This study was set up to establish the prevalence of Listeria monocytogenes in the tonsils of sows and fattening pigs from five Finnish slaughterhouses and to evaluate the genetic similarity of L. monocytogenes strains isolated from the tonsils. A total of 271 pig tonsils (132 tonsils from fattening pigs and 139 from sows) from five different slaughterhouses in various parts of Finland were studied from June 1999 to March 2000. Overall, 14 and 4% of pig tonsils harbored L. monocytogenes and Listeria innocua, respectively. The prevalence of L. monocytogenes in tonsils of fattening pigs (22%) was significantly higher than in sows (6%). The isolates (n = 38) recovered from tonsils showed a wide genetic diversity by means of 24 different pulsed-field gel electrophoresis (PFGE) types presented by the strains. Moreover, in numerical analyses of restriction patterns, no association was found between the clustering of strains and the slaughterhouses, and strains showing a similar PFGE type were recovered from pigs of different slaughterhouses. The high prevalence of L. monocytogenes showing various PFGE types in the tonsils of pigs could indicate a potential source of contamination of pluck sets, carcasses, and the slaughterhouse environment and of subsequent processing steps. 相似文献

Yersinia pseudotuberculosis with limited genetic diversity is a common finding in tonsils of fattening pigs 总被引:3,自引:0,他引:3
A total of 425 pig tonsils, including 210 tonsils from fattening pigs and 215 from sows, from seven different abattoirs in Finland were studied for the occurrence of Yersinia pseudotuberculosis from 1999 to 2000. The mean prevalence of Y. pseudotuberculosis in fattening pig tonsils was 4%, varying from 0 to 10% between slaughterhouses. Y. pseudotuberculosis was not recovered from sow tonsils. All 30 Y. pseudotuberculosis isolates from eight pig tonsils were recovered after cold enrichment. Seventeen isolates from seven tonsils were found after cold enrichment for 14 days, followed by alkali treatment. Y. pseudotuberculosis was not isolated after direct plating, overnight enrichment, or selective enrichment. All 30 isolates belonged to bioserotype 2/0:3 and carried the virF gene in the virulence plasmid. The isolates exhibited calcium dependence and Congo red absorption. The pyrazinamidase test gave variable results. All isolates were characterized with pulsed-field gel electrophoresis (PFGE). Using SpeI, NotI, and XbaI enzymes, seven, five, and two different PFGE patterns were obtained, respectively. A total of 11 genotypes, gI to gXI, identified by a combination of the various SpeI, NotI, and XbaI profiles, were detected. Three pigs were found to carry more than one genotype. Fermentation of rye dough is often accompanied with an increase in folate content. In this study, three sourdough yeasts, Candida milleri CBS 8195, Saccharomyces cerevisiae TS 146, and Torulaspora delbrueckii TS 207; a control, baker's yeast S. cerevisiae ALKO 743; and four Lactobacillus spp., L. acidophilus TSB 262, L. brevis TSB 307, L. plantarum TSB 304, and L. sanfranciscensis TSB 299 originally isolated from rye sourdough were examined for their abilities to produce or consume folates. The microorganisms were grown in yeast extract-peptone-d-glucose medium as well as in small-scale fermentations that modelled the sourdough fermentation step used in rye baking. Total folate contents were determined using Lactobacillus rhamnosus (ATCC 7469) as the growth indicator organism. The microorganisms studied did not excrete folates into the media in significant amounts. Yeasts increased the folate contents of sterilised rye flour-water mixtures from 6.5 microg/100 g to between 15 and 23 microg/100 g after 19-h fermentation, whereas lactic acid bacteria decreased it to between 2.9 and 4.2 microg/100 g. Strains of Lactobacillus bulgaricus, L. casei, L. curvatus, L. fermentum, L. helveticus, Pediococcus spp., and Streptococcus thermophilus that were also tested gave folate contents after fermentation that varied between 2 and 10.4 microg/100 g. Although the four Lactobacillus spp. from sourdough consumed folates their effect on folate contents in co-cultivations was minimal. It was concluded that the increase of folate content during fermentation was mainly due to folate synthesis by yeasts. Fermentation of non-sterilised flour-water mixtures as such resulted in three-fold increases in the folate contents. Lactic acid bacteria (LAB) in retail, modified-atmosphere-packaged (MAP), marinated broiler meat strips on sell-by-day were mainly identified as Leuconostoc gasicomitatum. A total of 32 packages, three to five packages of seven differently marinated broiler meat products, were studied at the end of the producer-defined shelf life (at 6 degrees C, 7-9 days depending on the manufacturer). Prior to the microbiological analyses, appearance and smell of the product was checked and pH measured. Bacteria were cultured on MRS and Tomato Juice Agar (TJA), Rogosa SL agar (SLA), Plate Count Agar (PCA) and Streptomycin Thallium Acetate Agar (STAA) for the enumeration of LAB, lactobacilli, total bacterial count and Brochothrix thermosphacta, respectively. The average CFU/g of the 32 packages was 2.3 x 10(8) on PCA. The highest bacterial average, 3.1 x 10(8), was recovered on TJA, the corresponding CFU/g averages on MRS and SLA being 2.3 x 10(8) and 1.3 x 10(8), respectively. Despite the high LAB numbers detected, radical spoilage changes such as unpleasant odor, slime production and formation of gas were not seen. B. thermosphacta did not form a significant part of the bacterial population since none of the levels exceeded the spoilage threshold level of 10(5) CFU/g reported in previous studies for this organism. In order to characterize the dominating LAB population, as many as 85, 85 and 88 colonies from MRS, TJA and SLA, respectively, were randomly picked and cultured pure. LAB were identified to species level using a 16 and 23S rDNA HindIiI RFLP (ribotyping) database. Fifty-six of the 170 isolates picked from the non-selective LAB media (MRS and TJA) were identified as L. gasicomitatum, followed by Carnobacterium divergens (41 isolates), Lactobacillus sakei and Lactobacillus curvatus subsp. melibiosus (31 isolates) and L. curvatus subsp. curvatus (20 isolates) species. SLA proved not to be completely selective for lactobacilli because the growth of Leuconostoc spp. was not inhibited, Carnobacterium spp. were the only species not detected on SLA. 相似文献

