The objective of this study is to develop a procedure to analyze the motions of a floating pier comprised of several pontoons that are modeled as rigid bodies and connected to each other by flexible and rigid connectors. Recently, the use offloating piers has increased because of their advantages, such as faster and higher-quality construction, seismic force isolation for a full-scale mooring system, low dependence on local soil conditions and tides, ability to relocate or reconfigure the pier modules during the operation period and 75-100 years of repair-free service. A floating pier consists of a pier, access bridge, mooring system and fender system, each of which comes in many variations to suit different usages and construction considerations. The typical loads used in the design of these piers are dead loads, live loads, mooring loads, fender loads and environmental loads induced by wind, currents and waves. For numerical simulation, three types of piers are used: passenger piers, light-cargo piers and semi-heavy-cargo piers. The selected piers consist of several large pontoons joined by pivots and have a pile-based mooring system. These piers are modeled by SAP2000software as two-dimensional frames that are linked together. As the first step, each type of pier is subjected to loading, and its general behavior is assessed. According to this behavior, the major load combinations are described for the design of piers and analyzed to determine the behavior of the modules. Lastly, according to the analysis results and the safe use and stability considerations, such as the maximum draft and longitudinal gradient, the dimensions of each module in each pier type are presented.  相似文献

The present study investigates the mechanical behavior of a new generation of buried pipelines, dubbed the textured pipeline, which is subjected to strike-slip faulting. In conventional cylindrical pipelines, the axial and bending stresses brought about in their walls as a result of fault movement, lead to local buckling, which is construed as one of the major reasons contributing to pipeline failure. The present study has assessed 3-D numerical models of two kinds of buried textured pipelines, with 6 and 12 peripheral triangular facets, subjected to a strike-slip faulting normal to the axis of the pipelines, with and without internal pressure, with the two kinds of X65 and X80 steel, and with different diameter-to-thickness ratios. The results indicate that, because of specific geometry of this pipeline shell which is characterized by having lower axial stiffness and higher bending stiffness, compared to conventional cylindrical pipeline, they are considerably resistant to local buckling. The results of this study can be conceived of as a first step toward comprehensive seismic studies on this generation of pipelines which aim at replacing the conventional cylindrical pipelines with textured ones in areas subjected to fault movement.  相似文献

The 5-HT1B receptor is expressed on nerve terminals where it inhibits neurotransmitter release. When expressed ectopically in fibroblasts, the 5-HT1B receptor inhibits adenylyl cyclase. However, in the central nervous system, the effect of this receptor on neurotransmitter release appears to be cAMP-independent. We therefore investigated alternative effector systems that might be activated by the 5-HT1B receptor. We constructed a recombinant adenovirus that allows expression of high levels of the 5-HT1B receptor in a variety of cells. We chose cardiac ventricle myocytes because they express a muscarinic-gated, inwardly rectifying K+ channel (i[KACh]). In infected ventricle cells, both 5-HT and the muscarinic receptor agonist, carbachol, elicited a similar inwardly rectifying K+ current. The currents elicited by these agonists were pertussis-toxin sensitive and were not additive. These results suggest a common signal transduction pathway for 5-HT1B and muscarinic receptors in ventricle cells.  相似文献

Roll waves create unstable and turbulent behaviour in channels and chutes and also change the behaviour of water entering the energy dissipater structures. In the present study, the free surface of water in the chute is simulated by numerical models. The roll waves’ occurrence is investigated in the numerical model for the discharge up to 30 m3/s, the longitudinal slope varying from 17 to 20% and the width of chute varying from 3.4 to 4.0 m. The ratios of the hydraulic depth to the chute width and also the minimum velocity to the maximum velocity are calculated along the chute length for different discharges, widths and longitudinal slopes of the chute. Results indicated that for the chutes in which the ratio of hydraulic depth to the chute width less than 0.16 the ratio of minimum to maximum velocity is less than 0.75, the formation of roll waves is inevitable.  相似文献

Biofilms are complex structures formed by bacteria, fungi, or even viruses on biotic and abiotic surfaces, and they can be found in almost any part of the human body. The prevalence of biofilm-associated diseases has increased in recent years, mainly because of the frequent use of indwelling medical devices that create opportunities for clinically important bacteria and fungi to form biofilms either on the device or on the neighboring tissues. As a result of their resistance to antibiotics and host immunity factors, biofilms have been associated with the development or persistence of several clinically important diseases. The inability to completely eradicate biofilms drastically increases the burden of disease on both the patient and the healthcare system. Therefore, it is crucial to develop innovative ways to tackle the growth and development of biofilms. This review focuses on dental- and implant-associated biofilm infections, their prevalence in humans, and potential therapeutic intervention strategies, including the recent advances in pharmacology and biomedical engineering. It lists current strategies used to control the formation of clinically important biofilms, including novel antibiotics and their carriers, antiseptics and disinfectants, small molecule anti-biofilm agents, surface treatment strategies, and nanostructure functionalization, as well as multifunctional coatings particularly suitable for providing antibacterial effects to the surface of implants, to treat either dental- or implant-related bacterial infections.  相似文献

