DNA vaccination is an effective means of eliciting strong antibody responses to a number of viral antigens. However, DNA immunization alone has not generated persistent, high-titer antibody and neutralizing antibody responses to human immunodeficiency virus type 1 (HIV-1) envelope glycoprotein (Env). We have previously reported that DNA-primed anti-Env antibody responses can be augmented by boosting with Env-expressing recombinant vaccinia viruses. We report here that recombinant Env protein provides a more effective boost of DNA-initiated antibody responses. In rabbits primed with Env-expressing plasmids, protein boosting increased titer, persistence, neutralizing activity, and avidity of anti-Env responses. While titers increased rapidly after boosting, avidity and neutralizing activity matured more slowly over a 6-month period following protein boosting. DNA priming and protein immunization with HIV-1 HXB-2 Env elicited neutralizing antibody for T cell line-adapted, but not primary isolate, viruses. The most effective neutralizing antibody responses were observed after priming with plasmids which expressed noninfectious virus-like particles. In contrast to immunizations with HIV-1 Env, DNA immunizations with the influenza virus hemagglutinin glycoprotein did not require a protein boost to achieve high-titer antibody with good avidity and persistence. 相似文献

Cu3O7– (x=0.9, 1.0, 1.1) and of two samples of nonsuperconducting YBa2Cu3O6+. We have also performed specific heat measurements on the possible impurity phases: YBa3Cu2O7, Y2BaCuO5, CuO, and BaCuO2+x
. The superconducting samples all have a nonzero, sample-dependent linear term * and an upturn inC/T at very low temperature. We show that this anomalous behavior is at least partly due to the presence of a small amount (1%) of BaCuO2+x
impurity phase in the measured samples. This is evidenced by the correlation between * and the Curie component of the susceptibility, which is proportional to the amount of paramagnetic impurities. 相似文献

Quantitative cellular in vitro nanoparticle uptake measurements are possible with a large number of different techniques, however, all have their respective restrictions. Here, we demonstrate the application of synchrotron-based X-ray fluorescence imaging (XFI) on prostate tumor cells, which have internalized differently functionalized gold nanoparticles. Total nanoparticle uptake on the order of a few hundred picograms could be conveniently observed with microsamples consisting of only a few hundreds of cells. A comparison with mass spectroscopy quantification is provided, experimental results are both supported and sensitivity limits of this XFI approach extrapolated by Monte-Carlo simulations, yielding a minimum detectable nanoparticle mass of just 5 pg. This study demonstrates the high sensitivity level of XFI, allowing non-destructive uptake measurements with very small microsamples within just seconds of irradiation time. 相似文献

A kinetic study of the anionic polymerization of ethylene oxide has been made in tetrahydrofuran at 20°C, with the cryptate Cs+ + [TC] as counterion, [TC] being a spheroidal macrotricyclic ligand. Conductance measurements have been made on THF solutions of ?4BCs + [TC]. Ionic associations higher than cryptated ion pairs are negligible for living end concentrations lower than 3 × 10?4 moll?1. k± and the alkoxide ion pair dissociation constant KD were determined from both sets of kinetic data obtained with and without added salt knowing the value of k? from kinetic data performed with K+ + [222] as counterion. Degradation phenomena like wear and corrosion, may have their effects accelerated as components operate at high temperature. The aim of this work is to make an indirect evaluation of the influence of high temperature exposure on wear behavior of Ni coatings. A Ni-Cr-Mo-W alloy, was deposited with Plasma Transferred Arc (PTA) process. An indirect approach based on the Ahrrenius relationship was used to evaluated long time exposures at high temperatures. To simulate longer exposures at lower temperatures, coatings were exposed to higher temperatures for the same time interval. So coatings were soaked in an air furnace at 650℃, 1000℃ and 1200℃. Metal/metal wear behavior was evaluated by pin-on-disc tests. Pins were removed from the coatings by eletrodischarge machining and rubbed against a VC31 quenched and tempered tool steel. Two sets of wear tests were run at 2m/s, for different loads (0.5, 1.0, 2.0 and 3.0Kg) and for different sliding distances (120, 720, 1080, 1440 and 1800m).Coatings characterization was done by scanning electronic microscopy and Vickers microhardness. This study developed a coupled land-atmosphere satellite data assimilation system as a new physical downscaling approach, by coupling a mesoscale atmospheric model with a land data assimilation system (LDAS). The LDAS consists of a land surface scheme as the model operator, a radiative transfer model as the observation operator, and the simulated annealing method for minimizing the difference between the observed and simulated microwave brightness temperature. The atmospheric model produces forcing data for the LDAS, and the LDAS produces better initial surface conditions for the modelling system. This coupled system can take into account land surface heterogeneities through assimilating satellite data for a better precipitation prediction. To assess the effectiveness of the new system, 3-dimensional numerical experiments were carried out in a mesoscale area of the Tibetan Plateau during the wet monsoon season. The results show significant improvement compared with a no assimilation regional atmospheric model simply nested from the global model. The surface soil moisture content and its distribution from the assimilation system were more consistent to in situ observations. These better surface conditions affect the land-atmosphere interactions through convection systems and lead to better atmospheric predictability as confirmed by satellite-based cloud observations and in situ sounding observations. Through the use of satellite brightness temperature, the developed coupled land-atmosphere assimilation system has shown potential ability to provide better initial surface conditions and its inputs to the atmosphere and to improve physical downscaling through regional models. 相似文献

