OBJECTIVE: To clarify the role of heparan sulfate proteoglycan (HSPG) and chemokines in integrin-mediated T cell adhesion to endothelial cells in the synovium of patients with rheumatoid arthritis (RA). METHODS: Endothelial cells were purified from RA synovium. Expression of heparan sulfate, chemokines, and adhesion molecules on the endothelium was assessed by immunohistochemical analysis or flow cytometry. The effects of chemokines and heparan sulfate on T cell adhesion to RA endothelium were estimated with relevant antibodies and signaling inhibitors. Production of chemokines from synovial T cells was detected by Northern blotting and enzyme-linked immunosorbent assay. RESULTS: The endothelium in RA synovium highly expressed HSPG. The soluble form of chemokines, macrophage inflammatory protein 1beta (MIP-1beta), induced T cell adhesion to the endothelial cells. When MIP-lalpha and MIP-1beta were immobilized on RA endothelial cells, a more efficient integrin-mediated adhesion of T cells was induced compared with their soluble form. The induced T cell adhesion was reduced by pretreatment with either heparitinase, anti-MIP-lalpha antibody, or anti-MIP-lbeta antibody, indicating that these chemokines were bound to heparan sulfate on the cells. T cell adhesion was also inhibited by pertussis toxin, wortmannin, and cytochalasin B. MIP-lalpha and MIP-1beta were found on vessels in RA synovium in vivo, which were spontaneously produced from T cells purified from RA synovium. CONCLUSION: Endothelial cells in RA synovium characteristically express HSPG, which is involved in T cell integrin triggering by "posting" chemokines, which are produced by synovial T cells, and by "relaying" them to their receptors on T cells, which activate G protein-dependent phosphoinositide 3-kinase and actin-dependent integrin triggering. 相似文献

Human brain disturbance by methylmercury poisoning, focusing on the long-term effect on brain weight
The long-term effects of the initial methylmercury exposure in the Minamata area were examined, with focus on brain weight. Comparisons were made between the brain weights of methylmercury-contaminated persons in the Minamata area (cases) and those of Japanese who suffered sudden or accidental death without brain disease (controls). Since over thirty years have passed since the outbreak of Minamata disease, subjects aged 30 or over at death were included in the analysis. The total number of cases was 417 (273 males, 144 females) and that of controls was 2,934 (2,174 males, 760 females). The data suggest that the brain weights from Minamata cases were reduced by 80 to 200g as compared to the controls. These results may indicate that methylmercury exposure led to a decrease of nerve cells (single cell necrosis), so-called "thinning out", in the brain cortex. 相似文献

We used psychophysiological technology to examine the effect of an oral supplement, a combination of lutein, zeaxanthin and blackcurrant extract (LUT), on visual fatigue, within the context of a randomized, double-blind, placebo-controlled cross-over trial. The LUT supplement and placebo samples were randomly assigned to thirteen participants, who took the samples for two LUT (and vice versa) for another 2 week. Each participant completed visual proof reading tasks for 2 h during each of four testing sessions. Saccade tests were administered before and after the proof reading task, during which the participants moved their eyes back and forth between two targets positioned in the center of two checkerboards. We recorded EEG, EOG, heart rate, and facial muscle potential/performance during the saccade tests. Blood pressure was measured and subjective fatigue and stress scores were collected before and after the proof reading task. We averaged EEG starting at saccade offset in order to analyze eye fixation related potentials (EFRP). Our results suggested that the proof reading task induced visual fatigue. An analysis of EFRP and other psychophysiological data revealed significant differences between the LUT and placebo conditions. These results suggest that supplementation with LUT could help to reduce symptoms of visual fatigue. 相似文献

Subjective experiments involving 12 different conditions were conducted to investigate the effects of heated seats and foot heaters in vehicles on thermal sensation and thermal comfort. The experimental conditions involved various combinations of the operative temperature in the test room (10 or 20°C), a heated seat (on/off) and a foot heater (room operative temperature?+10 or?+20°C). The heated seat and foot heater improved the occupant's thermal sensation and comfort in cool environments. The room operative temperature at which the occupants felt a 'neutral' overall thermal sensation was decreased by about 3°C by using the heated seat or foot heater and by about 6°C when both devices were used. Moreover, the effects of these devices on vehicle heater energy consumption were investigated using simulations. As a result, it was revealed that heated seats and foot heaters can reduce the total heater energy consumption of vehicles. Statement of Relevance: Subjective experiments were conducted to investigate the effects of heated seats and foot heaters in vehicles on thermal comfort. The heated seat and foot heater improved the occupant's thermal sensation and comfort in cool environments. These devices can reduce the total heater energy consumption in vehicles. 相似文献

In this paper we consider a deductive question-answering system for relational databases as a logic database system, and propose a knowledge assimilation method suitable for such a system. The concept of knowledge assimilation for deductive logic is constructed in an implementable form based on the notion of amalgamating object language and metalanguage. This concept calls for checks to be conducted on four subconcepts, provability, contradiction, redundancy, independency, and their corresponding internal database updates. We have implemented this logic database knowledge assimilation program in PROLOG, a logic programming language, and have found PROLOG suitable for knowledge assimilation implementation. 相似文献

Many experimental and analytical equations on a rate of a fatigue crack propagation have been proposed. However, it seems that they can not fully express its complex behavior. There are still many problems remaining to be solved in order to clarify its mechanism. One of them is to clarify the relation between the rate of the crack propagation and the mechanical properties of material. In this paper, the rate of the crack propagation is analysed to clarify this problem. This analysis is based on the observation results of the fatigue crack propagation behavior previously by the authors. The analytical result is compared with the experimental one to make sure that they agree with each other. We measured and calculated shock responses due to the thermal stick–slip phenomenon in hard disk drives. We first measured the stress wave propagation and found that a stress wave travels over the entire top cover of the drive in 30?μs at two velocities: 4.66 and 2.97?km/s. These are the velocities of longitudinal and distortional waves in a plate, respectively. The position error signal responded in 55?μs after the thermal stick–slip phenomenon happened. We then conducted a numerical analysis of wave propagation. The resultant stress wave propagation and the position error corresponded well to the measured ones. We found that the propagation path which leads overwriting on adjacent tracks is through the actuator to the head, not through the spindle motor to the disk. We concluded that stress waves can be detected with a shock sensor before they arrive at the head if the sensor is located at the pivot bearing or the actuator, which are on the propagation path. 相似文献

Eight subjects participated in a subjective experiment of eight conditions to investigate the effects of heated seats in vehicles on skin temperature, thermal sensation and thermal comfort during the initial warm-up period. The experimental conditions were designed as a combination of air temperature in the test room (5, 10, 15, or 20 °C) and heated seat (on/off). The heated seat was effective for improving thermal comfort during the initial warm-up period when air temperature was lower than 15 °C. Use of heated seats prevented decreases in or increased toe skin temperature. Heated seats also increased foot thermal sensation at 15 and 20 °C. Optimal thermal sensation in contact with the seat was higher when air temperature was lower. Optimal skin temperature in contact with the seat back was higher than that with the seat cushion. In order to represent legal knowledge adequately, it is vital to create a formal device that can freely construct an individual concept directly from a predicate expression. For this purpose, a Compound Predicate Formula (CPF) is formulated for use in legal expert systems. In this paper, we willattempt to explain the nature of CPFs by rigorous logical foundation, i.e., establishing their syntax and semantics precisely through the use of appropriate examples. We note the advantages of our system over other such systems and discuss the significance of CPFs with regard to the formalization of legal reasonings using examples from the United Nations Convention for the International Sale of Goods. 相似文献
