BAF-L Modulates Histone-to-Protamine Transition during Spermiogenesis

Maturing male germ cells undergo a unique developmental process in spermiogenesis that replaces nucleosomal histones with protamines, the process of which is critical for testicular development and male fertility. The progress of this exchange is regulated by complex mechanisms that are not well understood. Now, with mouse genetic models, we show that barrier-to-autointegration factor-like protein (BAF-L) plays an important role in spermiogenesis and spermatozoal function. BAF-L is a male germ cell marker, whose expression is highly associated with the maturation of male germ cells. The genetic deletion of BAF-L in mice impairs the progress of spermiogenesis and thus male fertility. This effect on male fertility is a consequence of the disturbed homeostasis of histones and protamines in maturing male germ cells, in which the interactions between BAF-L and histones/protamines are implicated. Finally, we show that reduced testicular expression of BAF-L represents a risk factor of human male infertility.

Molecular Cytogenetic Characterization of Novel 1RS.1BL Translocation and Complex Chromosome Translocation Lines with Stripe Rust Resistance

Rye is the most important source for the genetic improvement of wheat. In this study, two stable wheat-rye primary 1RS.1BL translocation lines, RT855-13 and RT855-14, were selected and identified by acid polyacrylamide gel electrophoresis (A-PAGE), co-dominant PCR, and multi-color fluorescence in situ hybridization (MC-FISH) from the progeny of the crossing of the wheat cultivar Mianyang11 and a Chinese rye Weining. When more than two independent, simple reciprocal translocations are involved in a carrier, they are defined as complex chromosome translocations (CCT). The MC-FISH results also indicated that CCT occurred in RT855-13; namely that, besides 1RS.1BL translocation chromosomes, there are other two pairs of balanced reciprocal translocations. It was demonstrated that the interchange between a distal segment of 4B and long arm of 3D occurred in the RT855-13. The novel translocation chromosomes in wheat were recorded as 3DS.4BS^DS and 3DL-4BS^PS.4BL. Reports about CCT as a genetic resource in plant breeding programs are scarce. Both lines expressed high resistance to Puccinia striiformis f. sp. tritici, which are prevalent in China and are virulent on Yr9, and the CCT line RT855-13 retained better resistance as adult plants compared with RT855-14 in the field. Both lines, especially the CCT line RT855-13, exhibited better agronomic traits than their wheat parent, Mianyang11, indicating that both translocation lines could potentially be used for wheat improvement. The results also indicated that the position effects of CCT can lead to beneficial variations in agronomic and resistant traits, making them a valuable genetic resource to wheat breeding programs.     

A technical method to improve NOx/NH3 mixing ratio in SCR system and its engineering applications

A technical method for diagnosing the distribution of NO_x flux within the cross-section area in front of ammonia injection grid (AIG) was proposed for guiding the valve-tuning of AIG branch-pipes, in order to optimize the NO_x/NH₃ mixing ratio in the selective catalytic reduction (SCR) system of power plant. The weight coefficient of each branch-pipe in AIG system can be quantitatively determined with regard to the distribution of NO_x flux in the corresponding sub-zone of the cross-section area. The control strategy of the valves for different AIG branch-pipes can be achieved for guiding the NH₃ injection and improving the NO_x/NH₃ mixing ratio within the whole cross-section area in front of AIG. The technology has been applied on one side of the SCR system flue-gas tunnels (normally two tunnels for the SCR system called as A-side and B-side) of a 660 MW plant for more than one year. The ammonia consumption rate of the SCR system was reduced about 12.62% and the uniformity of outlet NO_x distribution was estimated to be greatly improved by about 79.01% with regard to the standard deviation. The rising rate of the flue gas resistance of the air preheater was slown down by 39.18% compared to that of the other flue-gas tunnel of SCR system. This implied that the formation of the sticky ammonium bisulfate (ABS) on air preheater was significantly inhibited through the application of this technology.
