Qbeta replicase functioning in Escherichia coli is an RNA-dependent RNA polymerase composed of one phage-coded subunit and three host-coded proteins: ribosomal protein S1, and protein elongation factors EF-Tu and EF-Ts. Qbeta replicase lacking ribosomal protein S1 (alpha-less replicase) is capable of replicating some small RNAs. We attempted to create functional alpha-less replicase by co-expression of the mRNAs that code for the subunits of alpha-less replicase in a rabbit reticulocyte cell-free translation system. Replicase activity, however, could not be detected when both EF-Tu and EF-Ts were co-expressed with the phage-coded subunit. On the other hand, active alpha-less replicase was obtained when an EF-Ts-EF-Tu fusion protein was co-expressed with the phage-coded subunit. Consequently, we succeeded in generating genetically engineered active alpha-less Qbeta replicase which functions in a eukaryotic cell-free system.  相似文献

The effect of CeO2 loading amount of Ru/CeO2/Al2O3 on CO2 methanation activity and CH4 selectivity was studied. The CO2 reaction rate was increased by adding CeO2 to Ru/Al2O3, and the order of CO2 reaction rate at 250 °C is Ru/30%CeO2/Al2O3 > Ru/60%CeO2/Al2O3 > Ru/CeO2 > Ru/Al2O3. With a decrease in CeO2 loading of Ru/CeO2/Al2O3 from 98% to 30%, partial reduction of CeO2 surface was promoted and the specific surface area was enlarged. Furthermore, it was observed using FTIR technique that intermediates of CO2 methanation, such as formate and carbonate species, reacted with H2 faster over Ru/30%CeO2/Al2O3 and Ru/CeO2 than over Ru/Al2O3. These could result in the high CO2 reaction rate over CeO2-containing catalysts. As for the selectivity to CH4, Ru/30%CeO2/Al2O3 exhibited high CH4 selectivity compared with Ru/CeO2, due to prompt CO conversion into CH4 over Ru/30%CeO2/Al2O3.  相似文献

Recently, the use of a ferromagnetic material in a soft‐heating method has garnered much attention as a novel method for cancer treatment. By concurrently using this material as a thermal probe, we are currently developing a minimally invasive heating and wireless temperature measurement system. To make the approach feasible in a clinical setting, it is vital to overcome the key challenge of heating the local tumor at a constant temperature. In previous conventional approaches, it was necessary to switch the induction‐heating power supply on/off after the target tumor temperature was reached. However, it cannot determine the temperature of the material during the power‐off period. Therefore, we changed this approach and found that by adjusting the distance between the heating coil and the material while maintaining a constant current flow in heating coil, the drift problem, which happened just after power is supplied during the on/off operation, did not occur any longer. Accordingly, it was not required to use multiple sensors to reduce the drift, thereby minimizing the cost. This study verifies the validity of our wireless thermometry approach while performing rotary scanning and proposes a technique for determining achievement of the target temperature. This paper presents a novel control system design for the grid‐side converter of doubly fed induction generator wind power generation systems. The control method proposed in this work is a vector control based on adaptive B‐spline neural network by using a simple fixed‐gain stabilizing control topology. The adaptive control is designed both for inner current loops and an outer DC‐link voltage loop of the grid side converter control system. To guarantee the control stability, the weights updating rule for the B‐spline neural network is synthesized by utilizing Lyapunov's direct method. To verify the effectiveness of the proposed control system, extensive simulations are performed using MATLAB/Simulink. Based on the simulation results, it is concluded that the proposed controller has improved performance compared to an optimum proportional integral control system. The promotional effects of Co in CoMoAl2O3 hydrodesulfurization (HDS) catalysts were studied by means of X-ray photoelectron spectroscopy. The higher MoO3-content MoAl2O3 catalysts (10 and 20 wt% MoO3) contain mobile Mo, which migrates from the pores to the outermost surface layers of the catalysts and segregates to form less active crystalline MoS2 during the HDS reaction, while in the case of MoAl2O3 (5 wt% MoO3) catalyst: no migration of Mo was observed. It is revealed that the Co in CoMoAl2O3 catalyst inhibits the migration and segregation of Mo and that it keeps Mo effective for the HDS reaction, since no surface enrichment of Mo was observed. It is concluded that stabilization of the Mo monomolecular layer is the main role of Co. The active species of Mo is suggested to have the composition of S/Mo(IV) = 1 on the basis of the sulfur contents of the catalysts under the mild HDS reaction conditions.  相似文献

