Poly(ethylene terephthalate) (PET) is a highly inert packaging material that exhibits low interaction with foodstuff and consequently a limited diffusion of migrants. Migration modeling can therefore be used as an alternative to experimental migration tests in order to confirm compliance of PET packaging materials with food laws. The most important factor for predicting migration using mathematical models is the diffusion coefficient of the migrant in PET. However, current models that predict this parameter are typically based on worst‐case scenarios and thereby significantly over‐estimate the degree of migration. The key parameter for developing more realistic migration models is the activation energy of diffusion of potential migrants in PET, but experimental data on this are scarcely available in the scientific literature. The aim of the present study was therefore to develop a fast and precise method for determining diffusion coefficients and activation energies of diffusion of organic compounds in PET. Activation energies of diffusion for 13 organic compounds in PET were determined via their diffusion coefficient temperature dependencies. The molecular weight and activation energy of diffusion for the compounds investigated in this study were correlated, offering a basis for a new approach in predicting diffusion coefficients for use in migration modeling. In earlier work, we have observed discrepancies relating to the early hydration of calcium aluminate cement (CAC) when comparing data from heat flow calorimetry of CAC paste with results from mortar strength tests using the crushing method. Here, we investigated on this phenomenon and found that the sand which is used as a filler exerts a major influence on CAC hydration resulting in acceleration. Furthermore, in particular fine filler materials such as, for example, microsilica, fine limestone powder, and especially α- and γ-Al2O3 also produced a strong hydration accelerating effect which is dependent on their specific surface area. The mechanism underlying the acceleration is that under alkaline conditions their negative surface charge attracts calcium ions as was confirmed via inductively coupled plasma atomic emission measurements. Such a layer generates favourable conditions for the nucleation of CAC hydration products (C-A-H phases). The resulting crystalline hydrates which form on the surface of the filler particles submerged in CAC cement pore solution were visualized via SEM imaging. This way, specifically selected fillers can significantly accelerate CAC hydration and save precious lithium salts which are commonly used to boost the early strength of CAC. 相似文献

Spliceosome-mediated RNA trans-splicing has become an emergent tool for the repair of mutated pre-mRNAs in the treatment of genetic diseases. RNA trans-splicing molecules (RTMs) are designed to induce a specific trans-splicing reaction via a binding domain for a respective target pre-mRNA region. A previously established reporter-based screening system allows us to analyze the impact of various factors on the RTM trans-splicing efficiency in vitro. Using this system, we are further able to investigate the potential of antisense RNAs (AS RNAs), presuming to improve the trans-splicing efficiency of a selected RTM, specific for intron 102 of COL7A1. Mutations in the COL7A1 gene underlie the dystrophic subtype of the skin blistering disease epidermolysis bullosa (DEB). We have shown that co-transfections of the RTM and a selected AS RNA, interfering with competitive splicing elements on a COL7A1-minigene (COL7A1-MG), lead to a significant increase of the RNA trans-splicing efficiency. Thereby, accurate trans-splicing between the RTM and the COL7A1-MG is represented by the restoration of full-length green fluorescent protein GFP on mRNA and protein level. This mechanism can be crucial for the improvement of an RTM-mediated correction, especially in cases where a high trans-splicing efficiency is required. 相似文献

Human pluripotent stem cells (hPSCs) are attractive resources for regenerative medicine, but medical applications are hindered by their tumorigenic potential. Previously, a hPSC-specific lectin probe, rBC2LCN, was identified through comprehensive glycome analysis by using high-density lectin microarrays. Herein, a lectin–doxorubicin (DOX) prodrug conjugate, with controllable photolysis activation for the elimination of tumorigenic human induced pluripotent stem cells, has been developed. rBC2LCN was fused with a biotin-binding protein, tamavidin (BC2Tama), and the fusion protein was expressed in Escherichia coli and purified by means of affinity chromatography. BC2Tama was then conjugated with doxorubicin-photocleavable biotin (DOXPCB). The BC2Tama–DOXPCB conjugates were observed to bind to hPSCs followed by internalization. Upon exposure to ultraviolet light, DOX was released inside the cells, which allowed specific killing of the hPSCs. Thus, BC2Tama–DOXPCB should be useful for the targeted elimination of hPSCs contained in hPSC-derived cell therapy products. This is the first report of the generation of lectin–prodrug conjugates. BC2Tama should be applicable for the targeted delivery of various types of biotinylated compounds into hPSCs. 相似文献

This study investigated the effects of variations in oil contents and fatty-acid composition, density, viscosity, acid values, saponification values, specific oxidative stability, and antioxidant concentration of Acrocomia totai kernel oil during fruit maturation. Fatty acids were quantified using 1H nuclear magnetic resonance (NMR) spectroscopy, gas chromatograph-flame-ionization detector (GC-FID), and Fourier transform Raman (FT-Raman) spectroscopy analyses. The results showed that all physicochemical characteristics and oil composition changed during the ripening stage. The CG-FID analysis showed a reduction in the unsaturated fatty-acid content (from 78.8% to 22.1%), with a proportional increase in the saturated fatty-acid contents (from 21.6% to 77.9%). The difference in the fatty-acid composition was confirmed by analysis of the 1H NMR and FT-Raman spectra. The degree of unsaturation was calculated to determine the oxidative stability of oil. These results suggest that the fruit's maturation contributes to the specific oxidative stability. The antioxidant concentration revealed higher contents of carotenoids in the ripe fruit (0.16 mg of carotenoid per 100 gKERNEL) when compared to the unripe fruit (0.05 mg of carotenoid per 100 gKERNEL). In the total phenolics analysis, there was no change in concentration over ripening time. These results show that kernel oil has physicochemical properties comparable with high-quality commercial vegetable oils, suggesting that it is a promising alternative to conventional vegetable oils. 相似文献

The last advances in the synthesis and shaping techniques of chalcogenide glasses suitable for infrared photonics are reported. Ball milling combined with spark plasma sintering allows the preparation of bulk chalcogenide glasses at lower temperature and at lower cost. Microstructuring of optical fibers results in enhanced nonlinear properties leading to the demonstration of low-threshold Brillouin laser at 1.55 μm, all-optical wavelength conversion and time-domain demultiplexing with a 170 Gb/s rate, and supercontinuum between 1.5 and 2.8 μm.
