Coastal marsh habitats are impacted by many disturbances, including habitat destruction, pollution, and the introduction of invasive species. The common reed, Phragmites australis, has been particularly invasive in the mesohaline regions of the Chesapeake Bay, but few studies have investigated its role in trophic interactions with North American marsh consumers. The marsh periwinkle snail Littoraria irrorata is a common grazer in marshes and grazes on the native grass Spartina alterniflora. Whether this snail grazes on Phragmites has not been addressed. We found Spartina leaves to be tougher than those of Phragmites, but despite this, snails consumed significantly more Spartina than Phragmites. Subsequent experiments demonstrated that Phragmites is chemically deterrent to snails by an unknown, moderately polar, compound. Further studies are required to more fully understand the interactions between Phragmites, herbivores, and Spartina, and how they may impact marsh ecosystems.  相似文献

Herein we report the development of a highly active, magnetically retrievable and reusable biocatalyst using multilayer enzyme coupled-magnetic nanoparticles (MNPs) prepared by layer-by-layer assembly using two well-studied enzymes, horseradish peroxidase (HRP) and glucose oxidase (GOX), as a model enzyme system. We show that by combining the use of a biocompatible linker as well as biospecific immobilisation, the first layer enzyme in our HRP(1)-MNP system retains the native activity of the enzyme in solution, and the overall catalytic activity of the multilayer enzyme system, HRP(x)-MNP, increases linearly with the increasing number of enzyme layers. Furthermore, the HRP(x)-MNP system can be conveniently retrieved by using an external magnetic field and reused for 10 consecutive cycles without apparent reduction of catalytic activity. We also report the development of a novel coupled bienzyme, GOX/HRP(x)-MNP, system that can perform bi-enzymatic reactions to couple the colourless GOX-catalyzed reaction to the chromophoric HRP-catalyzed reaction via H(2)O(2) production. This model bienzyme-MNP system can be used for simple, rapid colorimetric quantification of micromolar glucose.  相似文献

The electron stimulated desorption (ESD) yield was measured at the internal surface of tubular samples made of aluminium alloy and aluminium coated stainless steel. The ESD yields were measured as a function of electron accumulated dose to a maximum of 3 × 1022 e/m2 at electron energy of 500 eV, then as a function of electron energy between 40 eV and 5 keV. The ESD yields as a function of electron energy were linear for all gasses except CH4 and CO2.  相似文献

The present study is a supplement to Epmeier and Kliger (2005), which investigated the effect of three different modification methods (acetylation, modification with methylated melamine resin and heat treatment in vegetable oil) on four material properties (density, modulus of elasticity (MOE), creep deflection and relative creep) and their inter-relations. In this study, another modification method (furfurylation) and four additional material parameters (“relative stiffness”, i.e. MOE/density, moisture content, change in moisture content and anti-creep efficiency) and their correlations are included. In addition, the creep curves were analysed to assess the development of bending creep performance. The properties were assessed by experimental work on 132 specimens of Scots pine sapwood with dimensions of 45×70×1100 mm3. It was found that modification leads to significant changes in material properties. Furfurylation increases density and reduces creep deflection, relative creep and moisture content. Modified timber tends to deflect and creep significantly less than untreated timber. However, the extent of reduction in relative creep appears not to be related to the extent of reduction in creep deflection. Initial deflection and/or modulus of elasticity are suitable for predicting the creep deflection of untreated and modified timber.  相似文献

The HMI1 gene encodes a DNA helicase that localizes to the mitochondria and is required for maintenance of the mitochondrial DNA (mtDNA) genome of Saccharomyces cerevisiae. Identified based on its homology with E. coli uvrD, the HMI1 gene product, Hmi1p, has been presumed to be involved in the replication of the 80 kb linear S. cerevisiae mtDNA genome. Here we report the purification of Hmi1p to apparent homogeneity and provide a characterization of the helicase reaction and the ATPase reaction with regard to NTP preference, divalent cation preference and the stimulatory effects of different nucleic acids on Hmi1p-catalysed ATPase activity. Genetic complementation assays indicate that mitochondrial localization of Hmi1p is essential for its role in mtDNA metabolism. The helicase activity, however, is not essential. Point mutants that lack ATPase/helicase activity partially complement a strain lacking Hmi1p. We suggest several possible roles for Hmi1p in mtDNA metabolism.  相似文献

Our study compared methane (CH4) emissions from lactating dairy cows measured using the sulfur hexafluoride (SF6) tracer and open-circuit respiration chamber techniques. The study was conducted using 16 lactating Holstein-Friesian cows. In each chamber, the cow was fitted with the SF6 tracer apparatus to measure total CH4 emissions, including emissions from the rectum. Fresh ryegrass pasture was harvested daily and fed ad libitum to each cow with a supplement of 5 kg of grain/d. The CH4 emissions measured using the SF6 tracer technique were similar to those using the chamber technique: 331 vs. 322 g of CH4/d per cow. The accuracy of the SF6 tracer technique was indicated by considering the ratio of the CH4 emission measured using the SF6 tracer to the emission measured using the chamber for each cow on each day. The calculated ratio of 102.3% (SE = 1.51) was not different from 100%. A higher variability within cow between days was found for the SF6 tracer technique [coefficient of variation (CV) = 6.1%] than for the chamber technique (CV = 4.3%). The variability among cows was substantially higher than within cows, and was higher for the SF6 technique (CV = 19.6%) than for the chamber technique (CV = 17.8%). Our CH4 emission data were compared with whole-animal chamber studies conducted in Canada and Ireland. In the Canadian study the SF6 technique did not measure CH4 emissions from the rectum and emissions were 8% lower than those measured using the chamber, indicating that emissions from the rectum may be greater than previously measured (1%). The relationship between CH4 emission and dry matter intake was examined for our data and for that reported in the Canadian study. There was a difference in the slopes of the regressions derived from our data and that from Canada; 17.1 vs. 20.8 g of CH4/kg of dry matter intake. A difference between the 2 locations was expected based on the difference in diet composition for these 2 studies. The SF6 tracer technique is reasonably accurate for inventory purposes and for evaluating the effects of mitigation strategies on CH4 emissions.  相似文献