The isoflavonoids, equol, formononetin, daidzein, genistein, biochanin A, and O-demethylangolensin (O-DMA), were analyzed from commercial cartons of skimmed Finnish milk by HPLC-diode array detector (DAD)-FL. Oxide-supported transition-metal clusters and nanoparticles have attracted significant attention owing to their important role as components of model catalysts, sensors, solar cells and magnetic recording devices. For small clusters, functionality and structure are closely interrelated. However, knowledge of the structure of the bare cluster is insufficient as the interaction with the chemical environment might cause drastic structural changes. Here we show by ab initio simulations based on the density functional theory that the reaction with molecular oxygen transforms small, non-crystalline, magnesia-supported Pd-clusters to crystalline Pd(x)O(y) nano-oxide clusters that are in epitaxy with the underlying support. Restructuring of the Pd backbone is controlled by the electrostatic interaction with magnesia leading to a strong reduction of the O2 dissociation barrier. The supported Pd(x)O(y) clusters are likely to serve as Mars-van Krevelen oxygen reservoirs in catalytic oxidation reactions as observed for PdO overlayers and demonstrated here for the oxidation of CO molecules. 相似文献

The prevalence of Clostridium botulinum types A, B, E and F in river lampreys caught in Finnish rivers was determined for the first time using a quantitative PCR-MPN (most probable number) analysis. One of 67 raw whole lampreys (1.5%) was positive for the botulinum neurotoxin type E gene, with the estimated C. botulinum count being 100spores/kg. Two type E strains were isolated from the positive sample and confirmed as different genotypes by pulsed-field gel electrophoresis. Although the current procedure of bringing the charcoal-broiled lampreys to market has been without any further packaging or extended storage, interest towards increasing the shelf life of the product by vacuum-packaging is increasing. Our results demonstrate that C. botulinum type E may constitute a safety hazard in processed lampreys from the Baltic Sea area if packaging and extended shelf lives are to be used. To control the potential risk, a storage temperature of 3 degrees C or below should be recommended for these products. 相似文献
