Fatty acid desaturations in the carboxy‐terminal segment from C1—C10 are catalyzed in many, but not in all cases, by desaturase enzymes which are fused to their electron donor cytochrome b5. Several of these enzymes (“front‐end desaturases”) from a wide variety of organisms have been cloned and functionally expressed for proof of regio‐, stereo‐ and chain length‐selectivity. In most cases the actual status of the substrate chain, whether coenzyme A thioester or component of a membrane lipid, is not known. The cytochrome b5 domain is located N‐terminally, internally or C‐terminally. Compared to the free cytochrome b5 , the fused domains show a significant reduction of acidic amino acid residues on the surface of the four helices enclosing the heme group. It is discussed how this may contribute to hydrophobic domain pairing required for interdomain electron transport. This is in contrast to the mode of interaction of free cytochrome b5 with its partners, which is governed by electrostatic charge pairing. A look at crystallized or computer‐simulated models involving fused or free cytochrome b5 helps to outline the problems encountered by optimizing the docking of partners and the exchange of electrons between domains of different degrees of mobility.  相似文献

A feeding experiment with brown layers was carried out to find effects of graded levels of rapeseed (without, 15% and 30%) in layer diets on the fatty acid composition of egg yolk and related functional properties of such modified eggs. A considerable increase of the content of essential n-3 fatty acids in egg yolk fat was noticed by including graded levels of rapeseed in layer diets. The destruction force of a gel prepared from whole egg was improved by the highest rapeseed level (30%) and emulsifying capacity by medium inclusion of rapeseed in layer diets (15%). The thermal stability of an emulsion prepared from whole egg was not influenced by rape feeding. Viscosity of mayonnaise prepared from egg yolk and whole egg yolk was decreased by increased rapeseed levels in the diets. In tendency, organoleptic evaluation of sponge cake prepared from whole egg revealed better results for the control group. The baking height of sponge cake was improved by rapeseed inclusion in layer diets. It may be assumed that all these changes caused by rapeseed feeding are closely related to the mentioned changes in the fatty acid profile.  相似文献

Membrane processes are able to separate liquid mixtures as well as mixtures of gases or vapours according to the sizes, solubilities, rates of diffusion or charges of the components. A separation is also possible using membranes in which the transport is coupled to a specific chemical reaction between the permeand and the membrane (facilitated or carrier mediated transport). This paper discusses aspects related to carrier‐mediated separation processes such as the transport mechanisms, the choice of appropriate carriers, the incorporation of carriers in the polymeric membrane material, the coating of porous support membranes with thin selective layers of the carrier‐polymer and, finally, the choice of appropriate parameters for the separation process. The separation of gas mixtures by carrier‐membranes appears very promising from a technical point of view. Experimental membrane samples often show excellent separation factors, but they have not met the permeability requirements for an economic use so far.  相似文献

The yeast Saccharomyces cerevisiae transforms branched‐chain and aromatic amino acids into higher alcohols in the Ehrlich pathway. During microbiological culturing and industrial fermentations, this yeast is confronted with amino acids modified by reducing sugars in the Maillard reaction (glycation). In order to gain some preliminary insight into the physiological “handling” of glycated amino acids by yeasts, individual Maillard reaction products (MRPs: fructosyllysine, carboxymethyllysine, pyrraline, formyline, maltosine, methylglyoxal‐derived hydroimidazolone) were administered to two strains of S. cerevisiae in a rich medium. Only formyline was converted into the corresponding α‐hydroxy acid, to a small extent (10 %). Dipeptide‐bound pyrraline and maltosine were removed from the medium with concomitant emergence of several metabolites. Pyrraline was mainly converted into the corresponding Ehrlich alcohol (20–60 %) and maltosine into the corresponding α‐hydroxy acid (40–60 %). Five specific metabolites of glycated amino acids were synthesized and characterized. We show for the first time that S. cerevisiae can use glycated amino acids as a nitrogen source and transform them into new metabolites, provided that the substances can be transported across the cell membrane.  相似文献

Fatigue failure is a concern when high‐strength, high‐toughness silicon nitride ceramics are used in mechanical components and the growth of natural flaws will determine the usable upper bound strength. In this study a fracture resistance curve (R‐curve) model is incorporated into an established method for deducing natural flaw growth rates from a combination of strength and fatigue life data for smooth specimens. Experimental data for a commercial silicon nitride, SL200, were examined. When compared with results deduced using a constant fracture toughness model, the new method gives more physically realistic growth rate results. Specifically, by incorporating the R‐curve the deduced fatigue threshold is equal to the reported intrinsic toughness for crack propagation of 2.2 MPa√m, whereas the constant fracture toughness model gives a physically unrealistic threshold value. Furthermore, much better agreement is achieved with the growth rates measured using macroscopic compact‐tension specimens. Overall, it is concluded that the R‐curve effect should not be ignored when deducing the fatigue crack growth rates of natural flaws in high‐toughness silicon nitride ceramics.  相似文献

