Mineralocorticoid action is facilitated by 11 beta-hydroxysteroid dehydrogenase type II (11 beta HSD2), which metabolizes glucocorticoids and allows aldosterone to bind to the nonselective mineralocorticoid receptor. We have recently demonstrated the presence of the 11 beta HSD2 protein in a wide range of human epithelia, suggesting that it is the sole isoform endowing specificity in man. In the present study we have used an immunopurified polyclonal antibody (RAH23) raised against a C-terminal peptide derived from the cloned rat 11 beta HSD2 protein to perform immunohistochemical and molecular analysis in rat tissues. In frozen sections of rat kidney, strong staining was seen with the RAH23 antibody in the distal tubule; weaker staining was observed in the thick ascending loop of Henle and the medullary and papillary collecting ducts. Punctate cortical staining was observed in the fetus at 20 days gestation and in 8-day-old rats, with a noticeable increase in the staining pattern at 16 days of age. The kidney did not attain the adult pattern of staining until 28 days of age. Epithelia of ileum and colon also stained with RAH23, as did excretory ducts of the submandibular gland. Intrahepatic and excretory bile ducts displayed strong immunoreactivity in the epithelial lining. Rat adrenal glands showed evidence of the 11 beta HSD2 antigen in the zona fasciculata and zona reticularis, but not in the zona glomerulosa or medulla. Western blot analysis with the RAH23 antibody revealed strong bands in the kidney, colon, adrenal gland, and submandibular gland at 40 kDa, colinear with the migration of the cloned 11 beta HSD2 enzyme. A band of medium intensity was also seen at this size in the pancreas, whereas a band of moderate intensity was seen in the bile duct, and weaker bands were noticed in the stomach, small intestine, and liver, with a diffuse band at 36-42 kDa in the prostate. Strong bands were seen in the pancreas and prostate at 78 kDa, with weaker signals in the colon, adrenal, stomach, and bile duct. A number of tissues also displayed multiple bands at about 30 kDa. Enzymatic assays on tissue homogenates showed extensive conversion of corticosterone to its 11-dehydro product in an NAD-dependent manner in the submandibular gland, adrenal gland, and kidney, but not in the pancreas or prostate. This study confirms the ubiquitous presence of 11 beta HSD2 in sodium-transporting epithelia, demonstrates the high level of 11 beta HSD2 protein and enzyme activity in the rat adrenal, and suggests a possible role for the enzyme in the biliary system. Further studies are required to determine the relevance of the various molecular species to the activity, latency, and processing of the enzyme.  相似文献

Molybdenum black solar selective coatings have been produced on cobalt by immersion in a solution of ammonium paramolybdate and nickel sulphate. The cobalt was electroplated on nickel-plated copper prior to immersion in the paramolybdate solution. The maximum solar absorptance of the resulting molybdenum black coating was about 0.91. The minimum emittance was about 0.1 for coatings on cobalt deposited with an addition agent and about 0.23 for coatings on cobalt deposited without addition agent in the 60 °C plating solution. These differences have been related to the coating morphology determined by scanning electron microscopy. The emittance of coatings on cobalt deposited without addition agent decreases (or remains unchanged) during short-term heat treatment while that of coatings deposited on cobalt plated with addition agent increases somewhat. Reduction of the cobalt plating bath temperature to 45 °C can also lead to good initial coating properties but without the requirement for an addition agent in the cobalt plating bath. XPS studies show that the oxidation state of molybdenum in the coatings is approximately + 5 corresponding to Mo4O11. This reduces to + 4 after argon ion bombardment. Some cobalt may be present in the coatings in the form of CoO.  相似文献

Experimental tests showed that poststrike elevation in tongue-flicking rate (PETF) and strike-induced chemosensory searching (SICS) in the gila monster last longer than reported for any other lizard. Based on analysis of numbers of tongue-flicks emitted in 5-min intervals, significant PETF was detected in all intervals up to and including minutes 41–45. Using 10-min intervals, PETF lasted though minutes 46–55. Two of eight individuals continued tongue-flicking throughout the 60 min after biting prey, whereas all individuals ceased tongue-flicking in a control condition after minute 35. The apparent presence of PETF lasting at least an hour in some individuals suggests that there may be important individual differences in duration of PETF. PETF and/or SICS are present in all families of autarchoglossan lizards studied except Cordylidae, the only family lacking linguallly mediated prey chemical discrimination. However, its duration is known to be greater than 2-min only in Helodermatidae and Varanidae, the living representatives of Varanoidea. That prolonged PETF and SICS are typical of snakes provides another character supporting a possible a varanoid ancestry for Serpentes. Analysis of 1-min intervals showed that PETF occurred in the first minute. A review of the literature suggests that a pause in tongue-flicking and delay of searching movements are absent in lizards and the few nonvenomous colubrid snakes tested. The delayed onset of SICS may be a specific adaptation of some viperid snakes to allow potentially dangerous prey to be rendered harmless by venom following voluntary release after envenomation and preceding further physical contact with the prey.  相似文献

A two-dimensional horizontal dispersion equation is derived for the many ways that buoyancy modifies the spreading of a mono-disperse discharge of particles in a shallow-water flow. The particle-induced lateral density gradient gives rise to a secondary transverse flow which tends to carry particles outwards. Stratification with the consequent reduced mixing, changes the local vertical profile of the original flow and gives rise to a Burgers nonlinearity (slowing down of sinking particles or a speeding up of buoyant particles). Stratification also modifies the non-local horizontal distribution of the current, with an inflow towards particle-laden regions where the drag is reduced. A simple eddy-diffusivity turbulence model is used which permits explicit evaluation of most of the linear and nonlinear coefficients. Nonlinear effects are shown to be significant for particle volume concentrations of only 10 parts per million.  相似文献

The second order boundary layer effects are considered for the classical problem of the impulsive motion past a flat plate. Although Stewartson [1], [2] showed clearly how the effect of the leading edge requires a finite time to enter the boundary layer when only first order effects are included, we see here how this second order contribution is noticed immediately throughout the layer, and so becomes the dominant contribution after the Rayleigh behaviour during this time.  相似文献
