On the basis of the executive-attention theory of working memory capacity (WMC; e.g., M. J. Kane, A. R. A. Conway, D. Z. Hambrick, & R. W. Engle, 2007), the authors tested the relations among WMC, mind wandering, and goal neglect in a sustained attention to response task (SART; a go/no-go task). In 3 SART versions, making conceptual versus perceptual processing demands, subjects periodically indicated their thought content when probed following rare no-go targets. SART processing demands did not affect mind-wandering rates, but mind-wandering rates varied with WMC and predicted goal-neglect errors in the task; furthermore, mind-wandering rates partially mediated the WMC-SART relation, indicating that WMC-related differences in goal neglect were due, in part, to variation in the control of conscious thought. Nearly half a billion dollars in resources are lost each time a drug candidate is withdrawn from the market by the Food and Drug Administration (FDA) for reasons of liver toxicity. The number of late-phase drug developmental failures due to liver toxicity could potentially be reduced through the use of hepatocyte-based systems capable of modeling the response of in vivo liver tissue to toxic insults. With this article, we report progress toward the goal of realizing an array of primary hepatocytes for use in high-throughput liver toxicity studies. Described herein is the development of a 64 (8 x 8) element array of microfluidic wells capable of supporting micropatterned primary rat hepatocytes in coculture with 3T3-J2 fibroblasts. Each of the wells within the array was continuously perfused with medium and oxygen in a nonaddressable format. The key features of the system design and fabrication are described, including the use of two microfluidic perfusion networks to provide the coculture with an independent and continuous supply of cell culture medium and oxygen. Also described are the fabrication techniques used to selectively pattern hepatocytes and 3T3-J2 fibroblasts within the wells of the array. The functional studies used to demonstrate the synthetic and metabolic capacity of the array are outlined in this article. These studies demonstrate that the hepatocytes contained within the array are capable of continuous, steady-state albumin synthesis (78.4 microg/day, sigma = 3.98 microg/day, N = 8) and urea production (109.8 microg/day, sigma = 11.9 microg/day, N = 8). In the final section of the article, these results are discussed as they relate to the final goal of this research effort, the development of an array of primary hepatocytes for use in physiologically relevant toxicology studies.  相似文献

Aerosol mass spectrometers can be used to classify individual airborne particles on the basis of chemical composition. While positive ion mass spectra are normally used to characterize ultrafine particles (defined here as particles smaller than 200 nm in diameter), negative ion mass spectra can provide complementary information. To effectively utilize the negative ion mass spectra of ultrafine particles, it is important to understand biases in the formation and detection of negative ions. It is found that the intensity of negative ions is generally less than that of positive ions, due to the creation of electrons in the ablation process that must react to form negative ions. The ablation efficiency, defined as the probability that an ablated particle produces a detectable ion signal, exhibits both size and composition dependencies. The ablation efficiency for detection of negative ions follows the same trends as the ablation efficiency for the detection of positive ions: sodium chloride and ammonium nitrate have higher ablation efficiencies than oleic acid, and the ablation efficiency decreases with the particle diameter. The ablation efficiency of negative ions is less than or equal to the ablation efficiency of positive ions, and the relative difference increases as the particle diameter decreases. Pure ammonium sulfate particles exhibit an ablation efficiency too low to be measured in the present experiments. However, trace amounts of sulfate in mixed-composition particles can be readily detected in the negative ion mass spectra.  相似文献

We investigate the signal-to-noise ratio (SNR) for a bistatic coherent laser radar (CLR) system. With a bistatic configuration, the spatial resolution is determined by the overlap of the transmit beam and the virtual backpropagated local oscillator beam. This eliminates the trade-off between range resolution and the bandwidth of the transmitted pulse inherent in monostatic systems. The presented analysis is completely general in that the expressions can be applied to both monostatic and bistatic CLR systems. The heterodyne SNR is computed under the assumption of untruncated Gaussian optics and untruncated Gaussian beam profiles. The analysis also includes the effects of refractive turbulence. The results show that, for maximum SNR, small transmit and local oscillator beam profiles (e-1 intensity radius) are desired.  相似文献

Highly stereo-selective hydrogenation of trans-1,3-pentadiene to cis-2-pentene and isomerization of 1,4-pentadiene to cis-1,3-pentadiene were found to take place thermally at 423 K over Mo(CO)6/NaY. Relative hydrogenation rates of pentadiene isomers indicated that s-cis conformation of diene was crucial for the selective hydrogenation to occur. This was substantiated by an IR study.  相似文献

The computational advantages associated with the utilization of preconditioned iterative equation solvers are quantified for the reanalysis of perturbed shapes using continuum structural boundary element analysis (BEA). Both single- and multizone three-dimensional problems are examined. Significant redutions in computer time are obtained by making use of previously computed solution vectors and preconditioners in subsequent analyses. The effectiveness of this technique is demonstrated for the computation of shape response sensitivities required in shape optimization. Computer times and accuracies achieved using the preconditioned iterative solvers are compared with those obtained via direct solvers and implicit differentiation of the boundary integral equations. It is concluded that this approach employing preconditioned iterative equation solvers in reanalysis and sensitivity analysis can be competitive with if not superior to those involving direct solvers.  相似文献

Activation of the phosphatidylinositol (PtdIns) signal transduction system involves stimulation of phospholipase C (PLC) by hormones and other agonists to produce two second messengers, the inositol phosphate, Ins(1,4,5)P3 which releases calcium from intracellular stores, and diacylglycerol which activates protein kinase C (PKC). This study, using activators or inhibitors of PLC and PKC and a calcium ionophore, examined the role of the PtdIns system in mouse embryonic stem (ES) cells. The PLC inhibitor, U-73122, inhibited ES-cell proliferation and also inhibited PLC activation as evidenced by a decrease in inositol phosphate formation in response to fetal calf serum stimulation. The two PKC activators, the diacylglycerol analogue 1,2, dioctanoyl-sn-glycerol (DOG) and the phorbol ester 12-O-tetra-decanoyl phorbol 13-acetate (TPA), increased cell proliferation in a dose-dependent manner, as did the calcium ionophore, ionomycin. However, co-stimulation with either ionomycin and DOG or ionomycin and TPA resulted in a reduced number of cells. The PKC inhibitor, bisindolylmaleimide II (Bis II), significantly decreased the number of ES cells, mainly due to increased apoptosis. The possible feedback effect of PKC on PLC was examined by preincubating ES cells with either the PKC inhibitor Bis II or the activator TPA before stimulation of inositol phosphate production with fetal calf serum; preincubation with Bis II increased inositol phosphate formation whereas preincubation with TPA decreased inositol formation. These results indicate that the PtdIns system is involved in the control of ES-cell proliferation and apoptosis.  相似文献
