A PCR method for the effective detection of Coxiella burnetii in commercially available mayonnaise was developed. Sample preparations were isolated from 50 g portions of each mayonnaise product by four successive extraction steps in phosphate buffer with 2.0 M NaCl. These extracts were then centrifuged at 20,000 x g for 60 min. DNA was isolated from the solution containing the precipitate with a commercial kit, and amplified quantitatively using real-time PCR that targeted the com1 region of C. burnetii. The recoveries of C. burnetii from 2 kinds of commercial mayonnaise specimens, with a baseline control of 1 x 10(7) particles of the Nine Mile phase II strain, were 85.0 +/- 6.0% and 72.0 +/- 0.4%, respectively. The determination limit of this method was 500 C. burnetii particles per 50 g of mayonnaise. The DNA specimens isolated from 50 different commercial mayonnaise samples sold in Tokyo using this method were amplified using both nested PCR and real-time PCR. No contamination by C. burnetii was detected in any of the mayonnaise samples. 相似文献

We applied Pixon deconvolution as introduced in Part I to several practical, examples of low signal-to-noise ratio (SNR), electron energy-loss spectra with a goal toward restoring their fine spectral features and/or improving the energy resolution. We demonstrate that by directly fitting the two-dimensional spectral data recorded on the CCD; the method enables us to reveal fine spectral structures. The friction and wear behaviour of SiC, Si3N4 and SiC/Si3N4 composite ceramics were investigated with oscillating sliding (gross slip fretting) at room temperature. The influence of
counter body material and the humidity of the surrounding air was studied with a ball-on-disc configuration with different
ball materials (1000Cr6, Al2O3 SiC and Si3N4). The effect of RH on friction is marginal with exception of SiC (low friction) as counter body material. The wear behaviour,
however, is strongly affected by humidity, showing inverse trends for different counter body materials. Consequently, the
wear behaviour of a tribo couple can be improved by selecting an adequate mating material. The results reveal the necessity
to control RH in tribological tests. For estimation of the performance of tribo couples under varying environmental conditions,
a variation of RH is required. In tribo couples with single phase SiC, either as ball or disc, the tribological behaviour
of the system is dominated by SiC. The friction behaviour of the composite material is in between the behaviour of the two

The identification of 20 strains of yeasts isolated from foods by means of DNA sequence analysis with two kinds of universal primers for the rDNA region was examined, and the results were compared with those of the conventional phenotyping test using API 20C AUX. In the analysis of the 26S region, all 20 yeast strains tested were identified at the species level. In the ITS1 region, 16 strains were also classified at the species level. In addition, all results of DNA sequence analysis were consistent with those of the phenotyping test at the genus level. Furthermore, DNA sequence analysis was able to identify causative yeasts observed in two suspect foods, though phenotyping tests alone failed to identify them. 相似文献

An octadecylamino-group-introduced polymer chain grafted onto a porous sheet was impregnated with bis(2-ethylhexyl)hydrogen phosphate (HDEHP). A mixture of HDEHP and ethanol of various HDEHP concentrations was used for the impregnation. The porous sheet into which a C18H37NH group was introduced was immersed in HDEHP/ethanol solution before ethanol evaporation. The liquid permeability of a cartridge charged with the HDEHP-impregnated porous sheet in disk form prepared in 50 (v/v)% HDEHP/ethanol solution was 96% that of the starting-porous-disk-packed cartridge. The equilibrium binding capacity of the HDEHP-impregnated porous disk for yttrium ions was 0.32 mol per kg of the disk. In addition, the HDEHP-impregnated-porous-disc-packed cartridge was found to be applicable to the preconcentration of trace amounts of lanthanides in a multielement solution prior to their measurement by inductively coupled plasma mass spectrometry. 相似文献

We consider fixed-size estimation for a linear function of mean vectors from π i : N p (μ i , Σ i ), i = 1,…, k. The goal of inference is to construct a fixed-span confidence region with required accuracy. We find a new sample-size formulation and propose a two-stage estimation methodology to give the fixed-span confidence region satisfying the probability requirement approximately. We show that the proposed methodology greatly reduces the sample size to enjoy the asymptotic first-order consistency when the dimensionality p is extremely high. With the help of simulation studies, we show that the proposed methodology is still efficient even when p is moderate. We give an actual example to illustrate how it should be done by using the proposed methodology in the inference. 相似文献

The electronic structural analysis of the conductive transparent films was carried out using bulk sensitive hard X-ray photoelectron spectroscopy (HAXPES). The In2O3-ZnO film has amorphous structure before and after annealed, and the conduction band spectrum around Fermi level showed the similar spectra with that of as-deposited amorphous In2O3 film. In these amorphous films, the conduction band minimum locates at the deeper level than the crystalline In2O3 film. The electronic state which comes from randomness of amorphous structure possibly exists around this level or below. These electrons are expected to act as scattering center. We concluded that the electron mobility depends on the density of this electronic state. 相似文献

Numerical results of three-dimensional separated flow and heat transfer in a rectangular channel with a suddenexpansion are presented in this paper.Numerical simulations of Navier-Stokes and energy equations are carriedout using the finite difference method.The results of three-dimensional calculations are compared with thetwo-dimensional ones,and effects of the aspect ratio of channel upon the flow are shown.The transition fromsymmetric to asymmetric flow appears at lower Reynolds number as increasing the aspect ratio.The details oflocal heat transfer characteristics in two different separated flow regions on two downstream walls are clarified.Two-dimensionality of the flow and heat transfer almost disappears for the aspect ratio considered. 相似文献
