Ca2+ was precipitated with potassium antimonate in vitellogenic follicles of the fruit fly Drosophila melanogaster and the distribution of the precipitates formed was studied by electron microscopy. The microvilli of the oolemma in mid- and late vitellogenic follicles were lined with precipitates. The chemical composition of the precipitates was analysed by electron spectroscopic imaging (ESI). The images produced by inelastically scattered electrons at specific ionization edges were compared, and the nonspecific background signals were subtracted by an image processing system. The presence of Ca2+, antimony and oxygen in the precipitates formed could be demonstrated. The elemental composition of the precipitates and of yolk spheres was also analysed by electron energy-loss spectroscopy (EELS). With respect to the precipitates, signals at the calcium L2,3-edge, the oxygen K-edge and the antimony M4,5-edge were recorded without deconvolution and background subtraction. The yolk spheres, which were free of precipitates, gave the characteristic signal of the nitrogen K-edge. The applied techniques combine good ultrastructural resolution with the possibility of analysing the elemental composition of histochemical reaction products and cellular structures.  相似文献

The extracellular sheath material and some intracellular cell components of cyanobacteria and phosphate-accumulating sewage bacteria were analysed by electron spectroscopic imaging (ESI) and electron energy-loss spectroscopy (EELS). The specimens were embedded in water-soluble Nanoplast resin without any previous fixation and ultrathin sections were examined in a Zeiss CEM 902 microscope. A high sulphur content was detected in the inner sheath of the cyanobacterium Gloeothece. The elemental composition of some cell components and inclusion bodies, such as carboxysomes and cyanophycin, was determined by ESI and EELS. In addition, the phosphate content in specific granules of phosphate-accumulating sewage bacteria was estimated by EELS and nuclear magnetic resonance spectroscopy.  相似文献

Indoor and outdoor BTX levels in German cities   总被引:4,自引:0,他引:4  
On the basis of the ongoing study INGA (INdoor exposure and Genetics in Asthma), Germany's most detailed and standardized epidemiological study on indoor exposure to both allergens in house dust and volatile compounds in the air of the home environment has been performed. The purpose of this paper is to describe the spatial and seasonal variability of indoor and outdoor BTX (Benzene, toluene, ethyl benzene, ortho-xylene, meta- and para-xylene) concentrations for the study period from June 1995 to November 1996. Within this framework, air concentrations of volatile organic compounds (BTX) were measured in 204 households in Erfurt (Eastern Germany) and 201 households in Hamburg (Western Germany). BTX sampling was conducted over one week using OVM 3500 passive diffusion sampling devices in the indoor (living room and bedroom) and outdoor environment (outside the window of the living room). Indoor and outdoor median BTX concentrations in Erfurt were slightly, but significantly higher than those in Hamburg. This gap was most pronounced in the levels of indoor toluene (37.3 microg/m3 for Erfurt and 20.5 microg/m3 for Hamburg, P < 0.0001). In both cities, winter indoor and outdoor concentrations for the five compounds exceeded the summer values. Outdoor concentrations of ethyl benzene and ortho-xylene were very low (50% < L.D.). In general, the indoor BTX air concentrations were significantly higher than the outdoor concentra- tions, the lowest I/O ratios were found in the case of benzene. Living room and bedroom values for the five compounds were highly correlated (Spearman coefficient 0.5-0.9). Despite the better insulation of the homes in West Germany, no indication for the expected higher indoor concentrations of BTX in the West could be found. The strong and yet undiscovered indoor source for toluene in East Germany might lead to a further increase in the indoor air load in those homes in the East, which undergo renovations which will lead to improved insulation.  相似文献

