Cell therapy of the post-infarcted myocardium is still far from clinical use. Poor survival of transplanted cells, insufficient regeneration, and replacement of the damaged tissue limit the potential of currently available cell-based techniques. In this study, we generated a multilayered construct from adipose-derived mesenchymal stromal cells (MSCs) modified to secrete stem cell factor, SCF. In a rat model of myocardium infarction, we show that transplantation of SCF producing cell sheet induced activation of the epicardium and promoted the accumulation of c-kit positive cells in ischemic muscle. Morphometry showed the reduction of infarct size (16%) and a left ventricle expansion index (0.12) in the treatment group compared to controls (24–28%; 0.17–0.32). The ratio of viable myocardium was more than 1.5-fold higher, reaching 49% compared to the control (28%) or unmodified cell sheet group (30%). Finally, by day 30 after myocardium infarction, SCF-producing cell sheet transplantation increased left ventricle ejection fraction from 37% in the control sham-operated group to 53%. Our results suggest that, combining the genetic modification of MSCs and their assembly into a multilayered construct, we can provide prolonged pleiotropic effects to the damaged heart, induce endogenous regenerative processes, and improve cardiac function. 相似文献

Pro-inflammatory cytokines like interleukin-1β (IL-1β) are upregulated during early responses to tissue damage and are expected to transiently compromise the mechanical microenvironment. Fibroblasts are key regulators of tissue mechanics in the lungs and other organs. However, the effects of IL-1β on fibroblast mechanics and functions remain unclear. Here we treated human pulmonary fibroblasts from control donors with IL-1β and used Atomic Force Microscopy to unveil that IL-1β significantly reduces the stiffness of fibroblasts concomitantly with a downregulation of filamentous actin (F-actin) and alpha-smooth muscle (α-SMA). Likewise, COL1A1 mRNA was reduced, whereas that of collagenases MMP1 and MMP2 were upregulated, favoring a reduction of type-I collagen. These mechanobiology changes were functionally associated with reduced proliferation and enhanced migration upon IL-1β stimulation, which could facilitate lung repair by drawing fibroblasts to sites of tissue damage. Our observations reveal that IL-1β may reduce local tissue rigidity by acting both intracellularly and extracellularly through the downregulation of fibroblast contractility and type I collagen deposition, respectively. These IL-1β-dependent mechanical effects may enhance lung repair further by locally increasing pulmonary tissue compliance to preserve normal lung distension and function. (1) Background: Neurogenesis is considered to be a potential brain repair mechanism and is enhanced in stroke. It is difficult to reconstruct the neurogenesis process only from the histological sections taken from different animals at different stages of brain damage and restoration. Study of neurogenesis would greatly benefit from development of tissue-specific visualization probes. (2) Purpose: The study aimed to explore if overexpression of ferritin, a nontoxic iron-binding protein, under a doublecortin promoter can be used for non-invasive visualization of neurogenesis using magnetic resonance imaging (MRI). (3) Methods: Ferritin heavy chain (FerrH) was expressed in the adeno-associated viral backbone (AAV) under the doublecortin promoter (pDCX), specific for young neurons, in the viral construct AAV-pDCX-FerrH. Expression of the enhanced green fluorescent protein (eGFP) was used as an expression control (AAV-pDCX-eGFP). The viral vectors or phosphate-buffered saline (PBS) were injected intracerebrally into 18 adult male Sprague–Dawley rats. Three days before injection, rats underwent transient middle-cerebral-artery occlusion or sham operation. Animals were subjected to In vivo MRI study before surgery and on days 7, 14, 21, and 28 days after injection using a Bruker BioSpec 11.7 T scanner. Brain sections obtained on day 28 after injection were immunostained for ferritin, young (DCX) and mature (NeuN) neurons, and activated microglia/macrophages (CD68). Additionally, RT-PCR was performed to confirm ferritin expression. (4) Results: T2* images in post-ischemic brains of animals injected with AAV-pDCX-FerrH showed two distinct zones of MRI signal hypointensity in the ipsilesioned hemisphere starting from 14 days after viral injection—in the ischemic lesion and near the lateral ventricle and subventricular zone (SVZ). In sham-operated animals, only one zone of hypointensity near the lateral ventricle and SVZ was revealed. Immunochemistry showed that ferritin-expressing cells in ischemic lesions were macrophages (88.1%), while ferritin-expressing cells near the lateral ventricle in animals both after ischemia and sham operation were mostly mature (55.7% and 61.8%, respectively) and young (30.6% and 7.1%, respectively) neurons. RT-PCR confirmed upregulated expression of ferritin in the caudoputamen and corpus callosum. Surprisingly, in animals injected with AAV-pDCX-eGFP we similarly observed two zones of hypointensity on T2* images. Cellular studies also showed the presence of mature (81.5%) and young neurons (6.1%) near the lateral ventricle in both postischemic and sham-operated animals, while macrophages in ischemic lesions were ferritin-positive (98.2%). (5) Conclusion: Ferritin overexpression induced by injection of AAV-pDCX-FerrH was detected by MRI using T2*-weighted images, which was confirmed by immunochemistry showing ferritin in young and mature neurons. Expression of eGFP also caused a comparable reduced MR signal intensity in T2*-weighted images. Additional studies are needed to investigate the potential and tissue-specific features of the use of eGFP and ferritin expression in MRI studies. 相似文献

