CONCEPTS AND ASSUMPTIONS of self-in-relation theory of women's development, as proposed by feminist scholars at the Stone Center in Wellesley, Massachusetts, are applied in this article. This theory was used as a framework to guide staff development and mentorship roles of advanced practice nurses. How the theory served to direct these roles and assisted in development of a professional practice model is discussed. Exemplars from the authors' practices and professional practice outcomes after 2 years of theory implementation are presented.  相似文献

DNA vaccination is an effective means of eliciting strong antibody responses to a number of viral antigens. However, DNA immunization alone has not generated persistent, high-titer antibody and neutralizing antibody responses to human immunodeficiency virus type 1 (HIV-1) envelope glycoprotein (Env). We have previously reported that DNA-primed anti-Env antibody responses can be augmented by boosting with Env-expressing recombinant vaccinia viruses. We report here that recombinant Env protein provides a more effective boost of DNA-initiated antibody responses. In rabbits primed with Env-expressing plasmids, protein boosting increased titer, persistence, neutralizing activity, and avidity of anti-Env responses. While titers increased rapidly after boosting, avidity and neutralizing activity matured more slowly over a 6-month period following protein boosting. DNA priming and protein immunization with HIV-1 HXB-2 Env elicited neutralizing antibody for T cell line-adapted, but not primary isolate, viruses. The most effective neutralizing antibody responses were observed after priming with plasmids which expressed noninfectious virus-like particles. In contrast to immunizations with HIV-1 Env, DNA immunizations with the influenza virus hemagglutinin glycoprotein did not require a protein boost to achieve high-titer antibody with good avidity and persistence.  相似文献

The recent emergence of the discrete fractional Fourier transform has spurred research activity aiming at generating Hermite–Gaussian-like (HGL) orthonormal eigenvectors of the discrete Fourier transform (DFT) matrix F. By exploiting the unitarity of matrix F – resulting in the orthogonality of its eigenspaces pertaining to the distinct eigenvalues – the problem decouples into finding orthonormal eigenvectors for each eigenspace separately. A Direct Sequential Evaluation by constrained Optimization Algorithm (DSEOA) is contributed for the generation of optimal orthonormal eigenvectors for each eigenspace separately. This technique is direct in the sense that it does not require the generation of initial orthonormal eigenvectors as a prerequisite for obtaining the final optimal ones. The resulting eigenvectors are optimal in the sense of being as close as possible to samples of the Hermite–Gaussian functions. The technique is found to be numerically robust because total pivoting is allowed in performing the QR matrix decomposition step. The DSEOA is proved to be theoretically equivalent to each of the Gram–Schmidt algorithm (GSA) and the sequential orthogonal Procrustes algorithm (SOPA). However the three techniques are algorithmically quite distinct. An extensive comparative simulation study shows that the DSEOA is by far the most numerically robust technique among all sequential algorithms thus paying off for its relatively long computation time.  相似文献

Formation of stable actin filaments, critically important for actin functions, is determined by the ionic strength of the solution. However, not much is known about the elements of the actin fold involved in ionic-strength-dependent filament stabilization. In this work, F-actin was destabilized by Cu2+ binding to Cys374, and the effects of solvent conditions on the dynamic properties of F-actin were correlated with the involvement of Segment 227-235 in filament stabilization. The results of our work show that the presence of Mg2+ at the high-affinity cation binding site of Cu-modified actin polymerized with MgCl2 strongly enhances the rate of filament subunit exchange and promotes the filament instability. In the presence of 0.1 M KCl, the filament subunit exchange was 2–3-fold lower than that in the MgCl2-polymerized F-actin. This effect correlates with the reduced accessibility of the D-loop and Segment 227-235 on opposite filament strands, consistent with an ionic-strength-dependent conformational change that modulates involvement of Segment 227-235 in stabilization of the intermonomer interface. KCl may restrict the mobility of the α-helix encompassing part of Segment 227-235 and/or be bound to Asp236 at the boundary of Segment 227-235. These results provide experimental evidence for the involvement of Segment 227-235 in salt-induced stabilization of contacts within the actin filament and suggest that they can be weakened by mutations characteristic of actin-associated myopathies.  相似文献

