Four proteases (CP‐I, ‐II, ‐111 and ‐IV) were isolated and purified from hepatopancreas of crawfish. Based upon their activities with known inhibitors, they resembled trypsin. All were cross‐reactive with polyclonal antibodies raised against CP‐II, indicating they shared structural components. Each of these was able to inactivate orange peel pectinesterase, tomato pectinesterase and pectate lyase C at room temperature under nondenaturing conditions. Using matrix‐assisted‐laser‐desorption time of flight mass spectrometry (MALDl‐TOF MS), an analysis of peptides generated during proteolysis of pectate lyase C from Erwinia chrysenthemi showed similar peptide patterns for the four crawfish proteases, indicating a common specificity for each isozyme. However, the cleavage patterns were different from those obtained by the action of bovine trypsin on pectate lyase C. These studies indicate the four proteases from the hepatopancreas of crawfish are isozymes which may be used for the inactivation of pectinolytic enzymes.  相似文献

Some composites of multiwalled carbon nanotubes, which were chemically treated in acidic and/or hydrogen peroxide solution, and platinum nanoparticles were prepared by the simple reduction in glycerol solution. Carboxylated and/or hydroxyl MWNTs were structurally analysed using X-ray photoelectron spectroscopy. In addition, the MWNT?CPt composites were characterized by XRD and TEM in detail. The electrochemical oxidation of some basic alcohols, which was catalyzed by the MWNT?CPt composites, was analysed by cyclic voltammetry. Their catalytic activities were studied with cyclic voltammograms of alcohols.  相似文献

Exogenously delivered mesenchymal stromal cells (MSCs) are therapeutically beneficial owing to their paracrine effect; they secrete various cytokines, nucleic acids, and proteins. Multiple bioengineering techniques can help MSC cultures to release secretomes by providing stem cell niche-like conditions (both structurally and functionally). Various scaffolds mimic the natural extracellular matrix (ECM) using both natural and synthetic polymers, providing favorable environments for MSC proliferation and differentiation. Depending on material properties, either topographically or elastically structured scaffolds can be fabricated. Three-dimensional scaffolds have tunable substrate rigidities and structures, aiding MSC cultivation. Decellularized ECM-derived hydrogels are similar to the natural ECM, thus improving the paracrine effects of MSCs. Here, we discuss recent research on the application of scaffolds to maximize the immunomodulatory function of MSCs.  相似文献

Quantum nonlocality is tested for an entangled coherent state, interacting with a dissipative environment. A pure entangled coherent state violates Bell's inequality regardless of its coherent amplitude. The higher the initial nonlocality, the more rapidly quantum nonlocality is lost. The entangled coherent state can also be investigated in the framework of 2 X 2 Hilbert space. The quantum nonlocality persists longer in 2 X 2 Hilbert space. When it decoheres it is found that the entangled coherent state fails the nonlocality test, which contrasts with the fact that the decohered entangled state is always entangled.  相似文献

A discrete time domain modelling is presented for the pulse-width modulated series resonant converter (PWM-SRC) with a discontinuous current mode. This nonlinear system is linearized about its equilibrium state to obtain a linear discrete time model for the investigation of small signal performances such as the stability and transient response. The usefulness of this small signal model is verified by dynamic simulation.  相似文献

The present study was designed to examine the physicochemical and sensory properties in cholesterol‐reduced Camembert cheese made by crosslinked β‐cyclodextrin (β‐CD). The composition of cholesterol‐reduced Camembert cheese was similar to the control and the cholesterol reduction reached 90.6%. No difference was found in the total amount of short‐chain free fatty acids between the cholesterol‐reduced cheese and the control at every ripening period. The release of butyric and capric acid mostly contributed to the increase of total amount of short‐chain free fatty acids in both groups. The cholesterol‐reduced cheese produced similar amounts of individual free amino acids to the control in all periods. The scores of all rheological characteristics except for springiness were continuously increased up to 2 weeks’ ripening and decreased thereafter. Mouldy characteristics in both appearance and flavour were increased dramatically through the ripening period in both cholesterol‐reduced and the control cheeses. Based on these results, no significant difference was found in most physicochemical and sensory properties between cholesterol‐reduced Camembert cheese and the control. Therefore, we may find it possible to develop cholesterol‐reduced Camembert cheese using crosslinked β‐cyclodextrin.  相似文献

We report the cytotoxicity of the ginseng saponin metabolite, Compound K (20‐O‐D‐glucopyranosyl‐20(S)‐protopanaxadiol, IH901) on various human leukemia cell lines. Compound K had the most effect on U937, a human monocytic leukemia cell line among the tested cell lines. Compound K‐treated U937 cells showed characteristics of apoptosis: an exposure of phosphatidylserine from the inner cell membrane to the outer cell membrane, the formation of apoptotic bodies and DNA fragmentation. Compound K induced apoptosis by up‐regulating Bax, disrupting the mitochondrial membrane potential, and by activating caspase 9 and caspase 3. Therefore, we suggest that Compound K inhibits U937 cell growth by inducing apoptosis through the up‐regulation of Bax and caspase activation.  相似文献