The charge distributions of an improved opposed flow unipolar diffusion charger were measured using a tandem differential mobility analyzer (DMA) set up in a size range of approximately 20–400 nm. The charger is intended to be used in a portable aerosol sizer to measure particle size distributions. The determined charge distributions were represented by lognormal distributions, and a set of equations and coefficients was developed to calculate the charge distributions. These equations can be easily implemented in software for size distribution measurements. The agreement between the mathematically derived and measured charge distributions is very good, with regression coefficients R 2 > 0.96. The investigations showed that approximately 55% of 20-nm particles remain uncharged, while up to 25 elementary charges need to be considered for multiple charge correction of 400-nm particles. Comparison with the Fuchs theory delivered satisfying agreement with the measured average charge levels, but charge distributions cannot be described by the Fuchs theory, likely caused by the charger geometry.

On‐line liquid chromatography‐gas chromatography (LC‐GC) has been applied to the detection of vegetable oils in milk fat using β‐sitosterol as marker. The method involves transesterification of the fat, pre‐separation of the sterol fraction from other lipid constituents and on‐line transfer to the capillary GC system. The on‐line approach avoids time‐consuming sample preparation steps prior to GC analysis. The suitability of this analytical approach was tested with model mixtures of milk fat with cotton and rapeseed oil. The method allows the detection of adulterations at low levels. Simultaneous quantification of cholesterol in milk fat is also possible. Considering the rapid sample preparation, the described method can be applied for screening of large sample numbers.  相似文献

Raman spectra have been measured for pellets of five samples of high‐density polyethylene (HDPE), seven samples of low‐density polyethylene (LDPE), and six samples of linear low‐density polyethylene (LLDPE). The obtained Raman spectra have been compared to find out characteristic Raman bands of HDPE, LDPE, and LLDPE. Principal component analysis (PCA) was applied to the Raman spectra in the 1600–650 cm?1 region after multiplicative scatter correction (MSC) to discriminate the Raman spectra of the three different PE species. They are classified into three groups by a score plot of PCA factor 1 vs. 2. HDPE with high density and high crystallinity gives high scores on the factor 1 axis, while LDPE with low density and low crystallinity yields negative scores on the same axis. It seems that factor 1 reflects the density or crystallinity. A PC weight loadings plot for factor 1 shows six upward peaks corresponding to the bands arising from the crystalline parts or alltrans ? (CH2)n? groups and seven downward peaks ascribed to the bands of the amorphous or anisotropic regions and those arising from the short branches. Partial least‐squares (PLS‐1) regression was applied to the Raman spectra after MSC to propose calibration models that predict the density, crystallinity, and melting points of the polyethylenes. The correlation coefficient was calculated to be 0.9941, 0.9800, and 0.9709 for the density, crystallinity, and melting point, respectively, and their root‐mean‐square error of cross validation (RMSECV) was found to be 0.0015, 3.3707, and 2.3745, respectively. The loadings plot of factor 2 for the prediction of melting point is largely different from those for the prediction of density and crystallinity. © 2002 Wiley Periodicals, Inc. Summary: Non‐Newtonian fluid behavior has significant influence on quantities in chemical engineering like power input, mixing time, heat transfer etc. In the laminar flow region, the concept of effective viscosity by Metzner and Otto is well established. In the transition region between laminar and turbulent flow, the existing concepts use three and even more empirical parameters to determine the specific power input. Here, a unified and general but simple approach is introduced to calculate the power input for shear thinning fluids over the whole flow region using just one empirical parameter. The Metzner‐Otto relation is obtained as a limiting case for the laminar region. The empirical parameter of the new approach is related to the Metzner‐Otto constant. The concept is validated for eight different stirrer systems. Mixing time and maximum shear rate and heat transfer can also be calculated using this approach. The new concept presented should also be applicable for other apparatuses, e.g., static mixers.

A high-temperature gradient HPLC method has been developed for the analysis of polyethylene–polypropylene blends. For the first time it was possible to separate these polyolefin blends by a chromatographic technique which is operating at 140 °C. Blends of a commercial polypropylene and a medium molar mass linear polyethylene were separated using a mobile phase of ethylene glycol monobutylether (EGMBE) and 1,2,4-trichlorobenzene (TCB) and silica gel as the stationary phase. With the use of n-decanol as sample solvent, a precipitation–redissolution mechanism for polyethylene (PE) was established while polypropylene (PP) is eluted in size exclusion mode.  相似文献
