This case study uncovers a turning point in the production of play space in Amsterdam. Whereas over the first half of the twentieth century the creation of play spaces used to be the primary responsibility of the Amsterdam civil society, this situation started to change after the Second World War. Between 1947 and 1970, the Amsterdam Urban Planning Department created over 700 public play spaces. These spaces were little niches in the urban public domain, specifically designed and constructed to enable city children's play. This remarkable change from a predominantly private to a public intervention, is explained through a rapid increase of the number of children (the post-war baby-boom), the existence of the General Extension Plan (AUP) with its detailed age specific approach and the fruitful collaboration between powerful urban planners and politically dominant socialist politicians.  相似文献

The gallium gradient in Cu(In,Ga)Se2 (CIGS) layers, which forms during the two industrially relevant deposition routes, the sequential and co‐evaporation processes, plays a key role in the device performance of CIGS thin‐film modules. In this contribution, we present a comprehensive study on the formation, nature, and consequences of gallium gradients in CIGS solar cells. The formation of gallium gradients is analyzed in real time during a rapid selenization process by in situ X‐ray measurements. In addition, the gallium grading of a CIGS layer grown with an in‐line co‐evaporation process is analyzed by means of depth profiling with mass spectrometry. This gallium gradient of a real solar cell served as input data for device simulations. Depth‐dependent occurrence of lateral inhomogeneities on the µm scale in CIGS deposited by the co‐evaporation process was investigated by highly spatially resolved luminescence measurements on etched CIGS samples, which revealed a dependence of the optical bandgap, the quasi‐Fermi level splitting, transition levels, and the vertical gallium gradient. Transmission electron microscopy analyses of CIGS cross‐sections point to a difference in gallium content in the near surface region of neighboring grains. Migration barriers for a copper‐vacancy‐mediated indium and gallium diffusion in CuInSe2 and CuGaSe2 were calculated using density functional theory. The migration barrier for the InCu antisite in CuGaSe2 is significantly lower compared with the GaCu antisite in CuInSe2, which is in accordance with the experimentally observed Ga gradients in CIGS layers grown by co‐evaporation and selenization processes. Epidermal growth factor receptor (EGFR) is a validated tumor marker overexpressed in various cancers such as squamous cell carcinoma (SSC) of the head and neck and gliomas. We constructed protein-drug conjugates based on the anti-EGFR Designed Ankyrin Repeat Protein (DARPin) E01, and compared the bivalent DARPin dimer (DD1) and a DARPin-Fc (DFc) to the monomeric DARPin (DM) and the antibody derived scFv425-Fc (scFvFc) in cell culture and a mouse model. The modular conjugation system, which was successfully applied for the preparation of protein-drug and -dye conjugates, uses bio-orthogonal protein-aldehyde generation by the formylglycine-generating enzyme (FGE). The generated carbonyl moiety is addressed by a bifunctional linker with a pyrazolone for a tandem Knoevenagel reaction and an azide for strain-promoted azide-alkyne cycloaddition (SPAAC). The latter reaction with a PEGylated linker containing a dibenzocyclooctyne (DBCO) for SPAAC and monomethyl auristatin E (MMAE) as the toxin provided the stable conjugates DD1-MMAE (drug-antibody ratio, DAR = 2.0) and DFc-MMAE (DAR = 4.0) with sub-nanomolar cytotoxicity against the human squamous carcinoma derived A431 cells. In vivo imaging of Alexa Fluor 647-dye conjugates in A431-xenografted mice bearing subcutaneous tumors as the SCC model revealed unspecific binding of bivalent DARPins to the ubiquitously expressed EGFR. Tumor-targeting was verified 6 h post-injection solely for DD1 and scFvFc. The total of four administrations of 6.5 mg/kg DD1-MMAE or DFc-MMAE twice weekly did not cause any sequela in mice. MMAE conjugates showed no significant anti-tumor efficacy in vivo, but a trend towards increased necrotic areas (p = 0.2213) was observed for the DD1-MMAE (n = 5).  相似文献

The static distribution of work among tasks is not possible in many parallel applications. Therefore, it is essential to implement convenient and efficient abstractions for ‘work sharing’ on multicomputers. This paper compares the utility of two operating system facilities for the implementation of such ‘work sharing’: (1) a system for the migration of processes from heavily to less loaded processors and (2) a more general OS construct for the implementation of arbitrary distributed objects. Both were implemented as extensions to the Intel iPSC/1 operating system on a 32-node hypercube. Their experimental evaluation is based on a parallel implementation of a branch-and-bound algorithm. Two sets of results are attained. First, the necessity of the constructs for dynamic work sharing is demonstrated for applications with dynamic data domains, such as parallel branch-and-bound algorithms. This is followed by measurements that demonstrate the acceptable cost of process migration for a specific parallel branch-and-bound algorithm. These measurements are then compared with results attained with the construct for the implementation of distributed objects. Second, when using branch-and-bound to solve the Travelling Salesperson Problem (TSP), evaluation of the resulting parallel TSP program shows that some analytical and simulation results attained in past, published work may not hold.  相似文献

Thanks to their high energy density and thermal conductivity, metallic Phase Change Materials (mPCM) have shown great potential to improve the performance of thermal energy storage systems. However, the commercial application of mPCM is still limited due to their corrosion behavior with conventional container materials. This work first addresses on a fundamental level, whether carbon-based composite-ceramics are suitable for corrosion critical components in a thermal storage system. The compatibility between the mPCM AlSi12 and the Liquid Silicon Infiltration (LSI)-based carbon fiber reinforced silicon carbide (C/C-SiC) composite is then investigated via contact angle measurements, microstructure analysis, and mechanical testing after exposure. The results reveal that the C/C-SiC composite maintains its mechanical properties and microstructure after exposure in the strongly corrosive mPCM. Based on these results, efforts were made to design and manufacture a container out of C/C-SiC for the housing of mPCM in vehicle application. The stability of the component filled with mPCM was proven nondestructively via computer tomography (CT). Successful thermal input- and output as well as thermal storage ability were demonstrated using a system calorimeter under conditions similar to the application. The investigated C/C-SiC composite has significant application potential as a structural material for thermal energy storage systems with mPCM.  相似文献