The selectivity of encapsulation of leflunomide and teriflunomide by native α-, β- and γ-cyclodextrins was investigated through 1H NMR and molecular modeling. Thermodynamic analysis revealed the main driving forces involved in the binding. For α-cyclodextrin, the partial encapsulation was obtained while deep penetration was characterized for the other two cyclodextrins, where the remaining polar fragment of the molecule is located outside the macrocyclic cavity. The interactions via hydrogen bonding are responsible for high negative enthalpy and entropy changes accompanying the complexation of cyclodextrins with teriflunomide. These results were in agreement with the molecular modeling calculations, which provide a clearer picture of the involved interactions at the atomic level. 相似文献

A 25 nm thick α-alumina layer was deposited on a turbine-grade silicon nitride by sol-gel dip coating and subsequent heat treatment in air at 1200°C. This layer had a nanometer grain structure. Silicon nitride protected by this thin layer showed a significant improvement in oxidation resistance over its uncoated counterpart after 200 cyclic exposures in air at 1250°C. Quasiliving cationic polymerization of styrene was obtained in the system 2-phenyl-2-propano-/AlCl3·OBu2/Bu2O in a mixture of 1,2-dichloroethane and n-hexane (55:45 viv) at -15 °C. The molecular weights of the polymers (Mn) increased in direct proportion to the monomer conversion. However, the experimental Mns are essentially higher than theoretical ones, indicating that slow initiation relative to propagation takes place. The molecular weight distributions were broad (Mw/Mn2.5), probably due to the slow initiation and slow exchange between reversibly terminated and propagating species. 相似文献

Technical lignins are complex, irregular, polyphenolic compounds obtained in large quantities as by-products of the pulp and paper industries or according to current biorefinery setups. The availability of kraft lignin is increasing due to larger scale retrieval from process liquors, which opens new possibilities for further refining or new applications of such lignins. In the present study, sequential ultrafiltration of kraft lignin was performed to fractionate the lignin and to elucidate molar mass-dependent changes in lignin structure. Two industrial black liquors and three precipitated lignins were fractionated, and their functional groups were determined, providing molar mass-dependent profiles. Interrelations between structural parameters and functional groups, the molecular weight ranges, and the different lignin sources are discussed. This will help to establish structure-property-application relationships (SPARs) for technical lignins which are required for any future large-scale application. 相似文献

Cu2+ and Ni2+ complexes of chitosan/poly(vinyl amine) (CS/PVAm) composites were prepared. The metal-CS/PVAm complexes were characterized by FT-IR spectroscopy, SEM-EDX, X-ray diffraction (XRD), and magnetic moment determination. FT-IR spectra of the metal-CS/PVAm complexes showed the characteristic bands of anhidroglucose unit were affected by the metal complexation, and new bands assigned to Me–N and Me-O bonds were observed. SEM images of the surface of metal-composite complexes show the influence of metal ion on the morphology of the complexes, the strong binding of Cu2+ and Ni2+, involving most of the amino groups, leading to a dense surface structure. The chemical composition on the surface of metal-CS/PVAm complexes was determined from EDX measurements. XRD provided information about the amorphous or crystalline nature of the composite and metal-composite complexes. Using magnetic susceptibility method, the oxidation degree of metal ions from the polymer phase, the homogeneous distribution of the ligand groups from the volume of the CS/PVAm beads, and the existence of the antiferromagnetic interactions between the metal ions were determined. 相似文献

In this paper a detailed study of the (ZrO2)1‐x(Y2O3)x (x=0.025–0.15), (ZrO2)1‐x(Sc2O3)x (x = 0.06 – 0.11) and (ZrO2)1‐x‐y(Sc2O3)x(Y2O3)y (x=0.07 – 0.11; y=0.01 – 0.04) solid solution crystals grown by skull melting technique is presented. The structure, phase composition, and ion conductivity of the obtained crystals were investigated by X‐ray diffraction, transmission electron microscopy, Raman scattering spectroscopy, and impedance spectroscopy. Maximum conductivity as (ZrO2)1‐x(Y2O3)x and (ZrO2)1‐x(Sc2O3)x solid solution crystals is observed for the compositions containing 10 mol% stabilizing oxide, and the conductivity of 10ScSZ is ~3 times higher than for 10YSZ. Experiments on crystal growth (ZrO2)1‐x‐y(Sc2O3)x(Y2O3)y solid solutions showed that uniform, transparent crystals 7Sc3YSZ, 7Sc4YSZ, 8Sc2YSZ, 8Sc3YSZ, 9Sc2YSZ, 9Sc3YSZ, 10Sc1YSZ, and 10Sc2YSZ are single phase crystal containing t″ phase. It is established that a necessary condition of melt growth of (ZrO2)1‐x‐y(Sc2O3)x(Y2O3)y single‐phase crystals is the total concentration of the stabilizing oxides from 10 to 12 mol%. The addition of Y2O3 affects the (ZrO2)1‐x‐y(Sc2O3)x(Y2O3)y solid solution conductivity different ways and depends on the Sc2O3 content in the starting composition. The effects of structure, phase composition, concentration, and type of stabilizing oxides on the electrical characteristics of obtained crystals are discussed. 相似文献

Succinic acid has been pertracted with TOA using free liquid membranes without or with 1‐octanol. The addition of the alcohol led to the increase of up to 2.8–3 times of the acid's initial and final mass flows. At the same time, the influence of 1‐octanol on the transport capacity of the pertraction system was negative, its addition inducing the accumulation of succinic acid into the liquid membrane. A mathematical model describing the acid accumulation inside the liquid membrane has been developed for pertraction systems without and with 1‐octanol and offers good concordance with the experimental data. © 2012 Canadian Society for Chemical Engineering 相似文献
