The gutless hydrothermal tubeworm Riftia pachyptila Jones relies mainly on its chemoautotrophic bacterial symbionts to supply nutrients in the form of secreted organic compounds resulting from fixation and incorporation of CO2. In this study, symbionts were purified, tested for viability, and incubated in the presence of labeled CO2. We demonstrated that purified symbionts can be used as a viable alternative to experiments with bacterial cultures. Several organic acids, sugars, and amino acids were labeled, but their fraction of the total label stayed generally constant during the incubation times used. However, increasing fractions of succinate and, to a lesser degree, glutamate were excreted into the incubation medium, indicating that these are probably the main carbon-containing compounds transferred from the symbionts to the host. Glutamate could also account for the transport of nitrogen from the symbionts to the host.  相似文献

The authors investigated the role of homosexual arousal in exclusively heterosexual men who admitted negative affect toward homosexual individuals. Participants consisted of a group of homophobic men (n?=?35 ) and a group of nonhomophobic men (n?=?29); they were assigned to groups on the basis of their scores on the Index of Homophobia (W. W. Hudson & W. A. Ricketts, 1980). The men were exposed to sexually explicit erotic stimuli consisting of heterosexual, male homosexual, and lesbian videotapes, and changes in penile circumference were monitored. They also completed an Aggression Questionnaire (A. H. Buss & M. Perry, 1992). Both groups exhibited increases in penile circumference to the heterosexual and female homosexual videos. Only the homophobic men showed an increase in penile erection to male homosexual stimuli. The groups did not differ in aggression. Large collaborative centers are a common model for accomplishing integrated environmental health research. These centers often include various types of scientific domains (e.g., chemistry, biology, bioinformatics) that are integrated to solve some of the nation's key economic or public health concerns. The Superfund Research Center (SRP) at Oregon State University (OSU) is one such center established in 2008 to study the emerging health risks of polycyclic aromatic hydrocarbons while using new technologies both in the field and laboratory. With outside collaboration at remote institutions, success for the center as a whole depends on the ability to effectively integrate data across all research projects and support cores. Therefore, the OSU SRP center developed a system that integrates environmental monitoring data with analytical chemistry data and downstream bioinformatics and statistics to enable complete "source-to-outcome" data modeling and information management. This article describes the development of this integrated information management system that includes commercial software for operational laboratory management and sample management in addition to open-source custom-built software for bioinformatics and experimental data management.  相似文献

The relationship between protein motions (i.e., dynamics) and enzymatic function has begun to be explored in β-lactamases as a way to advance our understanding of these proteins. In a recent study, we analyzed the dynamic profiles of TEM-1 (a ubiquitous class A β-lactamase) and several ancestrally reconstructed homologues. A chief finding of this work was that rigid residues that were allosterically coupled to the active site appeared to have profound effects on enzyme function, even when separated from the active site by many angstroms. In the present work, our aim was to further explore the implications of protein dynamics on β-lactamase function by altering the dynamic profile of TEM-1 using computational protein design methods. The Rosetta software suite was used to mutate amino acids surrounding either rigid residues that are highly coupled to the active site or to flexible residues with no apparent communication with the active site. Experimental characterization of ten designed proteins indicated that alteration of residues surrounding rigid, highly coupled residues, substantially affected both enzymatic activity and stability; in contrast, native-like activities and stabilities were maintained when flexible, uncoupled residues, were targeted. Our results provide additional insight into the structure-function relationship present in the TEM family of β-lactamases. Furthermore, the integration of computational protein design methods with analyses of protein dynamics represents a general approach that could be used to extend our understanding of the relationship between dynamics and function in other enzyme classes.  相似文献

Synchrotron-based techniques (high-resolution photoemission, in-situ X-ray absorption spectroscopy, and time-resolved X-ray diffraction) have been used to study the destruction of SO2 and the water-gas shift (WGS, CO + H2O → H2 + CO2) reaction on a series of gold/ceria systems. The adsorption and chemistry of SO2 was investigated on Au/CeO2(111) and AuO x /CeO2 surfaces. The heat of adsorption of the molecule on Au nanoparticles supported on stoichiometric CeO2(111) was 4–7 kcal/mol larger than on Au(111). However, there was negligible dissociation of SO2 on the Au/CeO2(111) surfaces. The full decomposition of SO2 was observed only after introducing O vacancies in the ceria support. AuO x /CeO2 surfaces were found to be much less chemically active than Au/CeO2(111) or Au/CeO2−x (111) surfaces. In a separate set of experiments, in-situ time-resolved X-ray diffraction and X-ray absorption spectroscopy were used to monitor the behavior of nanostructured {Au + AuO x }–CeO2 catalysts under the WGS reaction. At temperatures above 250 °C, a complete AuO x → Au transformation was observed with high catalytic activity. Photoemission results for the oxidation and reduction of Au nanoparticles supported on rough ceria films or a CeO2(111) single crystal corroborate that cationic Auδ+ species cannot be the key sites responsible for the WGS activity at high temperatures. The active sites in {Au + AuO x }/ceria catalysts should involve pure gold nanoparticles in contact with O vacancies of the oxide.  相似文献

