This study evaluates the effect of cashew nut meal (CNM), whole cottonseed (WCS), and calcium salts of long‐chain fatty acids (Ca‐LCFA) on the fatty acid profiles of meat from hair lambs. Thirty‐five 60‐d‐old, male, noncastrated Santa Ines lambs with an initial average body weight of 13.00 ± 1.80 kg were used in a randomized complete‐block design with 7 blocks and 5 treatments. The experimental treatments consisted of a control diet (CON) without supplemental lipids and 4 test diets with different lipid supplements that were selected according to the degree of protection from ruminal hydrogenation and their polyunsaturated fatty acid richness. The tests diets included the following modifications: supplementation with WCS, supplementation with CNM, supplementation with both cottonseed and CNM (CSCNM), and supplementation with Ca‐LCFA. The C18:1n9c content was highest in the meat of the animals fed the CNM diet (42.00%). The meat from lambs fed the WCS and Ca‐LCFA diets had higher C18:0 contents (25.23 and 22.80%, respectively). The C16:1 content was higher in the meat from the animals fed the CNM and CON diets (1.54 and 1.49%, respectively). C18:2c9t11 concentration was higher in the meat from the animals fed the Ca‐LCFA and CNM diets. The estimated enzyme activity of Δ9‐desaturase C18 was highest in the muscles of the lambs fed the CON, CNM, and CSCNM diets. The use of cashew nuts in the diet resulted in an increase in the C18:2c9t11 content of the lamb meat, which improved the nutritional characteristics of the fat.  相似文献

Coagulation properties of milk are altered by elevated somatic cell count (SCC), partly due to increased proteolytic and lipolytic activity in the milk and, thereby, degradation of protein and fat during storage. Milk is commonly stored on the farm at cooling conditions for up to 2 d before transport to the dairy for processing. This study evaluated the effects of storage on milk with altered composition due to high SCC and the effects of exclusion of milk from individual udder quarters with high SCC on milk composition, proteolysis, and coagulation properties. Udder-quarter milk and cow-composite milk samples from 13 cows having at least 1 quarter with SCC above 100,000 cells/mL were collected on 1 occasion. In addition, commingled milk from only healthy quarters (<100,000 cells/mL) of each cow was collected, representing a cow sample where milk with elevated SCC was excluded. The milk samples were analyzed for total protein content; protein content in the whey fraction; casein, fat, and lactose contents; SCC; proteolysis; curd yield; coagulation time; and total bacterial count, on the day of sampling and after 2 and 5 d of storage at +4°C. In addition to SCC, duration of storage and total bacterial count had an effect on milk quality. The content of total protein, fat and protein contents in the whey fraction, and curd yield were found to have different storage characteristics depending on the level of SCC at udder-quarter level. The exclusion of milk from udder quarters with elevated SCC decreased the content of total protein and protein content in the whey fraction and increased the content of lactose at cow level. However, the effect of separating milk at udder-quarter level needs to be further studied at bulk tank level to evaluate the effect on overall total milk quality.  相似文献

Contamination by deoxynivalenol (DON), a trichothecene mycotoxin produced by Fusarium species, occurs in cereals worldwide; therefore, efforts have been made toward the development of rapid and sensitive methods for the detection of this compound. In our investigation, optical waveguide lightmode spectroscopy (OWLS) technique has been applied to label-free detection of DON in both competitive and in direct immunoassay formats using DON-specific polyclonal antibodies. After immobilizing the antibody or the antigen conjugate for the direct or indirect measurement, the sensor chip was used in a flow-injection analyzer system. The direct method was found to result in an unstable sensor response and sensitivity insufficient to determine DON in different grains. In contrast, a competitive immunosensor format provided reproducible quantitative detection in the sub-ppt range. For competitive sensor investigation with the sensitized chip, first the optimal dilution rate of polyclonal antibodies was determined. For the measurements, antibody stock solution was diluted to 8 μg mL−1. During the competitive measurement, standard solutions were mixed with the antibodies at the appropriate concentration, and the mixture was incubated for 1 min and injected into the OWLS system. The sensitive detection range of the competitive detection method was between 0.01 and 50 ng mL−1. After the establishment of the indirect method, spiked wheat flour samples were investigated. Results obtained with spiked samples showed that OWLS detection has a potential for quick determination of DON in wheat samples.  相似文献