The effects of clay (montmorillonite and kaolinite) in the hydrogels were investigated on various properties such as syneresis and strength of thermal and salinity situations in one of the southern Iranian oil reservoirs. The X-ray diffraction (XRD) patterns exhibited a significant increase in interplanar spacing between the montmorillonite clay layers, varying from the initial value of 12.43 °A to 19.45 °A, which evidences the intercalation formation. It was revealed that even increasing of the interlayer spacing due to kaolinite modification had no effect on the clay compositions. Formation water was used to study the strength of the hydrogel in the presence of ions. The results indicated that 15 wt% increase of kaolinite clay (modified and non-modified) leads to 20% decrease of the hydrogels’ syneresis. The diffusion of polymer chains between the clay layers increased the elastic modulus (G′) of the prepared hydrogels with modified kaolinite and montmorillonite, where the maximum value of G’ was observed in 3 wt% of montmorillonite. Finally, the thermogravimetric analysis (TGA) indicated an increase in the thermal stability of the mentioned hydrogels.  相似文献

Spontaneous mutation in the lysosomal acid phosphatase 2 (Acp2) mouse (nax—naked-ataxia mutant mouse) correlates with severe cerebellar defects including ataxia, reduced size and abnormal lobulation as well as Purkinje cell (Pc) degeneration. Loss of Pcs in the nax cerebellum is compartmentalized and harmonized to the classic pattern of gene expression of the cerebellum in the wild type mouse. Usually, degeneration starts in the anterior and posterior zones and continues to the central and nodular zones of cerebellum. Studies have suggested that the p75 neurotrophin receptor (NTR) plays a role in Pc degeneration; thus, in this study, we investigated the p75NTR pattern and protein expression in the cerebellum of the nax mutant mouse. Despite massive Pc degeneration that was observed in the nax mouse cerebellum, p75NTR pattern expression was similar to the HSP25 pattern in nax mice and comparable with wild type sibling cerebellum. In addition, immunoblot analysis of p75NTR protein expression did not show any significant difference between nax and wild type sibling (p > 0.5). In comparison with wild type counterparts, p75NTR pattern expression is aligned with the fundamental cytoarchitecture organization of the cerebellum and is unchanged in the nax mouse cerebellum despite the severe neurodevelopmental disorder accompanied with Pc degeneration.  相似文献

This paper describes the production, purification, and immobilization of l-asparaginase II (ASNase II) in chitosan nanoparticles (CSNPs). ASNase II is an effective antineoplastic agent, used in the acute lymphoblastic leukemia chemotherapy. Cloned ASNase II gene (ansB) in pAED4 plasmid was transformed into Escherichia coli BL21pLysS (DE3) competent cells and expressed under optimal conditions. The lyophilized enzyme was loaded into CSNPs by ionotropic gelation method. In order to get optimal entrapment efficiency, CSNP preparation, chitosan/tripolyphosphate (CS/TPP) ratio, and protein loading were investigated. ASNase II loading into CSNPs was confirmed by Fourier transform infrared (FTIR) spectroscopy, and morphological observation was carried out by transmission electron microscopy. Three absolute CS/TPP ratios were studied. Entrapment efficiency and loading capacity increased with increasing CS and TPP concentration. The best ratio was applied for obtaining optimal ASNase II-loaded CSNPs with the highest entrapment efficiency. Size, zeta potential, entrapment efficiency, and loading capacity of the optimal ASNase II-CSNPs were 340 ± 12 nm, 21.2 ± 3 mV, 76.2% and 47.6%, respectively. The immobilized enzyme showed an increased in vitro half-life in comparison with the free enzyme. The pH and thermostability of the immobilized enzyme was comparable with the free enzyme. This study leads to a better understanding of how to prepare CSNPs, how to achieve high encapsulation efficiency for a high molecular weight protein, and how to prolong the release of protein from CSNPs. A conceptual understanding of biological responses to ASNase II-loaded CSNPs is needed for the development of novel methods of drug delivery.  相似文献

Epoxy composites were prepared with different contents of hydrophobic silica aerogel particles to investigate their mechanical, thermal, and hydrophobic properties. The composites, particularly the one containing 1 wt % silica aerogel, showed remarkable toughness and yielding behaviors compared to neat epoxy, with its brittle behavior. As the content of silica aerogel increased, the thermal properties of the composites (e.g., thermal conductivity and thermal stability) improved. This was due to the very low thermal conductivity and high thermal stability of the silica aerogel particles. Moreover, the use of the hydrophobic silica aerogel led to the development of composites with hydrophobic properties. To examine the hydrophobicity more deeply, a series of water‐uptake tests were performed, and the results show that the composite with 3 wt % silica aerogel absorbed 50% less water than the neat epoxy. © 2017 Wiley Periodicals, Inc.