A polymer-supported Pd catalyst was investigated in hydrogenation of citral (3,7-dimethyl-2,6-octadienal), which is a stereoisomer with an isolated and a conjugated double bond as well as a carbonyl group. The catalyst was a fibrous polymer-supported catalyst modified with functional groups and immobilized metals. The enantiomeric composition of seven monoterpene hydrocarbons in headspace volatiles of spruce sawdust and seedlings (Picea abies), pine seedlings (Pinus sylvestris), and branches of juniper (Juniperus communis) was determined by gas chromatographic separation on a -Cyclodextrin column. For the six monoterpenes, -pinene, camphene, -pinene, sabinene, limonene, and -phellandrene, both enantiomers were present, whereas for 3-carene only the (+)-configuration was found. The amount of each enantiomer varied considerably both in relation to total amount of all of them, and for the six pairs also in relation to the opposite enantiomer. One olfactory receptor neuron in the pine weevil (Hylobius abietis) showed a strong response to -pinene when stimulated with all four headspace materials via a GC equipped with a DB-WAX column. The same neuron was subsequently tested with repeated stimulations via the GC effluent containing the (+)- or (–)-enantiomer. A marked better response to (+)- than to (–)--pinene was elicited. Another olfactory receptor neuron that responded strongly to limonene when stimulated with the spruce volatiles was tested for enantiomers of limonene. This neuron responded more strongly to (–)- than to (+)-limonene, when stimulated alternately with each of the limonene enantiomers. Discrimination between enantiomers by plant olfactory receptor neurons suggests that the enantiomeric ratios of volatile compounds may be important in host location by the pine weevil.  相似文献

The influence of silica fume on concrete properties represents an important technical research. In general, silica fume tends to improve both mechanical characteristics and durability of concrete. Thus the electrical properties of concrete containing silica fume can be studied to clarify its physical performance during hydration. The electrical conductivity of neat cement, mortar and concrete pastes was measured during setting and hardening. The ordinary Portland cement was partially replaced by different amounts of silica fume by weight. The changes in the electrical conductivity were reported during setting and hardening after gauging with water. The results of this study showed that the electrical conductivity can be used as an indication for the setting characteristics as well as the structural changes of the hardened pastes made with and without silica fume.  相似文献

Grinding tests were conducted in a laboratory steel ball mill, to investigate the grind-ability of some natural and artificial materials as “additions” for incorporation in Portland cement. Strength tests were performed on mixed cements composed of 71% Portland cement clinker, 25% addition and 4% gypsum. In one series of experiments, the components of the mixed cement were separately ground and in another one they were simultaneously ground. It was found from surface production curves and from compressive strength data that when materials which are harder to grind than clinker such as sand are interground with clinker, the resulting cement develops higher early strength than a corresponding cement in which the pozzolanic addition is softer than clinker. For materials which are softer to grind than clinker, separate grinding of the cement components is better than combined grinding, particularly for the development of early strength.  相似文献

Different posttreatment methods, such as heat treatment, mechanical processing, sealing, etc., are known to be capable to improve microstructure and exploitation properties of thermal spray coatings. In this work, a plasma electrolytic oxidation of aluminum coatings obtained by arc spraying on aluminum and carbon steel substrates is carried out. Microstructure and properties of oxidized layers formed on sprayed coating as well as on bulk material are investigated. Oxidation is performed in electrolyte containing KOH and liquid glass under different process parameters. It is shown that thick uniform oxidized layers can be formed on arc-sprayed aluminum coatings as well as on solid material. Distribution of alloying elements and phase composition of obtained layers are investigated. A significant improvement of wear resistance of treated layers in two types of abrasive wear conditions is observed.