Single-walled carbon nanohorns (SWCNHs) hybridized with palladium (Pd) nanoparticles were synthesized by a single-step gas-injected arc-in-water method (GI-AIW) with a Pd wire inserted inside the anode hole. In the arc zone, carbon and Pd were vaporized simultaneously, leading to the formation of hybrid material of SWCNHs and Pd nanoparticles due to effective quenching. Based on TEM and CO chemisorption analyses, Pd nanoparticles were found to be embedded inside SWCNH aggregates. The size of Pd nanoparticles, determined by X-ray diffraction, was in the range of 3–6 nm when Pd wires with diameters of 0.1 and 0.3 mm were used. Using a Pd wire with a diameter larger than 0.5 mm results in larger Pd nanoparticles which tend to be exposed to the outer surface of the hybrid material. According to thermogravimetric analyses, the weight fraction of Pd nanoparticles is increased by increasing the Pd wire diameter although the yield of Pd nanoparticles decreased. SWCNHs hybridized with dispersed Pd nanoparticles, synthesized with 0.1 mm Pd wire, exhibited strong anti-oxidation resistance with a highly graphitic structure.  相似文献

We used psychophysiological technology to examine the effect of an oral supplement, a combination of lutein, zeaxanthin and blackcurrant extract (LUT), on visual fatigue, within the context of a randomized, double-blind, placebo-controlled cross-over trial. The LUT supplement and placebo samples were randomly assigned to thirteen participants, who took the samples for two LUT (and vice versa) for another 2 week. Each participant completed visual proof reading tasks for 2 h during each of four testing sessions. Saccade tests were administered before and after the proof reading task, during which the participants moved their eyes back and forth between two targets positioned in the center of two checkerboards. We recorded EEG, EOG, heart rate, and facial muscle potential/performance during the saccade tests. Blood pressure was measured and subjective fatigue and stress scores were collected before and after the proof reading task. We averaged EEG starting at saccade offset in order to analyze eye fixation related potentials (EFRP). Our results suggested that the proof reading task induced visual fatigue. An analysis of EFRP and other psychophysiological data revealed significant differences between the LUT and placebo conditions. These results suggest that supplementation with LUT could help to reduce symptoms of visual fatigue.  相似文献

A new reactor concept of innovative water reactor for flexible fuel cycle (FLWR) is under development at Japan Atomic Energy Agency in cooperation with Japanese reactor suppliers. A design of 1,356 MWe high conversion boiling water reactor-type FLWR core, which has an instantaneous conversion ratio of 1.04, negative void coefficient, high burnup of 65 GWd/t, and 15-month operational cycle length, has been constructed. So far, studies on thermal-hydraulic characteristics have been performed for tight lattice core. Evaluation methods for the critical power and the pressure drop under both the steady and the transient states have been established, and a modified TRAC-BF1 code has been developed for the thermal-hydraulic design of the FLWR. In this paper, the thermal feasibility of the designed 1356MWe FLWR core is analyzed by using the modified TRAC-BF1 code. The analysis is first carried out for the current core design. It is confirmed that no boiling transition (BT) occurs under the steady state. However, the minimum critical power ratio (MCPR) is only about 1.08, and the BT is confirmed occurring under the postulated abnormal transient processes. Therefore, concretizations of the conditions that ensure the thermal feasibility of a natural circulation-type FLWR and a forced circulation-type FLWR are performed. As for the results, for a forced circulation-type FLWR, the operation-limited MCPR (OLMCPR) is 1.32, and the necessary minimum core coolant flow rate is 640 kg/(m2s). For a natural circulation-type FLWR, the OLMCPR is 1.19, and the necessary minimum core coolant flow rate is 560 kg/(m2s).  相似文献

Resorcinol-formaldehyde hydrogels were synthesized by sol-gel polycondensation of resorcinol with formaldehyde in a slightly basic aqueous solution. RF cryogels, RF xerogels, and RF xerogels (MW gels) were respectively prepared from RF hydrogels by freeze drying, hot air drying, and microwave drying. Carbon cryogels, carbon xerogels and carbon MW gels were subsequently obtained by pyrolyzing RF drygels in an inert atmosphere. Freeze drying and microwave drying were effective to prepare mesoporous RF drygels and carbon gels. RF cryogels and carbon cryogels showed high mesoporosity over wide ranges of the molar ratio of resorcinol to catalyst (R/C) and the ratio of resorcinol to water (R/W) used in sol-gel polycondensation. Although RF xerogels had a few mesopores, carbon xerogels had no mesopores. RF MW gels and carbon MW gels showed mesoporosity if appropriate values of R/C and R/W were selected.  相似文献

The catalytic activity in the polymerization of styrene has been examined using commercially available simple rare earth metal compounds such as Sm(OiPr)3, Sm(acac)3, Sm(OCOMe)3, SmI2(THF)2 or SmCl3 coupled with Et3Al or methylaluminoxane (MAO). Among these compounds, the Sm(OiPr)3/AlEt3 system shows the highest catalytic activity, especially in the presence of a minor amount of toluene at 60 °C. The random copolymerization of styrene with methyl methacrylate suggests that the present polymerization proceeds with a radical polymerization mechanism. (C5Me5)SmCl3Na(THF) and (C5Me5)SmCl3Li(THF) systems exhibit relatively low catalytic activity, even in the presence of AlEt3. © 2001 Society of Chemical Industry  相似文献