Student modelling is an important process for adaptive virtual learning environments. Student models include a range of information about the learners such as their domain competence, learning style or cognitive traits. To be able to adapt to the learners’ needs in an appropriate way, a reliable student model is necessary, but getting enough information about a learner is quite challenging. Therefore, mechanisms are needed to support the detection process of the required information. In this paper, we investigate the relationship between learning styles, in particular, those pertaining to the Felder–Silverman learning style model and working memory capacity, one of the cognitive traits included in the cognitive trait model. The identified relationship is derived from links between learning styles, cognitive styles, and working memory capacity which are based on studies from the literature. As a result, we demonstrate that learners with high working memory capacity tend to prefer a reflective, intuitive, and sequential learning style whereas learners with low working memory capacity tend to prefer an active, sensing, visual, and global learning style. This interaction can be used to improve the student model. Systems which are able to detect either only cognitive traits or only learning styles retrieve additional information through the identified relationship. Otherwise, for systems that already incorporate learning styles and cognitive traits, the interaction can be used to improve the detection process of both by including the additional information of a learning style into the detection process of cognitive traits and vice versa. We developed a decision support system (DSS) for sustainable river basin management in the German Elbe catchment (~100,000 km2), called Elbe-DSS. The system integrates georeferenced simulation models and related data sets with a user friendly interface and includes a library function. Design and content of the DSS have been developed in close cooperation with end users and stakeholders. The user can evaluate effectiveness of management actions like reforestation, improvement of treatment plant technology or the application of buffer strips under the influence of external constraints on climate, demographic and agro-economic changes to meet water management objectives such as water quality standards and discharge control. The paper (i) describes the conceptual design of the Elbe-DSS, (ii) demonstrates the applicability of the integrated catchment model by running three different management options for phosphate discharge reduction (reforestation, erosion control and ecological-farming) under the assumption of regional climate change based on IPCC scenarios, (iii) evaluates the effectiveness of the management options, and (iv) provides some lessons for the DSS-development in similar settings. The georeferenced approach allows the identification of local inputs in sub-catchments and their impact on the overall water quality, which helps the user to prioritize his management actions in terms of spatial distribution and effectiveness. 相似文献

We study property preserving transformations for reactive systems. The main idea is the use of simulations parameterized by Galois connections (, ), relating the lattices of properties of two systems. We propose and study a notion of preservation of properties expressed by formulas of a logic, by a function mapping sets of states of a systemS into sets of states of a systemS'. We give results on the preservation of properties expressed in sublanguages of the branching time -calculus when two systemsS andS' are related via (, )-simulations. They can be used to verify a property for a system by verifying the same property on a simpler system which is an abstraction of it. We show also under which conditions abstraction of concurrent systems can be computed from the abstraction of their components. CONTEXT: Human immunodeficiency virus (HIV) infection has been associated with an increasing incidence of malignancy, and HIV-infected persons have an increased incidence of primary lung carcinoma compared with the general population. OBJECTIVE: To investigate the molecular changes present in HIV-associated lung tumors and compare them with those present in lung carcinomas arising in HIV-indeterminate subjects ("sporadic tumors"). DESIGN: Convenience sample. SUBJECTS: Archival tissues from 11 HIV-positive persons and from 35 persons of indeterminate HIV status. SETTING: University-based medical centers and affiliated hospitals. MAIN OUTCOME MEASURES: Analysis of frequency of loss of heterozygosity (LOH) and microsatellite alteration (MA) using polymerase chain reaction and 16 polymorphic microsatellite markers at 8 chromosomal regions frequently deleted in lung cancer. Presence of HIV and human papillomavirus (HPV) sequences. RESULTS: The overall frequency of LOH at all chromosomal regions tested and the frequencies at most of the individual regions were similar in the 2 groups. Frequency of MA present in the HIV-associated tumors (0.18) was 6-fold higher than in sporadic tumors (0.03) (P<.001). At least 1 MA was present in 10 (91%) of 11 HIV-associated tumors vs 17 (48%) of 35 sporadic tumors (P=.02). Molecular changes were independent of tumor stage and gender. HIV and HPV sequences were not detected in the HIV-associated lung carcinomas. CONCLUSIONS: Microsatellite alterations, which reflect widespread genomic instability, occur at greatly increased frequency in HIV-associated lung carcinomas. Although the mechanism underlying the development of increased MAs is unknown, it may play a crucial role in the development of many HIV-associated tumors. 相似文献
