Staphylococcus aureus is one of the most prevalent pathogens associated with several types of biofilm-based infections, including infections of chronic wounds. Mature staphylococcal biofilm is extremely hard to eradicate from a wound and displays a high tendency to induce recurring infections. Therefore, in the present study, we aimed to investigate in vitro the interaction between S. aureus biofilm and fibroblast cells searching for metabolites that could be considered as potential biomarkers of critical colonization and infection. Utilizing advanced microscopy and microbiological methods to examine biofilm formation and the staphylococcal infection process, we were able to distinguish 4 phases of biofilm development. The analysis of staphylococcal biofilm influence on the viability of fibroblasts allowed us to pinpoint the moment of critical colonization—12 h post contamination. Based on the obtained model we performed a metabolomics analysis by 1H NMR spectroscopy to provide new insights into the pathophysiology of infection. We identified a set of metabolites related to the switch to anaerobic metabolism that was characteristic for staphylococcal biofilm co-cultured with fibroblast cells. The data presented in this study may be thus considered a noteworthy but preliminary step in the direction of developing a new, NMR-based tool for rapid diagnosing of infection in a chronic wound. 相似文献
Coumarins belong to a group of secondary metabolites well known for their high biological activities including antibacterial and antifungal properties. Recently, an important role of coumarins in plant resistance to pathogens and their release into the rhizosphere upon pathogen infection was discovered. It is also well documented that coumarins play a crucial role in the Arabidopsis thaliana growth under Fe-limited conditions. However, the mechanisms underlying interplay between plant resistance, accumulation of coumarins and Fe status, remain largely unknown. In this work, we investigated the effect of both mentioned factors on the disease severity using the model system of Arabidopsis/Dickeya spp. molecular interactions. We evaluated the disease symptoms in Arabidopsis plants, wild-type Col-0 and its mutants defective in coumarin accumulation, grown in hydroponic cultures with contrasting Fe regimes and in soil mixes. Under all tested conditions, Arabidopsis plants inoculated with Dickeya solani IFB0099 strain developed more severe disease symptoms compared to lines inoculated with Dickeya dadantii 3937. We also showed that the expression of genes encoding plant stress markers were strongly affected by D. solani IFB0099 infection. Interestingly, the response of plants to D. dadantii 3937 infection was genotype-dependent in Fe-deficient hydroponic solution. 相似文献

Expanded bed behavior was modeled by using the Richardson-Zaki correlation between the superficial velocity of the feed stream and the void fraction of the bed. A polydisperse material, Chelating excellose® (70-210 Μm in diameter, 1.21 g/cm3 in density), which has Ni2+ ions for the selective binding of histidine-tagged proteins, was used as the resin. A method to modify the Stokes expression to express the terminal settling velocity of the resins by introducing two empirical parameters, the effective diameter of the resins and an exponent for(ρp-ρ)/Μ term, was developed. Combined use of the Richardson-Zaki correlation and the modified Stokes expression was successful in modeling the bed expansion by incorporating physical properties of feed streams and the resins. 相似文献
Porous carbon beads were prepared by carbonizing at 700 and 1000 °C under N2 (NN-series) or CO2 atmospheres (CO-series) and a subsequent oxidization with boiling nitric acid solution (13%, v/v) for 3 h (ANN- and ACO-series). BET surface area of both CO- and NN-series samples tends to increase with increasing carbonization temperature, but CO-series samples show higher BET surface areas (700 °C: 610 m2/g, 1000 °C: 780 m2/g) compared with those of NN-series samples (700 °C: 380 m2/g, 1000 °C: 580 m2/g). After acid oxidation, BET surface areas of NN-series samples increased from 580 to 650 m2/g, whereas those of CO-series samples decreased from 780 to 600 m2/g. On subsequent acid oxidation, acidic surface functional groups increased in both CO- and NN-series samples, but the CO-series samples tend to have much more acidic surface functional groups. Adsorption of halogenated methanes, such as tetra-, tri- and dichloromethanes, onto the samples of CO-, NN-, ACO- and ANN-series was thought to be driven initially by the basicity, but the acidic functional groups that could attract adsorbates via dipole-dipole interaction might hinder the adsorption of subsequently incoming adsorbates. 相似文献
Verifiably encrypted signature schemes can convince a verifier that a given ciphertext is an encryption of an ordinary signature on a given message and the ordinary signature can be recovered by the third party, called adjudicator. In 2010, Rückert et al. proposed a general construction for the verifiably encrypted signatures, and then, they also showed that there exist the lattice-based verifiably encrypted signature schemes. Their constructions are very insightful, but their schemes need an extra adjudication setup phase and Merkle trees, so they have large parameters and keys, that is, they are inefficient. Also, their schemes provide only the limited signature capacity because the signing keys should be reissued after generating \(k\)th verifiably encrypted signatures. To overcome the weaknesses of Rückert et al.’s scheme, we construct a verifiably encrypted signature scheme based on the hard lattice problems. Our scheme provides the full functionality, i.e., the signatures can be generated without any limitations and does not need any extra adjudication setup phases. Moreover, the size of the secret keys in our scheme is constant. Our scheme provides unforgeability, opacity, extractability, and abuse-freeness in the random oracle model. 相似文献
The research focuses on the perception of the Mucharz reservoir in relation to costs and benefits for two types of communities. The primary aim of the paper is to compare the social impact of dam projects among both resettled and non-resettled local communities. The secondary aim of the paper is to test multivariate analysis of variance (MANOVA) as a tool to examine the social impact assessment (SIA) of dam projects. The studied dam is located on the Skawa River within the Polish Carpathians. We used in-depth interviews with a non-resettled (N?=?96) and resettled (N?=?57) local population. To analyse the collected data, we performed multivariate analysis of variance. The main results indicate that the social assessment of dam projects differs significantly depending on whether one belongs to the group of resettled or non-resettled population. MANOVA is an effective tool for the analysis of the social impact assessment (SIA) of dam projects.
Spontaneous charging/discharging processes of polymers: polypyrrole and poly(N-methylpyrrole), doped with perchlorate (anion exchanging PPy and PMPy, respectively) and poly(4-styrenesulphonate) ions (cation exchanging PPy(PSS) and PMPy(PSS)) occurring in aqueous electrolyte solutions were studied using different electrochemical techniques. These reactions (oxidation of the polymer by dissolved oxygen and discharge of the polymer redox capacitance) are usually undesired processes leading to alteration of the charge accumulated in the polymer films.
