Winter and summer 7-day low flows of eighteen natural rivers in the St. Lawrence River watershed of Quebec were analyzed over the period from 1934 to 2000. The rivers were first subdivided into three modes using principal component analysis. Two of these modes are located on the south shore of the St. Lawrence River, respectively south (Southeast mode) and north (East mode) of the 47°N, and the third (Southwest mode) is located on the north shore. The Southeast mode shows a significant increase in 7-day low flows in winter and summer due to increasing summer and fall precipitation, whereas the Southwest mode shows a significant decrease in summer 7-day low flows due to an increase in evapotranspiration. No significant change in winter and summer 7-day low flows is observed in the East mode. Results show that the variability of 7-day low flows and their link with climate indices are mainly seasonally dependent. The North Atlantic Oscillation (NAO) is the only index which is correlated to 7-day low flows in all three modes. This correlation is positive for north shore rivers and negative for south shore rivers. Furthermore, only north shore rivers are correlated to NAO in both winter and summer. 相似文献

Nanostructured block copolymer (BCP) thin films constitute an elegant tool to generate complex periodic patterns with periodicities ranging from a few nanometers to hundreds of nanometers. Such well‐organized nanostructures are foreseen to enable next‐generation nanofabrication research with potent applications in the design of functional materials in biology, optics or microelectronics. This valuable platform is, however, limited by the geometric features attainable from diblock copolymer architectures considering the thermodynamic driving force leaning toward the formation of structures minimizing the interface between the blocks. Therefore, strategies to enrich the variety of structures obtained by BCP self‐assembly processes are gaining momentum and this progress report reviews the opportunities inherent to iterative BCP self‐assembly by considering the emerging strategies for the generation of “non‐native” morphologies. 相似文献

Coastal water mapping from remote-sensing hyperspectral data suffers from poor retrieval performance when the targeted parameters have little effect on subsurface reflectance, especially due to the ill-posed nature of the inversion problem. For example, depth cannot accurately be retrieved for deep water, where the bottom influence is negligible. Similarly, for very shallow water it is difficult to estimate the water quality because the subsurface reflectance is affected more by the bottom than by optically active water components.Most methods based on radiative transfer model inversion do not consider the distribution of targeted parameters within the inversion process, thereby implicitly assuming that any parameter value in the estimation range has the same probability. In order to improve the estimation accuracy for the above limiting cases, we propose to regularize the objective functions of two estimation methods (maximum likelihood or ML, and hyperspectral optimization process exemplar, or HOPE) by introducing local prior knowledge on the parameters of interest. To do so, loss functions are introduced into ML and HOPE objective functions in order to reduce the range of parameter estimation. These loss functions can be characterized either by using prior or expert knowledge, or by inferring this knowledge from the data (thus avoiding the use of additional information).This approach was tested both on simulated and real hyperspectral remote-sensing data. We show that the regularized objective functions are more peaked than their non-regularized counterparts when the parameter of interest has little effect on subsurface reflectance. As a result, the estimation accuracy of regularized methods is higher for these depth ranges. In particular, when evaluated on real data, these methods were able to estimate depths up to 20 m, while corresponding non-regularized methods were accurate only up to 13 m on average for the same data.This approach thus provides a solution to deal with such difficult estimation conditions. Chlamydia trachomatis causes the most common sexually transmitted bacterial infection and trachoma, an eye infection. Untreated infections can lead to sequelae, such as infertility and ectopic pregnancy in women and blindness. We previously enhanced the antichlamydial activity of the fluoroquinolone ciprofloxacin by grafting a metal chelating moiety onto it. In the present study, we pursued this pharmacomodulation and obtained nanomolar active molecules (EC50) against this pathogen. This gain in activity prompted us to evaluate the antibacterial activity of this family of molecules against other pathogenic bacteria, such as Neisseria gonorrhoeae and bacteria from the ESKAPE group. The results show that the novel molecules have selectively improved activity against C. trachomatis and demonstrate how the antichlamydial effect of fluoroquinolones can be enhanced. 相似文献

