To evaluate the contributions of common noise sources to total annual noise exposures among urban residents and workers, we estimated exposures associated with five common sources (use of mass transit, occupational and nonoccupational activities, MP3 player and stereo use, and time at home and doing other miscellaneous activities) among a sample of over 4500 individuals in New York City (NYC). We then evaluated the contributions of each source to total noise exposure and also compared our estimated exposures to the recommended 70 dBA annual exposure limit. We found that one in ten transit users had noise exposures in excess of the recommended exposure limit from their transit use alone. When we estimated total annual exposures, 90% of NYC transit users and 87% of nonusers exceeded the recommended limit. MP3 player and stereo use, which represented a small fraction of the total annual hours for each subject on average, was the primary source of exposure among the majority of urban dwellers we evaluated. Our results suggest that the vast majority of urban mass transit riders may be at risk of permanent, irreversible noise-induced hearing loss and that, for many individuals, this risk is driven primarily by exposures other than occupational noise.  相似文献

The application of quantitative real-time PCR (qPCR) technologies for the rapid identification of fecal bacteria in environmental waters is being considered for use as a national water quality metric in the United States. The transition from research tool to a standardized protocol requires information on the reproducibility and sources of variation associated with qPCR methodology across laboratories. This study examines interlaboratory variability in the measurement of enterococci and Bacteroidales concentrations from standardized, spiked, and environmental sources of DNA using the Entero1a and GenBac3 qPCR methods, respectively. Comparisons are based on data generated from eight different research facilities. Special attention was placed on the influence of the DNA isolation step and effect of simplex and multiplex amplification approaches on interlaboratory variability. Results suggest that a crude lysate is sufficient for DNA isolation unless environmental samples contain substances that can inhibit qPCR amplification. No appreciable difference was observed between simplex and multiplex amplification approaches. Overall, interlaboratory variability levels remained low (<10% coefficient of variation) regardless of qPCR protocol.  相似文献

Campylobacter continues to be one of the bacterial pathogens most frequently associated with human gastrointestinal illness worldwide. Because Campylobacter primarily colonizes the intestines of animals used for food production, food products of animal origin can become contaminated with this pathogen and thus represent a significant risk factor. Despite application of numerous physical and chemical interventions to control Campylobacter during food processing, the high isolation rate of this pathogen from some retail meat products indicates that Campylobacter is able to persist from animal slaughterhouses through food systems. Given the fastidious growth requirements and high susceptibility of this pathogen to environmental conditions, the ability of Campylobacter to attach to food and food-related surfaces is likely to play an important role in food contamination and movement through food systems. This review was compiled to (i) describe how the attachment of Campylobacter to surfaces influences the prevalence and survival of the organism through food systems, (ii) examine the potential factors affecting the ability of Campylobacter to attach to surfaces, and (iii) suggest strategies for controlling this attachment process.  相似文献

The effects of different salt concentrations (6%, 15%, 18% and 24% NaCl (w/w)) on the conformational changes of cod muscle proteins during brine salting were examined in this study. Proteins were extracted from the brine salted samples with solutions of 1 M (5.9%) and 4 M (23.4%) NaCl and the quantity of salt soluble proteins (SSP), disulfide bond (S–S), total sulfhydryl (SH) and available SH content in the soluble fraction were determined. Increased salt concentration in cod muscle promoted protein denaturation and aggregation. The SSP and total SH content decreased, whereas the S–S bond and available SH content increased with increased salt concentration in cod muscle. Disulfide bond formation correlated (r = −0.6) with a decrease in total SH groups. Higher SSP and available SH groups of the samples at lower brine concentrations was explained by smaller concentration gradients and salt diffusion rates, resulting in stronger salting-in at early stages of the brining process. There was a significant difference in conformational changes in proteins extracted with a salt solution of 1 and 4 M, mainly due to a different degree of protein aggregation.  相似文献

In the present study, a biomimetic nanoconstruct (BNc) with a multimodal imaging system is engineered using tumor homing natural killer cell membrane (NKM), near‐infrared (NIR) fluorescent dye, and gadolinium (Gd) conjugate‐based magnetic resonance imaging contrast agent onto the surface of a polymeric nanoparticle. The engineered BNc is 110 ± 20 nm in size and showed successful retention of NKM proteins. The magnetic properties of the BNc are found to be tunable from 2.1 ± 0.17 to 5.3 ± 0.5 mm ?1 s?1 under 14.1 T, by adjusting the concentration of Gd‐lipid conjugate onto the surface of the BNc. Confocal imaging and cell sorting analysis reveal a distinguishable cellular interaction of the BNc with MCF‐7 cells in comparison to that of bare polymeric nanoparticles suggesting the tumor homing properties of NKM camouflage system. The in vitro cellular interaction results are further confirmed by in vivo NIR fluorescent tumor imaging and ex vivo MR imaging, respectively. Pharmacokinetics and biodistribution analysis of the BNc show longer circulation half‐life (≈9.5 h) and higher tumor accumulation (10% of injected dose) in MCF‐7 induced tumor‐bearing immunodeficient NU/NU nude mice. Owing to the proven immunosurveillance potential of NK‐cell in the field of immunotherapy, the BNc engineered herein would hold promises in the design consideration of nanomedicine engineering.  相似文献

In this paper the practical density of long‐term DNA storage is increased. Specifically, the DNA weight loading of silica sphere DNA storage is increased to 3.4 wt%, a ten‐fold increase compared to the previous state‐of‐the‐art. By applying a Layer‐by‐Layer (LbL) design with alternating layers of DNA and a polycationic molecule, namely polyethyleneimine (PEI), another dimension to DNA surface binding onto magnetic nanoparticles is added. A protective silica layer is grown on top of the multilayered nanoparticles to shield the DNA from external sources of damage. Accelerated aging experiments of the nanoparticles and the subsequent quantification of DNA stability via qPCR show a significantly lower degradation rate compared to unprotected DNA. The novel material is compared to previous DNA storage technologies, outperforming those in DNA storage density as well as stability. Finally, the storage of an 83 kB digital file in DNA through a successful readout of a 4991 oligonucleotide pool is demonstrated from particle encapsulation, through accelerated aging, to sequencing.  相似文献

This paper presents a design of a comb finger shape and calculation of a trapezoidal-shaped electrostatic comb-drive actuator (TECA) in order to aim a higher electrostatic force density and larger displacement in comparison with the typical rectangular-shaped electrostatic comb-drive actuator (RECA). Relation between a beam’s stiffness and a driving voltage has been examined to predict a pull-in effect occurring in TECA. Micro fabrication and characterization of TECA and RECA systems are performed by using a standard SOI-MEMS technology. Theoretical and experimental results confirm the strong points of TECA’s structure (similar to the dimensions of RECA) such as a larger number of movable comb finger arrayed at the same length and larger displacement. At driving voltages of 47.9 and 50 (V), the calculation and measurement displacement of TECA are approximately 2.2 and 1.78 times larger than that of RECA, respectively.
