The first part of this publication dealt with the effect of synthesis conditions on short and long-chain branching of low density polyethylene; this second part reports on the effect on the molar mass, the molar mass distribution and the polydispersity that can be determined from these parameters. The average molar mass was measured with a membrane osmometer and the molar mass distribution by gel permeation chromatography, using the same samples as those used for the branching analysis. These samples were produced in continuously operating stirred autoclaves under widely differing polymerization conditions. The number-average molar mass falls as the polymerization temperature and the initiator concentration rises and the pressure decreases. When the polymerization temperature is high, the molar mass distribution is narrower and less asymmetrical. When the polymerization temperature is low, there are broader, more asymmetrical distributions with a larger higher molar mass fraction. The polydispersity calculated from the molar mass distribution falls accordingly as the polymerization temperature rises. The polyethylene samples produced in a squat stirred autoclave have a higher polydispersity than those produced in a narrow autoclave. These findings have been confirmed on commercial products produced in various types of reactors. 相似文献

The ability of Salmonella to form complex surface-associated communities, called biofilms, contributes to its resistance and persistence in both host and non-host environments and is especially important in food processing environments. In this review, the different types of abiotic (plastic, glass, cement, rubber, and stainless steel) and biotic surfaces (plant surfaces, epithelial cells, and gallstones) on which Salmonella biofilms have been described are discussed, as well as a number of commonly used laboratory setups to study Salmonella biofilm formation (rdar morphotype, pellicle formation, and biofilms on polystyrene pegs). Furthermore, the structural components important during Salmonella biofilm formation are described (curli and other fimbriae, BapA, flagella, cellulose, colanic acid, anionic O-antigen capsule and fatty acids), with special attention to the structural variations of biofilms grown on different surfaces and under different conditions. Indeed, biofilm formation is strongly influenced by different environmental signals, via a complex regulatory network. An extensive overview is given on the current understanding of this genetic network and the interactions between its different components (CsgD, RpoS, Crl, OmpR, IHF, H-NS, CpxR, MlrA, c-di-GMP, BarA/SirA, Csr, PhoPQ, RstA, Rcs, metabolic processes and quorum sensing). To further illustrate that biofilm formation is a mechanism of Salmonella to adapt to different environments, the resistance of Salmonella biofilms against different stress factors including desiccation stress, disinfectants (e.g. hypochlorite, glutaraldehyde, cationic tensides and triclosan) and antibiotics (e.g. ciprofloxacin) is described. Finally, a number of Salmonella biofilm inhibitors, identified through bottom-up- and top-down-approaches, are discussed, such as surfactin, glucose, halogenated furanones, 4(5)-aryl 2-aminoimidazoles, furocoumarins and salicylates. Also the potential of combination therapy (e.g. combinations of triclosan and quaternary ammonium salts or halogenated furanones and antibiotics/disinfectants) and nano- and micro-emulsions to inhibit Salmonella biofilm formation is discussed. Insight into the pathogen's complex biofilm process will eventually lead to further unraveling of its intricacies and more efficient strategies to combat Salmonella biofilms. 相似文献

The method of hierarchical imprinting was used for the first time in order to synthesize polymers with a high affinity for
nisin. The concept of this approach is demonstrated by filling the pores of silica particles, which contain immobilized peptidic
templates, with a mixture of monomers/initiator, followed by polymerization and subsequent dissolution of the silica template.
This method leaves imprinted polymers with binding sites located at the surface that are capable of recognizing larger molecules
with the same immobilized epitope. The results of the high-performance liquid chromatography analyses illustrate that the
highest retention factors and imprinting factors of nisin on nisin C-terminal analogously imprinted polymers could be obtained
with acetonitrile/water mixtures containing more than 80% water. Furthermore, nisin was strongly retained on polymers that
were imprinted with a longer amino acid sequence because of better sterical accessibility of the binding sites for the C terminus
of nisin. The retention of nisin (k = 21.65) under aqueous elution conditions results from selective ionic interactions and hydrophobic interactions. Furthermore,
the polymers exhibited selectivity for the template Lys-Ala and the structurally related dipeptide Ala-Lys. The results of
the present work show that these hierarchically imprinted polymers could be used for the chromatographic separation of the