Ultra high purity Zn (m6N5) was obtained through triple vacuum distillation using an m4N5 Zn as input material. High-volatile impurities were removed from the zinc matrix by vacuum evaporation, while traces having lower volatility than the matrix remained in the residual material after finishing of the entire purification process. The time required for the removal of the main high-volatile impurity (Cd) was monitored using a connected gas source mass spectrometer. During distillation the Zn vapor of the distillate was condensed on the cap of a glassy carbon crucible which was water cooled by a movable copper block. The temperatures were optimized for the removal of the volatiles and for the distillation process using vapor pressure data. High resolution glow discharge mass spectrometry was applied for the analysis and purity evaluation of the distilled and input zinc. The analysis confirmed the reduction of the metallic impurities from 43 mg kg−1 to 0.5 mg kg−1 (m6N5) after three consecutive vacuum distillations. An increase in the grain size and a decrease in the micro-hardness were observed for the purified Zn material.  相似文献

We combine a THz time-domain spectroscopy system with a robotic arm. With this scheme, the THz emitter and receiver can be positioned perpendicular and at defined distance to the sample surface. Our system allows the acquisition of reflection THz tomographic images of samples with an arbitrarily shaped surface.  相似文献

Recently we reported the results of a screen of the Pathogen Box in which we identified 4-(2-amino-5-(4-(methylsulfonyl) phenyl) pyridin-3-yl)-2-methoxyphenol (MMV010576, 1 ) as our priority antitrypanosomal hit. This compound had previously been identified as a potent and selective antiplasmodial agent, where a focused optimization campaign, resulted in a medium-sized library of compounds, with favorable drug-like properties, one of which (MMV048, 2 , 5-(4-(methylsulfonyl)phenyl)-6′-(trifluoromethyl)-[3,3′-bipyridin]-2-amine) is currently undergoing clinical trials for malaria. Accordingly, we investigated this library, in order to elucidate structural activity relationship details of this class of compounds as inhibitors of Trypanosoma brucei. Our study has identified several structural features important for antitrypanosomal activity, which are distinct from those required for antiplasmodial activity. Results from this study can be exploited to develop potent antitrypanosomal agents.  相似文献

Aiming for non-destructive testing and security applications, we investigate transmission-mode imaging and spectroscopic sensing using terahertz (THz) pulses from a commercial optical parametric oscillator (OPO) in combination with THz detectors based on antenna-coupled field-effect transistors (TeraFETs). The Q-switched OPO generates quasi-continuous-wave THz pulses with a peak power of up to 1 W at a repetition rate between 12 and 90 Hz. The pulses are frequency-tunable between 0.7 and 2.6 THz with a typical linewidth of 50 GHz. We explore detection with fast GaN/AlGaN TeraFETs which hold the potential for multi-pixel and homodyne detection.  相似文献

We studied dampness and mold in homes in relation to climate, building characteristics and socio‐economic status (SES) across Europe, for 7127 homes in 22 centers. A subsample of 3118 homes was inspected. Multilevel analysis was applied, including age, gender, center, SES, climate, and building factors. Self‐reported water damage (10%), damp spots (21%), and mold (16%) in past year were similar as observed data (19% dampness and 14% mold). Ambient temperature was associated with self‐reported water damage (OR=1.63 per 10°C; 95% CI 1.02‐2.63), damp spots (OR=2.95; 95% CI 1.98‐4.39), and mold (OR=2.28; 95% CI 1.04‐4.67). Precipitation was associated with water damage (OR=1.12 per 100 mm; 95% CI 1.02‐1.23) and damp spots (OR=1.11; 95% CI 1.02‐1.20). Ambient relative air humidity was not associated with indoor dampness and mold. Older buildings had more dampness and mold (P<.001). Manual workers reported less water damage (OR=0.69; 95% CI 0.53‐0.89) but more mold (OR=1.27; 95% CI 1.03‐1.55) as compared to managerial/professional workers. There were correlations between reported and observed data at center level (Spearman rho 0.61 for dampness and 0.73 for mold). In conclusion, high ambient temperature and precipitation and high building age can be risk factors for dampness and mold in homes in Europe.  相似文献
