DX-8951f, a water-soluble and non-pro-drug analogue of camptothecin, exhibits a strong inhibitory action on DNA topoisomerase I (Topo I) and in vitro cytotoxicity against various human cancer cell lines. In order to elucidate the mechanisms of its cytotoxicity, we established a DX-8951f-resistant cell line, SBC-3/DXCL1, from human small cell lung cancer cells (SBC-3) by stepwise exposure to DX-8951f. SBC-3/DXCL1 cells were approximately 400 times more resistant to DX-8951f than parent cells. The SBC-3/DXCL1 cells showed a high degree of cross-resistance to other Topo I inhibitors such as CPT-11, SN-38 and camptothecin, but not to non-Topo I targeting agents such as cisplatin, adriamycin, etoposide, and vincristine. The mechanisms of resistance of SBC-3/DXCL1 cells to DX-8951f were examined. Intracellular accumulation of DX-8951f by SBC-3 and SBC-3/DXCL1 cells did not differ significantly. Although the Topo I activity of nuclear extracts obtained from SBC-3/DXCL1 cells was the same as that of the parent cells, the Topo I of SBC-3/DXCL1 cells was resistant to the inhibitory effects of DX8951f and SN-38. Immunoblotting using anti-Topo I antibody demonstrated similar protein levels of Topo I in SBC-3 and SBC-3/DXCL1 cells. The active Topo I protein of SBC-3/DXCL1 was eluted by a high concentration of NaCl (0.4 N) compared with that of SBC-3 (0.3 N). DX-8951f stabilized the DNA-Topo I cleavable complex from SBC-3 cells, as measured by Topo I-mediated cleavage assay. In SBC-3/DXCL1 cells, DX-8951f also stabilized the DNA-Topo I complex, but with a 10-fold lower efficiency. These results suggest that a qualitative change in Topo I contributes, at least partially, to the resistance to DX-8951f in SBC-3/DXCL1 cells. Therefore, SBC-3/DXCL1 cells may have a unique mechanism of resistance to Topo I-directed antitumor drugs.  相似文献

During the operation, fluctuations of fuel and electricity costs significantly impact on the stability of the profit from ESCO business. Therefore, it is essential to reduce the risk of fuel and electricity cost fluctuations. Generally, a transaction called “financial derivative” is used as a measure of hedging against the fuel price fluctuation. In the case of ESCO business, it is necessary to manage the risk of both electricity and fuel price fluctuations because the variation in electricity price strongly affects the profit from ESCO as that in fuel price does.  相似文献

A new method for determining suitable single-mode fibre parameters is examined. Suitable fibre parameters are determinable by minimising cabling- and splicing-loss increases. The authors have been studying Chemical Heat Pumps (CHP) from the viewpoints of energy saving and environmental impact. The CHP can store thermal energy in the form of chemical energy by an endothermic reaction, and release it at various temperature levels during heat demand periods by exo/endothermic reactions. The authors have proposed in an earlier study a novel chemical heat pump (CHP) system for environmentally-friendly effective utilization of thermal energy in drying as a chemical heat pump dryer (CHPD). In this exploratory study, we test the effectiveness of operating the proposed CHPDs experimentally. Basic experiments on the CHPDs such as hot dry air production for convective drying are performed on lab-scale CHPD apparatuses using gas-solid reactions in calcium oxide/calcium hydroxide reactant beds. The proposed CHPDs are found to produce hot air by CHP operation for drying. The temperature levels of the produced hot air and the reaction rates/conversions are as good as in the case of hot water supply system using basically same CHP operation. The cold heat for air dehumidification is also found to be generated/recovered by the same CHPD system. The generated heat amounts can be increased by changing the operating conditions although the heat recovery must be enhanced for practical application of CHPDs.  相似文献

Relationships between α-tocopherol concentration in the muscle and development of lipid oxidation or volatiles in raw venison were studied. Fourteen Japanese Shika Deer (Cervus nippon) were fed various amounts (0–3.0 g of α-tocopheryl acetate per animal) during the different periods (0–37 days) and then M. longissimus thoracium et lumborum (LD muscles) with a range of α-tocopherol concentrations (4.1–15.1 mg/kg tissue) were obtained. For stabilizing the lipid during storage for 11 days under air, over ca. 9 mg of α-tocopherol per kg tissue were required based on levels of 2-thiobarbituric acid reacting substances (TBARS) numbers. Nine compounds were identified in headspace volatiles, and one of the volatiles was hexanal which has been recognized as off-flavour component. For depressing the hexanal evolution, at least ca. 9 mg of α-tocopherol per kg tissue were also required for 11 days’ storage. This value was much higher than other species. The reasons for higher requirement of α-tocopherol were possibly due to the higher concentration of unsaturated fatty acid and myoglobin in venison.  相似文献

A two-stage absorption heat transformer (TAHT) and its improved version, the cycle incorpaorating latent heat and sensible heat modes, are investigated by emplying energy-utilization diagrams (EUDs). Utilizing the exhaust gas of a gas turbine as the heat source and employing the water/lithium bromide as the working fluid, it has been concluded that this is an effective way of decreasing the exergy loss by combining latent and sensible heat exchange modes. Furthermore, the temperature of the useful heat is increased. This analysis can provide a basis for synthesis and development of absorption heat transformer systems.  相似文献

A thermodynamic assessment of the Bi-Sn-Zn ternary system was carried out by considering the experimental data including the phase equilibria and thermodynamic properties on the basis of the CALPHAD method. A set of optimized thermodynamic parameters has been obtained, which leads to a very good fit between calculation and experiments. In particular, the thermodynamic calculations in the Sn-rich portion are presented in view of the recent progress in Pb-free solder alloys.  相似文献

Phase equilibria in the bcc Fe-Ge system have been investigated by means of electron probe microanalysis and transmission electron microscopy. Two kinds of ordered phases, B2 and DO3 types, have been detected, the B2 phase being formed from the disordered A2 phase by a 2nd order transition. The DO3 phase is also formed from the B2 phase by a 2nd or higher order transition at higher temperatures. Below about 820 °C, however, the transition becomes a 1st order, and a miscibility gap between these ordered phases occurs in the composition range from 13 to 16 at. pct Ge. It has been also confirmed that the solubility of Ge in the bec phase shows a retrogression due to the formation of DO3 phase. On the basis of these experimental results, the phase diagram for the Fe-Ge system has been partly revised.  相似文献