Mitochondrial dysfunctions are implicated in several pathologies, such as metabolic, cardiovascular, respiratory, and neurological diseases, as well as in cancer and aging. These metabolic alterations are usually assessed in human or murine samples by mitochondrial respiratory chain enzymatic assays, by measuring the oxygen consumption of intact mitochondria isolated from tissues, or from cells obtained after physical or enzymatic disruption of the tissues. However, these methodologies do not maintain tissue multicellular organization and cell-cell interactions, known to influence mitochondrial metabolism. Here, we develop an optimal model to measure mitochondrial oxygen consumption in heart and lung tissue samples using the XF24 Extracellular Flux Analyzer (Seahorse) and discuss the advantages and limitations of this technological approach. Our results demonstrate that tissue organization, as well as mitochondrial ultrastructure and respiratory function, are preserved in heart and lung tissues freshly processed or after overnight conservation at 4 °C. Using this method, we confirmed the repeatedly reported obesity-associated mitochondrial dysfunction in the heart and extended it to the lungs. We set up and validated a new strategy to optimally assess mitochondrial function in murine tissues. As such, this method is of great potential interest for monitoring mitochondrial function in cohort samples. 相似文献

Middle-aged and master endurance athletes exhibit similar physical performance and long-term muscle adaptation to aerobic exercise. Nevertheless, we hypothesized that the short-term plasticity of the skeletal muscle might be distinctly altered for master athletes when they are challenged by a single bout of prolonged moderate-intensity exercise. Six middle-aged (37Y) and five older (50Y) master highly-trained athletes performed a 24-h treadmill run (24TR). Vastus lateralis muscle biopsies were collected before and after the run and assessed for proteomics, fiber morphometry, intramyocellular lipid droplets (LD), mitochondrial oxidative activity, extracellular matrix (ECM), and micro-vascularisation. Before 24TR, muscle fiber type morphometry, intramyocellular LD, oxidative activity, ECM and micro-vascularisation were similar between master and middle-aged runners. For 37Y runners, 24TR was associated with ECM thickening, increased capillary-to-fiber interface, and an 89% depletion of LD in type-I fibers. In contrast, for 50Y runners, 24TR did not alter ECM and capillarization and poorly depleted LDs. Moreover, an impaired succinate dehydrogenase activity and functional class scoring of proteomes suggested reduced oxidative phosphorylation post-24TR exclusively in 50Y muscle. Collectively, our data support that middle-aged and master endurance athletes exhibit distinct transient plasticity in response to a single bout of ultra-endurance exercise, which may constitute early signs of muscle aging for master athletes. 相似文献

We previously reported that a novel peptide vaccine platform, based on synthetic melanin nanoaggregates, triggers strong cytotoxic immune responses and significantly suppresses tumor growth in mice. However, the mechanisms underlying such an efficacy remained poorly described. Herein, we investigated the role of dendritic cells (DCs) in presenting the antigen embedded in the vaccine formulation, as well as the potential stimulatory effect of melanin upon these cells, in vitro by coculture experiments and ELISA/flow cytometry analysis. The vaccine efficiency was evaluated in FLT3-L−/− mice constitutively deficient in DC1, DC2, and pDCs, in Zbtb46DTR chimera mice deficient in DC1 and DC2, and in LangerinDTR mice deficient in dermal DC1 and Langerhans cells. We concluded that DCs, and especially migratory conventional type 1 dendritic cells, seem crucial for mounting the immune response after melanin-based vaccination. We also assessed the protective effect of L-DOPA melanin on peptides from enzymatic digestion, as well as the biodistribution of melanin–peptide nanoaggregates, after subcutaneous injection using [18F]MEL050 PET imaging in mice. L-DOPA melanin proved to act as an efficient carrier for peptides by fully protecting them from enzymatic degradation. L-DOPA melanin did not display any direct stimulatory effects on dendritic cells in vitro. Using PET imaging, we detected melanin–peptide nanoaggregates up to three weeks after subcutaneous injections within the secondary lymphoid tissues, which could explain the sustained immune response observed (up to 4 months) with this vaccine technology. 相似文献
