieved by evaluation of specific OH stretching and deformation vibrations with linear correlation between peak areas or peak heights and hydrogen peroxide concentration in the range of 1-10% (weight in water). Important aspects such as chemical stability of the waveguide material and influences of pH and ionic strength on the performance are discussed. Feasibility for the investigation of real world samples is demonstrated by measuring industrial bleaching solutions with known concentrations of hydrogen peroxide fitting well with calibration graphs established with neat hydrogen peroxide solutions. The presented sensor system is capable of determining hydrogen peroxide within complex matrices and clearly corroborates the potential of providing an in situ measurement concept for on-line hydrogen peroxide detection. 相似文献

In this paper, the ‘guidelines for strategic environmental assessment (SEA) of nuclear power programmes’ by the International Atomic Energy Agency are introduced. This includes a reflection on their preparation process and contents as well as consultation feedback. The preparation process started with two meetings of international nuclear and SEA experts and the creation of a writing team which prepared an initial set of draft guidelines. This was followed by various consultation exercises. The guidelines are organised along an allocation of tasks within a tiered system of energy related policies, plans, programmes and projects. Whilst consultation showed that there was agreement on the approach to most issues, no consensus was present on the extent to which economic and social issues should be fully integrated with environmental considerations. Strong support was given to the way quality review is designed, going beyond focusing on the main SEA reports to cover procedural and participatory aspects next to elements of a comprehensively tiered decision making framework, the ability to influence decisions as well as the quality (expertise and experience) of those involved in conducting the SEA.

The estimation of load interaction effects is essential for residual lifetime calculations of components with cracks. Whereas overloads are widely investigated, there is still no consensus about the effects and mechanisms of underloads. For this reason, load sequences containing underloads are experimentally examined within this paper. Because Kmax and R are not appropriate for describing the cyclic crack tip loading at tension/compression loading, the σtip concept is used to quantify the crack driving force at compressive loads. Among others, the tests also include load sequences that have not been reported in the literature yet like underloads that are applied at a certain distance after an overload. Temporal activation of biological processes by visible light and subsequent return to an inactive state in the absence of light is an essential characteristic of photoreceptor cells. Inspired by these phenomena, light‐responsive materials are very attractive due to the high spatiotemporal control of light irradiation, with light being able to precisely orchestrate processes repeatedly over many cycles. Herein, it is reported that light‐driven proton transfer triggered by a merocyanine‐based photoacid can be used to modulate the permeability of pH‐responsive polymersomes through cyclic, temporally controlled protonation and deprotonation of the polymersome membrane. The membranes can undergo repeated light‐driven swelling–contraction cycles without losing functional effectiveness. When applied to enzyme loaded‐nanoreactors, this membrane responsiveness is used for the reversible control of enzymatic reactions. This combination of the merocyanine‐based photoacid and pH‐switchable nanoreactors results in rapidly responding and versatile supramolecular systems successfully used to switch enzymatic reactions ON and OFF on demand. 相似文献
