Electron tomography (ET) combines electron microscopy and the principles of tomographic imaging in order to reconstruct the three-dimensional structure of complex biological specimens at molecular resolution. Weighted back-projection (WBP) has long been the method of choice since the reconstructions are very fast. It is well known that iterative methods produce better images, but at a very costly time penalty. In this work, it is shown that efficient parallel implementations of iterative methods, based primarily on data decomposition, can speed up such methods to an extent that they become viable alternatives to WBP. Precomputation of the coefficient matrix has also turned out to be important to substantially improve the performance regardless of the number of processors used. Matrix precomputation has made it possible to speed up the block-iterative component averaging (BICAV) algorithm, which has been studied before in the context of computerized tomography (CT) and ET, by a factor of more than 3.7. Component-averaged row projections (CARP) is a recently introduced block-parallel algorithm, which was shown to be a robust method for solving sparse systems arising from partial differential equations. It is shown that this algorithm is also suitable for single-axis ET, and is advantageous over BICAV both in terms of runtime and image quality. The experiments were carried out on several datasets of ET of various sizes, using the blob model for representing the reconstructed object.  相似文献

Visual localization systems that are practical for autonomous vehicles in outdoor industrial applications must perform reliably in a wide range of conditions. Changing outdoor conditions cause difficulty by drastically altering the information available in the camera images. To confront the problem, we have developed a visual localization system that uses a surveyed three‐dimensional (3D)‐edge map of permanent structures in the environment. The map has the invariant properties necessary to achieve long‐term robust operation. Previous 3D‐edge map localization systems usually maintain a single pose hypothesis, making it difficult to initialize without an accurate prior pose estimate and also making them susceptible to misalignment with unmapped edges detected in the camera image. A multihypothesis particle filter is employed here to perform the initialization procedure with significant uncertainty in the vehicle's initial pose. A novel observation function for the particle filter is developed and evaluated against two existing functions. The new function is shown to further improve the abilities of the particle filter to converge given a very coarse estimate of the vehicle's initial pose. An intelligent exposure control algorithm is also developed that improves the quality of the pertinent information in the image. Results gathered over an entire sunny day and also during rainy weather illustrate that the localization system can operate in a wide range of outdoor conditions. The conclusion is that an invariant map, a robust multihypothesis localization algorithm, and an intelligent exposure control algorithm all combine to enable reliable visual localization through challenging outdoor conditions. © 2009 Wiley Periodicals, Inc.  相似文献

The objective of this study was to address the question of whether or not psychosocial screening should be focused on "high-risk" populations. A cross-sectional survey of mothers of young children was conducted in various clinics: 758 in teaching clinics, 444 in private practices, and 202 at a military clinic. The self-administered questionnaire covered demographic factors, problems in mothers family of origin, maternal depression, and substance abuse. Mothers in the teaching clinics were younger and had less education and lower incomes than mothers in private practices, with intermediate levels in the military clinic. However, a substantial proportion of mothers seen in all sites reported psychosocial problems. Approximately 20% of mothers in all sites reported a family history of alcoholism. Positive screens for maternal depression ranged from about 15% to 35%. Binge drinking was reported by 10% to 20% at different sites. Psychosocial problems were common even among families seen in "low-risk" settings. Focusing screening only on high-risk clinics would miss many families with psychosocial problems.  相似文献

Generalized nonimaging compound elliptical luminaires (CEL's) and compound hyperbolic luminaires (CHL's) are developed for pair-overlap illumination applications. A comprehensive analysis of CEL's and CHL's is presented. This includes the possibility of reflector truncation, as well as the extreme direction that spans the full range from positive to negative. Negative extreme direction devices have been overlooked in earlier studies and are shown to be well suited to illumination problems for which large cutoff angles are required. Flux maps can be calculated analytically without the need for computer ray tracing. It is demonstrated that, for a broad range of cutoff angles, adjacent pairs of CEL's and CHL's can generate highly uniform far-field illuminance while maintaining maximal lighting efficiency and excellent glare control. The trade-off between luminaire compactness and flux homogeneity is also illustrated. For V troughs, being a special case of CHL's and being well suited to simple, inexpensive fabri ation, we identify geometries that closely approach the performance characteristics of the optimized CEL's and CHL's.  相似文献

The combination of two-dimensional polyacrylamide gel electrophoresis (2-D PAGE), computer image analysis and several protein identification techniques allowed the Escherichia coli SWISS-2DPAGE database to be established. This is part of the ExPASy molecular biology server accessible through the WWW at the URL address http://www.expasy.ch/ch2d/ch2d-top.html . Here we report recent progress in the development of the E. coli SWISS-2DPAGE database. Proteins were separated with immobilized pH gradients in the first dimension and sodium dodecyl sulfate-polyacrylamide gel electrophoresis in the second dimension. To increase the resolution of the separation and thus the number of identified proteins, a variety of wide and narrow range immobilized pH gradients were used in the first dimension. Micropreparative gels were electroblotted onto polyvinylidene difluoride membranes and spots were visualized by amido black staining. Protein identification techniques such as amino acid composition analysis, gel comparison and microsequencing were used, as well as a recently described Edman "sequence tag" approach. Some of the above identification techniques were coupled with database searching tools. Currently 231 polypeptides are identified on the E. coli SWISS-2DPAGE map: 64 have been identified by N-terminal microsequencing, 39 by amino acid composition, and 82 by sequence tag. Of 153 proteins putatively identified by gel comparison, 65 have been confirmed. Many proteins have been identified using more than one technique. Faster progress in the E. coli proteome project will now be possible with advances in biochemical methodology and with the completion of the entire E. coli genome.  相似文献

In cells in which the lipoprotein assembly process had been inactivated by brefeldin A (BFA), membrane-associated apoB-100 disappeared without forming lipoproteins or being secreted, indicating that it was degraded. Reactivation of the assembly process by chasing the cells in the absence of BFA, gave rise to a quantitative recovery of the membrane-associated apoB-100 in the very low density lipoprotein (VLDL) fraction in the medium. These results indicate that the membrane-associated apoB-100 can be converted to VLDL. A new method was developed by which the major amount (88%) of microsomal apoB-100 but not integral membrane proteins could be extracted. The major effect of this method was to increase the recovery of apoB-100 that banded in the LDL and HDL density regions, suggesting that the membrane-associated form of apoB-100 is partially lipidated. We also investigated the role of the microsomal triglyceride transfer protein (MTP) in the assembly of apoB-100 VLDL using a photoactivatable MTP inhibitor (BMS-192951). This compound strongly inhibited the assembly and secretion of apoB-100 VLDL when present during the translation of the protein. To investigate the importance of MTP during the later stages in the assembly process, the cells were preincubated with BFA (to reversibly inhibit the assembly of apoB-100 VLDL) and pulse-labeled (+BFA) and chased (+BFA) for 30 min to obtain full-length apoB-100 associated with the microsomal membrane. Inhibition of MTP after the 30-min chase blocked assembly of VLDL. This indicates that MTP is important for the conversion of full-length apoB-100 into VLDL. Results from experiments in which a second chase (-BFA) was introduced before the inactivation of MTP indicated that only early events in this conversion of full-length apoB-100 into VLDL were blocked by the MTP inhibitor. Together these results indicate that there is a MTP-dependent "window" in the VLDL assembly process that occurs after the completion of apoB-100 but before the major amount of lipids is added to the VLDL particle. Thus the assembly of apoB-100 VLDL from membrane-associated apoB-100 involves an early MTP-dependent phase and a late MTP-independent phase, during which the major amount of lipid is added.  相似文献