Consumer wishes have to be translated into product characteristics to implement consumer-oriented product development. Before this step can be made, insight in food-related behavior and perception of consumers is necessary to make the right, useful, and successful translation. Food choice behavior and consumers' perception are studied in many disciplines. Models of food behavior and preferences therefore were studied from a multidisciplinary perspective. Nearly all models structure the determinants related to the person, the food, and the environment. Consequently, the overview of models was used as a basis to structure the variables influencing food perception into a model for consumer-oriented product development. To this new model, referred to as food perception model, other variables like time and place as part of consumption moment were added. These are important variables influencing consumers' perception, and therefore of increasing importance to consumer-oriented product development nowadays. More than 1900 sediment-water partitioning coefficients were measured for 58 polychlorinated biphenyl (PCB) congeners in 53 historically contaminated sediments collected from 10 urban and rural waterways in the United States and Canada. Freely dissolved porewater concentrations were determined using passive sampling with polyoxymethylene. Measured total organic carbon (TOC)/water partitioning coefficients, K(TOC), ranged from one to nearly three orders-of-magnitude higher than typical literature values based on spiking experiments and model predictions. Although total PCB concentrations ranged from 0.08 to 194 mg/kg, the more highly contaminated sediments showed only slightly lower K(TOC) values than less-contaminated sediments. No correlation was observed between log K(TOC) values and sediment TOC, black carbon (BC), or BC/TOC fractions (r(2) typically <0.1). Utilizing a two-carbon model incorporating anthropogenic BC did not improve predictions over a one-carbon TOC model. A comparison of models recently validated for field data showed that a coal-tar poly parameter linear-free energy relationship (PP-LFER) and a Raoult's Law model were successful at predicting average log K(TOC) values, without the need for any calibration or fitting (within a factor of 10 more than 90% of the time, and within a factor of 30 more than 99% of the time). Predictions were further improved by the introduction of a Weathering Factor (WF) that accounts for the relative depletion of lower molecular weight congeners due to weathering. Highly weathered sediments (with a WF near 1) tended to follow the coal-tar PP-LFER and Raoult's Law model the closest. Less-weathered sediments (with WF ? 1) sorbed less than predicted by these models. Noncalibrated WF inclusive coal-tar PP-LFER and Raoult's Law models performed as well or better than a quantitative-structure activity relationship (QSAR) model calibrated specifically to the data. The present study aimed to evaluate the effect of single-layer centrifugation (SLC) through a species-specific colloid (Androcoll-B; patent pending, J. M. Morrell) on bull sperm quality. Computer-assisted sperm analysis of motility and flow cytometric analysis of sperm viability (SYBR-14/propidium iodide staining), chromatin integrity (acridine orange staining), reactive oxygen species production [Hoechst 33258-hydroethidine-2′,7′-dichlorodihydrofluorescein diacetate (HO-HE-DCFDA) staining], mitochondrial membrane potential (staining with JC-1 probe), and protein tyrosine phosphorylation (specific antibody staining) were performed on unselected and SLC-selected sperm samples. Single-layer centrifugation of bull spermatozoa resulted in the selection of a sperm population that had high mitochondrial membrane potential, a higher content of phosphorylated protein, and more reactive oxygen species than control samples. Sperm chromatin damage was lower in the SLC samples although sperm viability and motility did not differ between SLC samples and controls. These observations suggest that SLC of bull semen in a soybean-containing extender improved some, but not all, parameters of sperm quality. 相似文献

Reliable, sensitive, quantitative, and mobile rapid screening methods for pathogenic organisms are not yet readily available, but would provide a great benefit to humanitarian intervention units in disaster situations. We compared three different methods (immunofluorescent microscopy, IFM; flow cytometry, FCM; polymerase chain reaction, PCR) for the rapid and quantitative detection of Giardia lamblia and Cryptosporidium parvum (oo)cysts in a field campaign. For this we deployed our mobile instrumentation and sampled canal water and vegetables during a 2 week field study in Thailand. For purification and concentrations of (oo)cysts, we used filtration and immunomagnetic separation. We were able to detect considerably high oo(cysts) concentrations (ranges: 15-855 and 0-240 oo(cysts)/liter for Giardia and Cryptosporidium, respectively) in 85 to 300 min, with FCM being fastest, followed by PCR, and IFM being slowest due to the long analysis time per sample. FCM and IFM performed consistently well, whereas PCR reactions often failed. The recovery, established by FCM, was around 30% for Giardia and 13% for Cryptosporidium (oo)cysts. It was possible to track (oo)cysts from the wastewater further downstream to irrigation waters and confirm contamination of salads and water vegetables. We believe that rapid detection, in particular FCM-based methods, can substantially help in disaster management and outbreak prevention. 相似文献

Excitation-emission matrix (EEM) fluorescence was combined with parallel factor analysis (PARAFAC) to model base-extracted particulate (POM) and dissolved (DOM) organic matter quality in the Neuse River Estuary (NRE), North Carolina, before and after passage of Hurricane Irene in August 2011. Principle components analysis was used to determine that four of the PARAFAC components (C1-C3 and C6) were terrestrial sources to the NRE. One component (C4), prevalent in DOM of nutrient-impacted streams and estuaries and produced in phytoplankton cultures, was enriched in the POM and in surface sediment pore water DOM. One component (C5) was related to recent autochthonous production. Photoexposure of unfiltered Neuse River water caused an increase in slope ratio values (S(R)) which corresponded to an increase in the ratio C2:C3 for DOM, and the production of C4 fluorescence in both POM and DOM. Changes to the relative abundance of C4 in POM and DOM indicated that advection of pore water DOM from surface sediments into overlying waters could increase the autochthonous quality of DOM in shallow microtidal estuaries. Modeling POM and DOM simultaneously with PARAFAC is an informative technique that is applicable to assessments of estuarine water quality. 相似文献

Summary In addition to the mono- and dipyrrolidinohexosereductones described recently (Z Lebensm Unters Forsch (1984) 178:356) two further bittertasting compounds were isolated from heated mixtures of proline and sucrose (molar ratio 3: 1, 190 °C, 30 min) and also synthesized, namely 2,3-bis-(1-pyrrolidinyl)2-cyclopenten-l-one and 2,3-bis(1-pyrrolidinyl)-5methyliden-2-cyclopenten-l-one. The recognition thresholds (mmol/1) are 0.15–0.25 and 0.06–0.12 respectively. Both compounds have a burning bitter taste character. From a maltose-proline reaction mixture 2--glucosyl-5-hydroxy-5-methyl-4-piperidinyl-2-cyclopenten-1-one was isolated. When the dihydroxypyranone VI was heated with proline, 2,5-dihydroxy-5-methyl-4-prohnyl-2-cyclopenten-1-one and the cyclopent(b)azepinones XI and XII were among the compounds formed. These compounds also have a bitter taste character.Zusammenfassung Zusätzlich zu den kürzlich beschriebenen Mono- and Dipyrrolidinohexosereduktionen (Z Lebensm Unters Forsch (1984) 178:356) wurden zwei weitere bittere Verbindungen aus erhitzten Mischungen von Prolin and Saccharose (Molverhältnis 3:1, 190 °C, 30 min) isoliert and synthetisiert.
