Improvements in defect detection and sizing capabilities for non-destructive inspection techniques have been required in order to ensure the reliable operation and life extension of nuclear power plants. For the volumetric inspection, the phased array UT technique has superior capabilities for beam steering and focusing to objective regions, and real-time B-scan imaging without mechanical scanning. In contrast to the conventional UT method, high-speed inspection is realized by the unique feature of the phased array technique. A 256-channel array system has developed for the inspection of weldment of BWR internal components such as core shrouds. The TOFD crack sizing technique also can be applied using this system. For the surface inspection, potential drop techniques and eddy current techniques have been improved, which combined the theoretical analysis. These techniques have the crack sizing capability for surface breaking cracks to which UT method is difficult to apply. 1-O-Octadecyl-2-O-methyl-sn-glycero-3-phosphocholine (ET-18-OMe) has been reported to possess definite antitumor activity in vivo. Twenty-two alkyl lysophospholipid analogs were chemically synthesized, and their antitumor activity against mouse experimental tumors (Sarcoma 180, MM46, P388) was examined. Among them, 1-O-octadecyl-2-O-acetoacetyl-rac-glycerol-3-phosphocholine was found to show antitumor activity similar to ET-18-OMe with less acute toxicity. Intravenous injection of the ET-18-OMe withsn-3 configuration retarded the subcutaneous growth of Sarcoma 180 cells effectively, while the growth inhibition by thesn-1 isomer was much less effective. This stereospecificity was similar to that observed in their activities as platelet-activating factor (PAF) agonists. The acetoacetyl compound, another PAF agonist, showed similar stereospecific antitumor action in vivo. These findings suggest that some alkyl lysophospholipids may activate host cells to a cytostatic stage against tumor cells in vivo through binding to a PAF receptor. Our preliminary results indicated that the responsible cells under these conditions might be primarily immature macrophages present in the bone marrow. No appreciable or even adverse stereospecificity was observed in the different sets of experiments where the activity of ET-18-OMe against MM46 tumor cells in vivo or the direct cytotoxicity against human promyelocytic leukemia HL-60 cells in vitro was examined. Under, some conditions, the antitumor activity of ET-18-OMe in vivo may be revealed through direct cytotoxicity and/or modulation of the host defense system by “nonspecific” mechanisms. The formation of fluorescent products in the reaction of methyl linoleate hydroperoxide with glycine in aqueous emulsions
correlated directly with the decrease in diene conjugation and the increase in thiobarbituric acid (TBA) reactive substances.
These correlations also were reflected in the course of the oxidation of methyl linoleate in aqueous emulsions with glycine
and indicated that glycine reacted with products of peroxide decomposition as opposed to intermediates of autoxidation in
hydroperoxide formation. Thin layer chromatography (TLC) and selective solvent extraction demonstrated that the products of
the reaction contained many substances with a fluorescent spectrum similar to those of lipofuscin pigments. When methyl esters
of polyunsaturated fatty acids or other polyunsaturated lipids underwent oxidation adsorbed on silica gel particles, products
with similar fluorescent spectral properties were formed illustrating that fluorescent substances were formed in a variety

A 308 nm monochromatic excimer light (MEL) is widely used to treat patients with vitiligo. However, dose optimization still needs to be clarified. This study aimed to obtain objective evidence regarding various doses of MEL irradiation, induced cell level changes in vitro, and skin level alterations in vivo. Cultured human keratinocytes were irradiated with MEL using various doses. After irradiation at low doses, stem cell factor, endothelin-1, and glycoprotein nonmetastatic melanoma protein B, factors that activate and protect melanocytes, were found to be significantly elevated in keratinocytes. After irradiation using medium and high doses, inflammatory cytokines were induced. The amount of ATP released and the level of inflammasome activation, which are known to be related to interleukin-1β activation, were also increased. The back skin of guinea pigs and mice were irradiated with MEL at varying doses. After irradiation, an increase of epidermal melanin and epidermal melanocytes was confirmed, using the minimal erythemal dose or less. In rhododendrol-induced leukoderma guinea pigs, a much lower dose of MEL irradiation was effective, when compared with the effective dose for control guinea pigs. Our results suggest that a lower irradiation dose of MEL might be sufficient and more suitable for repigmentation in vitiligo treatment. 相似文献

The prognosis of multiple myeloma (MM) has drastically improved owing to the development of new drugs, such as proteasome inhibitors and immunomodulatory drugs. Nevertheless, MM is an extremely challenging disease, and many patients are still refractory to the existing therapies, thus requiring new treatment alternatives. Venetoclax is a selective, orally bioavailable inhibitor of BCL-2 that shows efficacy in MM not only as a single agent but also in combination therapy, especially for MM patients with translocation t(11;14). However, many patients are refractory to this drug. Here, we treated the MM cell lines KMS12PE and KMS27 with a combination treatment of venetoclax targeting BCL-2 and daratumumab targeting CD38 to evaluate the synergistic cytotoxicity of these drugs in vitro. MM cell lines were co-cultured with natural killer (NK) cells at an effector:target ratio of 0.3:1 in the presence of serial concentrations of daratumumab and venetoclax, and the resulting apoptotic MM cells were detected by flow cytometry using annexin V. These results indicated that the antibody-dependent cell-mediated NK cytotoxicity was enhanced in KMS12PE and KMS27 cells harboring t(11;14) with a high BCL-2 expression, suggesting that the combination treatment of venetoclax and daratumumab should be especially effective in patients with these characteristics. 相似文献

Partial discharge (PD) within voids in a solid insulating system is a symptom and/or a direct cause of deterioration of a generator winding. It is reported that PD is a very short duration current and/or voltage pulse, about 1 to 5 ns. Generally PD emits an electromagnetic wave which is a broadband range signal (≈ gigahertz). Therefore, we developed a new system for detecting the electromagnetic wave, especially microwaves (gigahertz) associated with a material defect of the winding. From the point of view of preventing accidents of insulation breakdown, it is important to assess the condition of the winding. We applied our system to a dielectric breakdown test to learn the relation between the characteristics of the microwaves and the insulation condition of the winding. The dielectric breakdown test is believed to be useful for estimating the minimum breakdown voltage which is a criterion of the insulation condition of the winding. Homogeneous grafting of styrene onto cellulose was carried out in a dimethylacetamide/lithium chloride solvent system. The grafted products were added to cellulose/polystyrene (PS) blends as compatibilizers. The thermal decomposition behavior of the blends was investigated by thermogravimetry. The thermal stability of the blends decreased with an increase in grafted product content. The crystallinity of the blends decreased with grafted product content. The microphase‐separated structures of the blends became finer with grafted product content. The glass transition temperatures for cellulose and PS in the blends were lowered with grafted product content. Differences in thermal decomposition behavior of the blends were correlated with compatibility.
