This study aims at identifying food ingredients that have potential to enhance satiety and therefore potentially can be used for preventing and treating obesity. To do so, effects of thirteen food ingredients on the secretion of gut satiety hormone glucagon-like peptide-1 (GLP-1) in enteroendocrine STC-1 and HuTu-80 cells were investigated. First, the effects of food ingredients on cell viability were investigated. This was done to determine if the individual ingredient affected cell viability, and in case so, to determine the ingredient concentration below which, the cell viability was unaffected. Enzyme-treated whey protein, palmitic acid, stearic acid, green tea extract, and hesperidin did not affect GLP-1 secretion. Functional soy protein, fractionated functional soy protein, acetylated monoglyceride, N-oleoylethanolamine, epigallocatechin-3-gallate, hesperetin, rosemary extract, and kale extract increased GLP-1 secretion in at least one cell line. However, the effects of the eight potent ingredients on GLP-1 secretion need to be further investigated in vivo.  相似文献

Transmission electron microscopy (TEM) makes it possible to obtain insight into the structure, composition and reactivity of photocatalysts, which are of fundamental interest for sustainable energy research. Such insight can be used for further material optimization. Here, we combine conventional TEM analysis of photocatalysts with environmental TEM (ETEM) and photoactivation using light. Two novel types of TEM specimen holder that enable in situ illumination are developed to study light-induced phenomena in photoactive materials, systems and photocatalysts at the nanoscale under working conditions. The technological development of the holders is described and two representative photo-induced phenomena are studied: the photodegradation of Cu?O and the photodeposition of Pt onto a GaN:ZnO photocatalyst.  相似文献

The purpose of this study was to estimate the genetic correlations among claw and leg health and potential indicator traits. Claw health was defined as absence of heel horn erosion, interdigital dermatitis, interdigital phlegmon, interdigital hyperplasia, laminitis, and sole ulcer. Leg health was defined as absence of hock infection, swollen hock, and bruising. The potential indicators were locomotion and foot and leg conformation, represented by rear leg side view, rear leg rear view, foot angle, and apparent hock quality and bone structure. The study was conducted using records from 429,877 Danish Holstein cows in first lactation. Binary health traits were divided into 3 subcategories: claw health, leg health, and absence of all claw and leg disorders. Genetic (rg) and phenotypic correlations were estimated using a bivariate linear sire model and REML. Estimated heritabilities were 0.01 for all 3 combined claw and leg health traits (on the observed binary scale), 0.09 for locomotion, 0.14 for rear leg rear view, 0.19 for rear leg side view, 0.13 for foot angle, 0.22 for apparent hock quality, and 0.27 for apparent bone structure. Heritabilities were 0.06 and 0.01 for claw health and leg health, respectively, when transformed to the underlying continuous scale. Claw and leg disorders are an increasing problem for Danish Holsteins, but genetic improvement of claw and leg health is challenging because the traits have low heritabilities. Claw and leg health were separate but correlated traits (rg = 0.35). Locomotion and rear leg rear view were useful indicator traits for claw health (rg = 0.46 and rg = 0.21, respectively), whereas hock quality and bone structure were useful indicators for leg health (rg = 0.42 and 0.26, respectively). Claw and leg health should be considered as separate traits in genetic evaluations that also include the useful indicator traits to compensate for low heritability of the health traits.  相似文献

Searching for the most rewarding sources of innovative ideas remains a key challenge in management of technological innovation. Yet, little is known about which combinations of internal and external knowledge sources are triggers for innovation. Extending theories about searching for innovation, we examine the effectiveness of different combinations of knowledge sources for achieving innovative performance. We suggest that combinations involving integrative search strategies – combining internal and external knowledge – are the most likely to generate product and process innovation. In this context, we present the idea that cognitively distant knowledge sources are helpful for innovation only when used in conjunction with knowledge sources that are closer to the focal firm. We also find important differences between product and process innovation, with the former associated with broader searches than the latter. Using a large-scale pooled sample of UK firms, we find overall support for our conjectures, particularly in terms of product innovation.  相似文献

The toxic and inflammatory potential of 5 different types of nanoparticles were studied in a sensitive model for pulmonary effects in apolipoprotein E knockout mice (ApoE-/-). We studied the effects instillation or inhalation Printex 90 of carbon black (CB) and compared CB instillation in ApoE-/- and C57 mice. Three and 24 h after pulmonary exposure, inflammation was assessed by mRNA levels of cytokines in lung tissue, cell composition, genotoxicity, protein and lactate dehydrogenase activity in broncho-alveolar lavage (BAL) fluid.  相似文献

Analysis of dyes extracted from textiles of historical interest can give valuable information as to where, when, and how the textiles were made. The most widely used method for extraction of colorants involves heating with HCl, which frequently decomposes glycosidic dye components to their parent aglycons, with consequent loss of information about the source of the dye. This is particularly true for flavonoid dyes, many of which are glycosides. We have developed or improved upon two mild textile extraction methods that use ethylenediaminetetraacetic acid (EDTA) and formic acid and are efficient in extracting dyes, but preserve glycosidic linkages. The relative efficiencies of the HCl, EDTA, and formic acid extraction methods are compared by analyzing extracts of dyed samples of silk using HPLC coupled with diode array and mass spectrometric detection. HPLC profiles of EDTA or formic acid extracts of silk dyed, for example, with pagoda tree buds and onionskins are clearly distinguishable as to the plant material used, whereas profiles of HCl extracts are not. Thus, extraction of textiles with EDTA or formic acid reagents can yield significantly more information about the original dyestuff than can extraction with a strong acid.  相似文献

Multivariate statistical process control (MSPC) based for example on principal component analysis (PCA) can make use of the information contained in multiple measured signals simultaneously. This can be much more powerful in detecting variations due to special causes than conventional single variable statistical process control (SPC). Furthermore, the PCA based SPC simplifies monitoring as it limits the number of control charts to typically two charts rather than one for each signal. However, the derived MSPC statistics may suffer from lack of sensitivity if only one or a few variables deviate in a given situation. In this paper we develop a new comprehensive control (COCO) chart procedure that considers both univariate statistics and multivariate statistics derived from PCA in a single plot that allows easy visualization of the combined data from a univariate and multivariate point of view. The method is exemplified using twenty analytical chromatographic peak areas obtained for purity analysis of a biopharmaceutical drug substance. The new control chart procedure detected two different types of faulty events in this study.  相似文献

Surface-mediated drug delivery is a recent concept, where active surface coatings are employed to deliver therapeutic cargo to cells. Herein, we explore the potential of liposomes embedded in polydopamine (PDA) coatings to serve as drug deposits stored on planar substrates. We quantify the PDA growth rate on glass by XPS and show that PDA coatings support myoblast adherence and proliferation. Further, PDA capping layers were deposited on glass substrates precoated with poly(L-lysine) and zwitterionic liposomes. Already thin PDA capping layers render liposome coated surfaces cell adhesive. We experimentally show for the first time, the internalization of a model hydrophobic cargo, that is, fluorescent lipids embedded within the lipid bilayer of liposomes by the cells from the surface. This is evident from the fluorescence exhibited by the cells grown on PDA coatings containing fluorescently labeled liposomes, with the highest fluorescent intensity found in the close proximity of the cell nuclei. The cargo uptake efficiency depends on the thickness of the PDA capping layer and the cell residence time on the coated substrates. Taken together, we demonstrate the first step toward the establishment of a versatile approach using liposomal drug deposits in polymer thin films for surface-mediated drug delivery.  相似文献
