CD complex response to cessation of induction with OKT3 may be predictive of rejection. Twenty-seven patients receiving renal allografts and OKT3 induction immunosuppression were retrospectively analyzed for CD complex repopulation and allograft rejection. Flow cytometric monitoring was utilized in all patients. Responder status groups were identified based on CD complex repopulation, with fast responders demonstrating CD complex repopulation above the determined cohort mean. Slow responders had repopulation below this mean. Student's t test yielded P<0.01 (CD2), P<0.02 (CD3), and P<0.01 (CD8). Nonresponder patients were identified with repopulation below the mean, but flat compared with depletion. All nine fast responders lost their graft or were treated for rejection. No slow responder experienced graft loss or rejection episodes. One nonresponder was treated for rejection. CD complex activity following OKT3 cessation correlates with future rejection. Identification of responder status provides insight into propensity to reject, allowing individual tailoring of immunosuppression to patient response.  相似文献

Affinity separation can be achieved by batch and fixed bed modes. Mathematical models including film mass transfer, intraparticle diffusion, and reversible reaction are formulated for both fixed bed and batch adsorption processes. Orthogonal collocation method is used to numerically evaluate the performance of batch and fixed bed adsorption. The efficiencies are evaluated in terms of both the solute recovery and adsorbent utilization. The effect of the following parameters is simulated for the comparison purpose: solute concentration, reaction kinetics, ligand content, and particle size. U937 cell growth in the presence of either chloramphenicol or ethidium bromide rapidly leads to respiratory deficiency. The novel finding of this report is that this response is paralleled by a specific increase in Se-dependent and independent glutathione peroxidase activities as well as of glutathione peroxidase and heme oxygenase mRNAs. Under the same experimental conditions, catalase activity and catalase mRNA do not show appreciable changes. These results can be explained by an increased formation of H2O2 at the early times of development of respiratory deficiency followed by induction of antioxidant enzymes.  相似文献

The use of a high-resistivity substrate extends the capability of standard digital CMOS technology to enable the integration of high-performance RF passive components. The impact of substrate resistivity on the key components of RF CMOS for system-on-chip (SoC) applications is discussed. The comparison includes the transistor, transmission line, inductor, capacitor and varactor, as well as the noise isolation. We also discuss the integration issues including latch-up and well-well isolation in a 0.35-/spl mu/m Cu metal pitch, 0.1-/spl mu/m-gate-length RF CMOS technology.  相似文献

A computational model is developed for the parametric investigation of single‐effect and series flow double‐effect LiBr/H2O absorption refrigeration systems. The effects of generator, absorber, condenser, evaporator and dead state temperatures are examined on the performance of these systems. The parameters computed are coefficient of performance (COP), exergy destruction rates, thermal exergy loss rates, irreversibility and exergetic efficiency. The results indicate that COP and exergetic efficiency of both the systems increase with increase in the generator temperature. There exist different optimum values of generator temperature for maximum COP and maximum exergetic efficiency. The optimum generator temperature is lower corresponding to maximum exergetic efficiency as compared to optimum generator temperature corresponding to maximum COP. The effect of increase in absorber, condenser and evaporator temperatures is to decrease the exergetic efficiency of both the systems. The irreversibility is highest in absorber in both systems. The peritectic transition, where δ -ferrite and liquid transform into γ -austenite, is an important aspect of product quality and process control in many ferrous and non-ferrous alloys. Due to the high temperatures at which the transition takes place, relatively little experimental work has been carried out to quantify the underlying phenomena. This paper presents recent work in visualizing the peritectic reaction stage of the transition. The visualization was carried out on the surface of hyperperitectic Fe-4.7 wt% Ni samples using a Confocal Scanning Laser Microscope (CSLM) equipped with a gold-image furnace. The focus of the research is to quantify the rate and shape evolution of the γ -austenite phase as it envelopes the δ -ferrite grains. Tip radii of 8 and 5 μ m were witnessed and it was found that, as these tips approached one-another their shape was altered and their growth rates decreased due to soft impingement of their respective diffusion fields. Comparing the separation distance at the point of soft impingement with the tip radius suggested a proportionality constant between tip radius of curvature and diffusion field size of between 1.1 and 1.3.  相似文献

Sarcosine dehydrogenase (SarDH) is a mitochondrial flavoenzyme involved in the oxidative degradation of choline to glycine. The absence of SarDH activity in humans is genetically transmitted and is the cause of an amino acid metabolism disorder called sarcosinemia. Tryptic fragments of the purified enzyme from rat liver were subjected to Edman degradation and the sequences obtained were used to clone the cDNA encoding the full length protein. The deduced amino acid sequence of SarDH shares an overall similarity of 47% with dimethylglycine dehydrogenase (Me2GlyDH), another flavoenzyme involved in the mitochondrial choline catabolism with a similar FAD-binding domain. Covalent binding of FAD to SarDH was demonstrated by the observation of strong fluorescence at 530 nm under excitation at 450 nm of the enzyme immunoprecipitated under denaturing conditions from liver extracts. The localization of SarDH immunoreactivity in the mitochondrial matrix was confirmed by Western-blot analysis of purified mitochondrial fractions. Finally, the tissue distribution of SarDH was investigated by Northern-blot analysis of total RNA and Western-blot analysis of total protein from several rat tissues. A strong expression in the liver, but also in the lung, pancreas, kidney, thymus, and oviduct was observed. We therefore suggest that the enzymes of the choline catabolism pathway are important also for metabolism in nonhepatic tissues.  相似文献

Immunization of proven fertile adult male monkeys (n=3) with a recombinant FSH receptor protein preparation (oFSHR-P) (representing amino acids 1-134 of the extracellular domain of the receptor Mr approximately 15KDa) resulted in production of receptor blocking antibodies. The ability of the antibody to bind a particulate FSH receptor preparation and receptors in intact granulosa cells was markedly (by 30-80%) inhibited by FSH. Serum T levels and LH receptor function following immunization remained unchanged. The immunized monkeys showed a 50% reduction (p<0.001) in transformation of spermatogonia(2C) to primary spermatocytes (4C) as determined by flow cytometry and the 4C:2C ratio showed a correlative change (R 0.81, p<0.0007) with reduction in fertility index (sperm counts X motility score). Breeding studies indicated that monkeys became infertile between 242-368 days of immunization when the fertility index was in the range of 123+/-76 to 354+/-42 (compared to a value of 1602+/-384 on day 0). As the effects observed are near identical to that seen following immunization with FSH it is suggestive that oFSHR-P can substitute for FSH in the development of a contraceptive vaccine.  相似文献

In an increasingly noisy society, methods of reducing noise are becoming more important. This work proposes an analog electroacoustic circuit for active control of narrow-band low-frequency acoustic noise using adaptive filtering techniques. The circuit aims at producing antinoise, which is acoustically added to the disturbing noise to produce an error signal that is fed back to the circuit. The proposed circuit is a modified Kerwin-Huelsman-Newcomb biquad filter that tunes itself to the incoming noise frequency using the zero tuning techniques. The circuit was implemented on a printed circuit board and it was successful in reducing noise by 15-20 dB in open space. Active noise control specifically for narrow-band noise cancellation using adaptive analog filters seems to be a better solution than its digital signal processing counterpart in speed, cost, and robustness.  相似文献
