Many organisms contain head‐to‐head isoprenoid synthases; we investigated three such types of enzymes from the pathogens Neisseria meningitidis, Neisseria gonorrhoeae, and Enterococcus hirae. The E. hirae enzyme was found to produce dehydrosqualene, and we solved an inhibitor‐bound structure that revealed a fold similar to that of CrtM from Staphylococcus aureus. In contrast, the homologous proteins from Neisseria spp. carried out only the first half of the reaction, yielding presqualene diphosphate (PSPP). Based on product analyses, bioinformatics, and mutagenesis, we concluded that the Neisseria proteins were HpnDs (PSPP synthases). The differences in chemical reactivity to CrtM were due, at least in part, to the presence of a PSPP‐stabilizing arginine in the HpnDs, decreasing the rate of dehydrosqualene biosynthesis. These results show that not only S. aureus but also other bacterial pathogens contain head‐to‐head prenyl synthases, although their biological functions remain to be elucidated. 相似文献

The fictive temperature of glass characterizes the glass network structure and its thermal history, and thereby can influence ion and water transport in the glass surface. In this study, IR specular reflectance (SR), refractive index, and density measurements were used to characterize and confirm the effects of glass sample processing, especially the fictive temperature/thermal‐history variations. The subsequent acid leaching of these glasses created leached surface layers due to interdiffusion and reaction of hydrous species in the surface; the hydrogen depth profiles obtained with secondary ion mass spectrometry (SIMS) confirmed enhanced leaching with increasing fictive temperature. Attenuated total reflectance (ATR) in the mid‐ and near‐IR indicated increases in both SiOH and H2O species with increasing fictive temperature. The relative intensities and shapes of the ATR peaks were found to vary between the samples suggesting that speciation of the hydrous reaction products (eg, strong and weakly hydrogen‐bonded OH) is also influenced by the original fictive temperature of the glass, but could not be quantitatively determined. 相似文献

This paper introduces the concept of migration cost in analyzing the interregional allocation of population. Migration cost gives the social planner an additional burden of population reallocation, and it has an important effect upon an individual's decisions on migration in a decentralized market mechanism. Interregional migration is influenced by migration cost, initial state of the population, etc. Though the difference of marginal productivity of labor and the consumption of composite good in one region is greater than that in another region, migration will not be desirable if the difference of net benefit between two regions from migration does not cover migration cost. Therefore, there will be a range of initial population allocation for which no migration occurs, not only for the social planner's problem but also for the individual's migration decision. Comparison of populations between social optimum and market equilibrium with central government intervention revealsmigration cost externality. Traffic emissions have been associated with a wide range of adverse health effects. Many schools are situated close to major roads, and as children spend much of their day in school, methods to reduce traffic‐related air pollutant concentrations in the school environment are warranted. One promising method to reduce pollutant concentrations in schools is to alter the timing of the ventilation so that high ventilation time periods do not correspond to rush hour traffic. Health Canada, in collaboration with the Ottawa‐Carleton District School Board, tested the effect of this action by collecting traffic‐related air pollution data from four schools in Ottawa, Canada, during October and November 2013. A baseline and intervention period was assessed in each school. There were statistically significant (P < 0.05) reductions in concentrations of most of the pollutants measured at the two late‐start (9 AM start) schools, after adjusting for outdoor concentrations and the absolute indoor–outdoor temperature difference. The intervention at the early‐start (8 AM start) schools did not have significant reductions in pollutant concentrations. Based on these findings, changing the timing of the ventilation may be a cost‐effective mechanism of reducing traffic‐related pollutants in late‐start schools located near major roads. 相似文献

Enhanced ammonia nitrogen removal using consistent biological regeneration and ammonium exchange of zeolite in modified SBR process 总被引:13,自引:0,他引:13
The modified zeo-SBR is recommended for a new nitrogen removal process that has a special function of consistent ammonium exchange and bioregeneration of zeolite-floc. Three sets of sequencing batch reactors, control, zeo-SBR, and modified zeo-SBR were tested to assess nitrogen removal efficiency. The control reactor consisted of anoxic-fill, aeration-mixing, settling, and decanting/idle phases, meaning that nitrogen removal efficiency was dependent on the decanting volume in a cycle. The zeo-SBR reactor was operated in the same way as the control reactor, except for daily addition of powdered zeolite in the SBR reactor. The operating order sequences in the zeo-SBR were changed in the modified zeo-SBR. Anoxic-fill phase was followed by aeration-mixing phase in the zeo-SBR, while aeration-mixing phase was followed by anoxic-fill phase in the modified zeo-SBR to carry NH4(+)-N over to the next operational cycle and to reduce total nitrogen concentration in the effluent. In the modified zeo-SBR, nitrification and biological regeneration occurred during the initial aeration-mixing phase, while denitrification and ammonium adsorption occurred in the following anoxic-fill phase. The changed operational sequence in the modified zeo-SBR to adapt the ammonium adsorption and biological regeneration of the zeolite-floc could enhance nitrogen removal efficiency. As a result of the continuous operation, the nitrogen removal efficiencies of the control and zeo-SBR were in 68.5-70.9%, based on the 33% of decanting volume for a cycle. The zeo-SBR showed a consistent ammonium exchange and bio-regeneration in the anoxic-fill and aeration-mixing phases, respectively. Meanwhile, the effluent total nitrogen of the modified zeo-SBR showed 50-60 mg N/L through ammonium adsorption of the zeolite-floc when the influent ammonium concentration was 315 mg N/L, indicating the T-N removal efficiency was enhanced over 10% in the same HRT and SRT conditions as those of control and zeo-SBR reactors. The ammonium adsorption capacity was found to be 6-7 mg NH4(+)-N/g FSS that is equivalent to 40 mg NH4(+)-N/L of ammonium nitrogen removal. 相似文献

This work concerns mechanical properties of an austempered ductile cast iron (ADI). Samples alloyed with copper and molybdenum were austenitized at 910 °C for 90 min and subsequently austempered in a salt bath over a range of temperatures from 350 °C to 410 °C to obtain favorable mechanical properties such as tensile strength, elongation, and fracture toughness. Laser hardening provides benefits over the conventional hardening processes, including minimum distortion in the parts and the absence of a quenchant. This process is also faster than conventional hardening processes and can be used for selective hardening of specific areas of components. One known problem with laser hardening in steels, however, is back tempering when a large area is hardened by multiple, overlapping passes. This study focused on the development of a numerical model to predict the back tempering in multi-track laser hardening. A tempering model was combined with existing models of thermal behavior and phase change kinetics, which were developed earlier in the authors’ group, to predict three-dimensional hardness profiles after multiple track laser hardening. The combined model was first validated through multi-track laser hardening tests and then used to predict and optimize the laser hardened case depth in multi-track laser hardening of AISI 4140 steel. The predictions and parameters optimized to obtain maximum case depth with the least variation along width of the hardened zone were experimentally verified.