Experiments were conducted to (1) determine whether the electroantennogram (EAG) can detect differences among the responses of antennae from males derived from the three strains ofOstrinia nubilalis (Hübner), and (2) characterize the EAG responses of each strain to isomeric forms of the natural pheromone, (E)- and (Z)-11-tetradecen-1-ol acetate (TDA), and analogs possessing differences in the terminal alkyl group, cyclopropyl (CPA), ortert-butyl (TBA).EAG responses differed among the strains in two ways: (1) Antennae fromZZ males always produced an EAG to (Z)-TDA with an extended duration of response. This signature EAG response was found to be unique to the antennal response ofZZ males to (Z)-TDA, thus providing a relatively easy method of distinguishing liveZZ males fromEE orZE males. Correlated with this longer EAG response was a longer disadaptation time, i.e., the EAG response ofZZ antennae disadapted more slowly (ca. 10 min) than the response ofEE antennae. (2) Strain differences in the relative EAG amplitudes to isomers and analogs were observed at the stimulus amounts eliciting the peak EAG amplitude as follows: TDA CPA > TBA forZZ males and both isomers; TDA > CPA TBA and CPA TDA > TBA forEE males and theE andZ isomers, respectively; CPA > TBA TDA forZE males and both isomers. Dose—response relationships were seen for all compounds if amplitude (peak height) of the EAG was used as a measure of response. However, if width of the EAG at half the peak height (peak width) was used, then only theZZ antennal response to (Z)-TDA resulted in a meaningful dose-response relationship. For all strains, the EAG amplitudes elicited by theZ isomers of any of the tested compounds were greater than those elicited by the correspondingE isomers. Therefore, correlations between the relative EAG and upwind flight responses were observed in theZZ (r = 0.86) andZE (r = 0.80) strains but were not correlated in theEE strain (r = 0.18). Temporal studies showed that adaptation, not postexcision deterioration, was responsible for the observed decreases in the EAG amplitude after repetitive stimulation or after stimulation with amounts in a descending order. Disa-daptation required at least 20 min for a moderate dose (10 g for 1 sec). Developmental studies showed that antennae from 2-day-old adults had the greatest EAG response.  相似文献

This paper describes the design of a piezoelectric actuated cutting tool and implementation of digital servo controls for machining surfaces with dynamically varying depth of cut. Through a flexure hinge, the tool holder could generate 50 μm travel at the tip of the cutting insert. Tool motion errors of less than 0.5 μm were achieved in tracking cyclic waveforms by employing a digital repetitive servo control. When applied to turning aluminum and steel workpieces with variable depth of cut using carbide tools, less than 5 μm machined surface errors were measured.  相似文献

Questions exist over the extent to which social media content may bypass, follow, or attract the attention of traditional media. This study sheds light on such dynamics by examining intermedia agenda‐setting effects among the Twitter feeds of the 2012 presidential primary candidates, Twitter feeds of the Republican and Democratic parties, and articles published in the nation's top newspapers. Daily issue frequencies within media were analyzed using time series analysis. A symbiotic relationship was found between agendas in Twitter posts and traditional news, with varying levels of intensity and differential time lags by issue. While traditional media follow candidates on certain topics, on others they are able to predict the political agenda on Twitter.  相似文献

The digital revolution has changed the daily work of archivists. As a result, archivists have refined their analog practices and initiated new ones to accommodate the unique information creation and access needs of users. While limitless storage and technological obsolescence may be surmountable challenges, appraisal continues to be a necessary component of this preservation enterprise. Macro-appraisal—a top-down functional assessment of an organization’s high-level decision making that defines the enduring value of specific bodies of documentary evidence of these creative or administrative processes—is used to facilitate the management of diverse bodies of born-digital records in archives and special collections.  相似文献

Adherence to accessibility guidelines for Web pages does not necessarily guarantee a usable or satisfying Web experience for all persons with disabilities. The needs of many of these individuals fall outside the guidelines for accessible content that most Web authors take into consideration. Many of these users wish, for example, that they simply could ‘enlarge’ what is on a Web page. They also express the wish that pages would be ‘less confusing’. To meet these needs, Web browsers and various software applications provide for a variety of ways in which page presentations can be altered. The effects of these alterations often have unexpected consequences. Some designs accommodate these alterations better than others. This article discusses one such application that allows users to control features of Web page presentation and explores design features that facilitate such control.  相似文献