During mammalian follicle development, a fluid-filled antrum develops in the avascular centre of the follicle. We investigated the hypothesis that follicular fluid contains osmotically-active molecules, sufficiently large so as to not freely escape the follicular fluid. Such molecules could generate an osmotic differential and thus recruit fluid from the surrounding vascularised stroma into the antrum. Follicular fluid was collected from bovine follicles classified histologically as healthy (n = 4 pools) or atretic (n = 4 pools). Dialysis of the follicular fluid at 300 kDa or 500 kDa resulted in a reduction in colloid osmotic pressure of 35% and 60%, respectively, in fluid from healthy follicles and 29% and 80% from atretic follicles. Digestion of follicular fluid with Streptomyces hyaluronidase, chondroitinase ABC or DNase 1 followed by dialysis resulted in reductions in osmotic pressure of 43%, 53% and 43% respectively for fluids from healthy follicles and 34%, 20% and 31% for atretic follicles. Digestion with collagenase I, proteinase K, heparanase 1 or keratanase had no significant effect on the osmotic pressure of follicular fluid of healthy follicles. Ion exchange and size exclusion, Western blotting and ELISA identified the proteoglycans versican and inter-alpha trypsin inhibitor and the glycosaminoglycan hyaluronan in follicular fluid. We conclude that these molecules or aggregates of them are of sufficient size to contribute to the osmotic potential of follicular fluid and thus recruit fluid into the follicular antrum. DNA may also contribute but it is probably not a component that is regulated for this role.  相似文献

This study aimed to determine whether reduced fetal ovary folliculogenesis in ewes undernourished during early/midpregnancy is associated with altered ovarian cell proliferation and/or the expression of apoptosis-regulating genes. Groups of ewes (n = 11-19) were fed either 100% (high; H) or 50% (low; L) of metabolisable energy requirements for live-weight maintenance during selected windows of gestation. All animals were killed at days 50, 65 or 110 of gestation. Between mating and slaughter, control animals were fed the H ration, while animals of other subgroups were fed the L ration from (a) mating to slaughter at 50, 65 or 110 days; (b) 0 to 30 days; (c) 31 to 50 or 65 days; or (d), in the day 110 slaughter group only, from 66 to 110 days. Bouin's-fixed fetal ovaries were examined for (a) Ki67 immunoexpression (proliferation) and (b) Bax and Mcl-1 (apoptosis-regulating genes) expression by in situ hybridisation (day 110) and immunohistochemistry (days 50, 65 and 110). At day 50, maternal nutrition had no effect on Ki67, predominant in germ cells, or Bax and Mcl-1, predominant in the oocytes. Restricted maternal food intake from 0 to 30 days significantly reduced staining for Ki67 in germ cells at day 65 (P < 0.05) but increased staining in granulosa cells at day 110 (P < 0.05). In animals fed the L ration for 110 days, primordial follicle Bax and Mcl-1 were significantly increased (Bax: P < 0.01; Mcl-1: P < 0.05). Granulosa cell Bax was also increased (P < 0.05). When the L ration was fed from 66 to 110 days, granulosa cell Bax (P < 0.05) and primordial follicle Mcl-1 (P < 0.01) were also significantly increased. In the fetal ovarian vasculature, animals underfed for 0-110 days had significantly elevated perivascular Mcl-1 (P < 0.001) and endothelial Bax expression (P < 0.05). Moreover, at day 110, endothelial Mcl-1 was increased by underfeeding from 0 to 30 days (P < 0.05). These data indicate that maternal undernutrition alters proliferation and the expression of apoptosis-regulating genes in the developing fetal ovary. The precise mechanism depends on the window of maternal food restriction.  相似文献

This paper builds a model of energy demand and supply for Ireland with a focus on evaluating, and providing insights for, energy efficiency policies. The demand-side comprises sectoral sub-models, with a detailed bottom–up approach used for the transport and residential sectors and a top–down approach used for the industry and services sectors. The supply side uses the linear programming optimisation features of the Open Source Energy Modelling System applied to electricity generation to calculate the least-cost solution. This paper presents the first national level model developed within the Long Range Energy Alternatives Planning software to combine detailed end-use analysis on the demand side with a cost-minimising optimisation approach for modelling the electricity generation sector. Through three scenarios over the period 2009–2020, the model examines the aggregate impact on energy demand of a selection of current and proposed energy efficiency policies. In 2020, energy demand in the energy efficiency scenario is 8.6 % lower than the reference scenario and 11.1 % lower in the energy efficiency?+?scenario.  相似文献
