Immune checkpoint inhibitors, which unleash a patient’s own T cells to kill tumors, are revolutionizing cancer treatment. Several independent studies suggest that higher non-synonymous mutational burden assessed by whole exome sequencing (WES) in tumors is associated with improved objective response, durable clinical benefit, and progression-free survival in immune checkpoint inhibitors treatment. Next-generation sequencing (NGS) is a promising technology being used in the clinic to direct patient treatment. Cancer genome WES poses a unique challenge due to tumor heterogeneity and sequencing artifacts introduced by formalin-fixed, paraffin-embedded (FFPE) tissue. In order to evaluate the data interoperability of WES data from different sources to survey tumor mutational landscape, we compared WES data of several tumor/normal matched samples from five commercial vendors. A large data discrepancy was observed from vendors’ self-reported data. Independent data analysis from vendors’ raw NGS data shows that whole exome sequencing data from qualified vendors can be combined and analyzed uniformly to derive comparable quantitative estimates of tumor mutational burden. 相似文献

In Japan the electricity market will open on April 1, 2004. Electric utility, Power Producer and Supplier (PPS), and Load Service Entity (LSE) will join the electricity market. LSEs purchase electricity based on the Market Clearing Price (:MCP) from the electricity market. LSEs supply electricity to the customers that contracted with the LSEs on a certain electricity price, and one to the customers that introduced Energy Storage System (:ES) on a time‐of‐use pricing. It is difficult for LSEs to estimate whether they have any incentive to promote customers to introduce ES or not. This paper evaluates the reduction of LSEs' purchasing cost from the electricity market and other LSEs' purchasing cost by introducing ES to customers. It is clarified which kind of customers has the effect of decreasing LSEs' purchasing cost and how much MCP of the whole power system the demand‐side energy storage systems change. Through numerical examples, this paper evaluates the possibility of giving the cost merit to both customers with energy storage systems and LSE by using real data for a year's worth of MCP. © 2006 Wiley Periodicals, Inc. 液动射流冲击器在深井模拟实验室的试验结果表明,其在实际应用中具有良好的可行性和应用前景。与此同时,也发现了一些问题,如由于喷嘴过流断面较小等原因导致的射流元件易损坏的问题,所以需要增大喷嘴过流断面。但这种改变必然对射流切换造成影响,其它结构参数必须重新设计。而大量模拟试验表明:增大喷嘴过流断面,调整其它相关结构参数,射流能够实现切换。为此,实际冲击器在设计时增设一阀,将信号道移到中板,改变劈形,排空孔下移且合二为一。重新设计的阀式液动射流冲击器与同规格射流冲击器相比,在冲击功及冲击频率方面相差不大,但压耗大幅度降低。 相似文献