During summer stratification a strip-mine lake in which fish were cultured in floating cages developed black water in its anoxic zone. The blackness was evidently produced by ferrous sulfide. Two types of black water stratification were observed. In one the blackness occurred throughout the anoxic zone but was more intense in the upper part, while iron and alkalinity were high throughout this zone. In the second the black water was confined to a meter-thick stratum within the anoxic zone, and iron and alkalinity were remarkably low except in two strata: the one with black water, and the one at the top of the anoxic zone.It is suggested that both types of stratification were caused by a high rate of sulfide production in the metalimnion. The relatively high temperature there (as compared to the hypolimnion) and the presumably higher concentrations of organic matter and ammonia, resulting from fish food and fish wastes, would account for this high rate of sulfide production. Because of sharp oxygen stratification two carbonate-forming processes, photosynthesis and sulfate reduction, occurred in close proximity to each other. In especially calm weather vertical density currents could be initiated by the calcium carbonate crystals and produce the second type of stratification. If the second type of stratification could be produced at will it might be used to improve the quality of strip-mine lake water.  相似文献

Summary: Coupling reactions between terminal functionalized polymer chains were chosen for the synthesis of star‐like polymers consisting of polystyrene and polystyrene‐block‐poly[styrene‐co‐(butyl acrylate)] arms. For the preparation of terminal functionalized polymer chains a side reaction of the 2,2,6,6‐tetramethylpiperidine‐N‐oxyl (TEMPO) mediated free radical polymerization of methacrylates could be used successfully to convert TEMPO terminated polymers into end functionalized polymers. The number of functionalized monomer units attached to the polymer chain is directly related to the TEMPO concentration during this reaction. Different polystyrenes and polystyrene‐block‐poly[styrene‐co‐(butyl acrylate)] block copolymers were functionalized with a variable number of epoxide and alcohol groups at the chain end. For the determination of the optimal reaction parameters for the coupling reactions between these polymer chains, epoxy functionalized polystyrenes were converted with hydroxy functionalized polystyrenes under basic and acidic conditions. By activation with sodium hydride or boron trifluoride star‐like polymers were synthesized under mild conditions. The transfer of the reaction conditions to coupling reactions between end functionalized polystyrene‐block‐poly[styrene‐co‐(butyl acrylate)] copolymers showed that star‐like polymers with more than 12 arms were formed using boron trifluoride as activating agent.

Cavitation and superstructure evolution of polymers during stretching play crucial roles to influence the mechanical properties of materials. In this study, we investigated deformation-mediated superstructures and cavitation of poly (l-lactide) (PLA) as well as their dependence on stretching temperatures by in-situ small-angle X-ray (SAXS) analysis coupled with mechanical testing. It is found that the cavitation and crystalline deformation are strongly influenced by stretching stress during deformation, which significantly depends on the stretching temperature. At lower stretching temperature (70 °C), the cavitation is initiated before the yielding and then stimulates the crystallite shearing. At higher stretching temperature (90 °C), however, the crystallites shear firstly and then crystalline deformation promotes the formation of cavities orientated along the stretching direction. High stretching temperature benefits the formation of relatively perfect crystals with high orientation. The results provide the basic knowledge of how to adjust the mechanical properties of polymer materials by controlling their superstructure in the deformation process.  相似文献

Visible/near-infrared spectroscopy has been evaluated for use in freshness prediction and frozen-thawed classification of farmed Atlantic salmon fillets, where fresh samples were stored as whole fish in ice. A handheld interactance probe for performing rapid measurements of single fillets and an imaging spectrometer for online analysis at an industrial speed of one fillet per second, have been used. Freshness as storage days in ice is predicted with an accuracy of 2.4 days for individual fillets, whereas frozen-thawed salmon fillets are completely separated from fresh fillets. The prediction results are comparable to previous results using the Quality Index Method with trained panelists. The region between 605 and 735 nm, which excludes interference by carotenoids and water, is appropriate for both frozen-thawed classification and freshness prediction of salmon fillets. The results indicate that the spectral changes are explained mainly by oxidation of heme proteins during the freeze–thaw cycle and during chilled storage in ice.  相似文献

Staphylococcus aureus remains a leading cause of food-poisoning with substantial impact on public health. Using a multiplex polymerase chain reaction-DNA enzyme immunoassay (PCR-DEIA), we studied the presence of genes encoding staphylococcal enterotoxin-like (SEl) superantigens sem, sen, and seo, associated with the enterotoxin gene cluster (egc), in 429 clinical Staphylococcus aureus isolates. 294 (68.5%) isolates tested positive for at least one of the three SEl genes. In contrast to the fixed gene combination seg/sei also located on egc, a substantial number of isolates (n = 108) were found to bear only one or two of the genes encoding SElM, SElN, and SElO. Regarding the origin of the S. aureus isolates, a significant difference (P = 0.022) was found for the possession of seo (61.2% of blood isolates versus 42.9% of nasal strains). Also sem (not significantly) was found more common in blood isolates (52.1% versus 40.5%). The survey of the newly described SEl genes sem-seo supports the concept that most clinical S. aureus isolates harbor subsets of pyrogenic toxin superantigens. The potential contribution of seo and sem to the pathogenic potential of S. aureus has to be further evaluated.  相似文献