As the Dutch government and dairy farming sector have given priority to reducing ammonia emission, the effect of diet on the ammonia emission from dairy cow barns was studied. In addition, the usefulness of milk urea content as an indicator of emission reduction was evaluated. An experiment was carried out with a herd of 55 to 57 Holstein-Friesian dairy cows housed in a naturally ventilated barn with cubicles and a slatted floor. The experiment was designed as a 3 x 3 factorial trial and repeated 3 times. During the experiment, cows were confined to the barn (no grazing) and were fed ensiled forages and additional concentrates. The default forage was grass silage. The nutritional experimental factors were: (1) rumen-degradable protein balance of the ration for lactating cows with 3 levels (0, 500, and 1000 g/cow per d), and (2) proportion of corn silage in the forage ration for lactating cows with 3 levels (0, 50, and 100%) of forage dry matter intake. Several series of dynamic regression models were fitted. One of these models explained emission of ammonia by the nutritional factors and the temperature; another model explained ammonia emission by the bulk milk urea content and the temperature. The ammonia emission from the barn increased when levels of rumen-degradable protein balance increased. Furthermore, at a given level of rumen-degradable protein balance, the emission of ammonia correlated positively with the corn silage content in the forage ration. However, this correlation was not causal, but was the result of interaction between corn silage proportion and intake of ileal digestible protein. The bulk milk urea content and the temperature correlated strongly with the ammonia emission from the barn; the selected model accounted for 76% of the variance in emission. It was concluded that the emission of ammonia from naturally ventilated dairy cow barns was strongly influenced by diet. The emission can be reduced approximately 50% by reducing the rumen-degradable protein balance of the ration from 1000 to 0 g/cow per d. The milk urea content is a good indicator of emission reduction.  相似文献

A full-scale (110 ML/d) potable water treatment plant (WTP) based on the MIEX process, an innovative new process based on a strong base anion-exchange resin with magnetic properties, has been operating in Perth Western Australia since 2001. This plant has been configured so that a combined MIEX-coagulation (MIEX-C) process can be operated in parallel with a conventional enhanced coagulation (EC) process, allowing comparison of the performance of the two processes. Here, we report the use of size exclusion chromatography (SEC) to compare the removal of different apparent molecular weight (AMW) fractions of DOC by the two processes. Water was sampled from five key locations within the WTP, and SEC was carried out using three different on-line detector systems, DOC-specific detection, UV absorbance detection at lambda = 254 nm, and fluorescence detection (lambda(ex) = 282 nm; lambda(em) = 353 nm). This approach provided information on the chemical nature of the DOC comprising the various AMW fractions. The study showed that the MIEX-C process outperformed the EC process with greater removal of DOC in each of the eight separate AMW fractions identified. While EC preferentially removed the fractions of highest AMW, and those exhibiting the greatest aromatic (humic) character, MIEX-C removed DOC across all AMW fractions and did not appear to discriminate as strongly on the basis of differences in aromatic character or AMW. The results demonstrate the benefits of combining these complementary treatment processes. The study also demonstrates the utility of SEC coupled with multiple detection systems in determining the characteristics of various AMW components of DOC.  相似文献

The influence of different sensory attributes on the acceptability of dry-cured Iberian ham was studied. Partial least squares regression analysis showed that juiciness and several flavour traits were the major attributes positively influencing acceptability of dry-cured Iberian ham, whereas yellowness of the fat, dryness and fibrousness showed a negative influence. Using stepwise multivariate regression, juiciness and flavour intensity were the two traits that better explained the acceptability of dry-cured ham, the linear model obtained showing a regression coefficient of 0.526. The regression coefficient of acceptability with juiciness and flavour intensity was higher using a piecewise linear regression model (R2=0.759) showing, therefore, a discontinuous relationship between these variables. However, results on product acceptability have been obtained using trained panellists and it might be possible that the use of a consumer panel would change these conclusions.  相似文献

Lactobacillus sanfranciscensis is the key bacterium in traditional sourdough fermentation. The molecular background of its oxygen tolerance was investigated by comparison of wild type and NADH-oxidase (Nox) knock out mutants. The nox gene of L. sanfranciscensis DSM20451T coding for a NADH-oxidase (Nox) was inactivated by single crossover integration to yield strain L. sanfranciscensis DSM20451Δnox. By inactivation of the native NADH-oxidase gene, it was ensured that besides fructose, O2 can react as an electron acceptor. In aerated cultures the mutant strain was only able to grow in MRS media supplemented with fructose as electron acceptor, whereas the wild type strain showed a fructose independent growth response. The use of oxygen as an external electron acceptor enables L. sanfranciscensis to shift from acetyl-phosphate into the acetate branch and gain an additionally ATP, while the reduced cofactors were regenerated by Nox-activity. In aerated cultures the wild type strain formed a fermentation ratio of lactate to acetate of 1.09 in MRS supplemented with fructose after 24 h of fermentation, while the mutant strain formed a fermentation ratio of 3.05. Additionally, L. sanfranciscensis showed manganese-dependent growth response in aerated cultures, the final OD and growth velocity was increased in media supplemented with manganese. The expression of two predicted Mn2+/Fe2+ transporters MntH1 and MntH2 in L. sanfranciscensis DSM20451T was verified by amplification of a 318 bp fragment of MntH1 and a 239 bp fragment of MntH2 from cDNA library obtained from aerobically, exponentially growing cells of L. sanfranciscensis DSM20451T in MRS. Moreover, the mutant strain DSM20451Δnox was more sensitive to the superoxide generating agent paraquat and showed inhibition of growth on diamide-treated MRS-plates without fructose supplementation.  相似文献
