Prior to the emergence of life, it is believed that only l-amino acids were selected for formation of protein and that d-amino acids were eliminated on the primitive Earth. Whilst homochirality is essential for life, the occurrence of proteins containing d-β-aspartyl (Asp) residues in various tissues from elderly subjects has been reported recently. Here, we demonstrate the presence of a d-β-Asp-containing protein in the cardiac muscle of heart, blood vessels of the lung, chief cells of the stomach, longitudinal and circular muscle of the stomach, small intestine and large intestine. Since the d-β-Asp residue occurs through a succinimide intermediate, this isomer may potentially be generated in proteins more easily than initially thought. Formation of the d-β-Asp residue in proteins may be related to stress.  相似文献

The present study demonstrates a novel mixing approach for achieving a good dispersion of carbon nanotubes (CNTs) in a styrene-butadiene rubber (SBR), which leads to a significant improvement in electrical properties. Our mixing technique consists of (1) pretreatment by ultrasonication to disentangle the bundles of CNTs in organic solvent and (2) “rotation-revolution” mixing of the CNTs with SBR without mechanical shear, which prevents CNTs from collapsing during the mixing process. The present mixing method does not require the addition of any dispersing agents (amphiphilic molecules) or chemical modification of the CNTs to obtain a good dispersion. Compared with a conventional Banbury mixing technique, our method leads to a significant decrease in the percolation threshold (less than 1 phr), where the electrical conductivity suddenly increases due to the formation of percolation networks of CNTs in SBR. This is because the aspect ratio of the CNTs was maintained even after the mixing process, whereas CNTs were broken during the conventional Banbury mixing. The effect of using different types of CNTs on electrical conductivity was also investigated. The results show that the percolation threshold is largely related to the structural quality (graphitization) of the CNTs as well as their aspect ratio.  相似文献

Catechins are extensively used in health care treatments. Nevertheless, there is scarce information about the feasibility of local administration with polyphenols for bone regeneration therapy, possibly due to lack of effective delivery systems. Here we demonstrated that the epigallocatechin-3-gallate-conjugated gelatin (EGCG/Gel) prepared by an aqueous chemical synthesis using 4-(4,6-dimethoxy-1,3,5-triazin-2-yl)-4-morpholinium chloride (DMT-MM) gradually disintegrated with time and facilitated bone formation in a critical size defect of a mouse calvaria. Conjugation of EGCG with the Gel generated cross-linking between the two molecules, thereby leading to a retardation of the degradation of the EGCG/Gel and to a delayed release of EGCG. The prepared EGCG/Gels represented significant osteogenic capability compared with that of the uncross-linked Gel and the cross-linked Gel with uncombined-EGCG. In vitro experiments disclosed that the EGCG/Gel induced osteoblastogenesis of a mouse mesenchymal stem cell line (D1 cells) within 14 days. Using fluorescently-labeled EGCG/Gel, we found that the fraction of EGCG/Gel adsorbed onto the cell membrane of the D1 cells possibly via a Gel-cell interaction. The interaction might confer the long-term effects of EGCG on the cells, resulting in a potent osteogenic capability of the EGCG/Gel in vivo. These results should provide insight into local controlled release of polyphenols for bone therapy.  相似文献

A macrocomparative analysis of the consonant systems of Indo-European (IE) and Austronesian (AN) language families elucidated 43 word-initial consonant-correspondence laws between proto-IE (pIE) and AN (proto-AN (pAN), proto-Malayo-Polynesian (pMP), MP, Oceanic (OC), etc.). pIE was concluded to have emerged from an eastern MP (E.MP) language closely akin to Bali-Sasak languages. Characteristic features in the emergence of new consonants from pre-existing consonant systems have been elucidated.  相似文献

Corrosion behavior of Fe-3 at.% Si alloy in 0.01 mol dm−3 HCl solution was investigated by using scanning electrochemical microscopy (SECM) as well as general electrochemistry. The rate of corrosion coupled with hydrogen evolution was initially 0.44 A m−2 but decreased significantly with time. Localized hydrogen evolution on the specimen surface was probed by an SECM system in which a force sensor was mounted to determine the probe height from the specimen surface. SECM images revealed that hydrogen evolution took place heterogeneously on the specimen surface depending on crystallographic orientation of substrate single grains in the initial stage and then became relatively homogeneous. Finally, a heterogeneous hydrogen distribution corresponding to the appearance of localized corrosion sites was observed.  相似文献

The irradiation behavior of uranium-plutonium mixed oxide fuels containing a large amount of silicon impurity was examined by post-irradiation examination. Influences of Si impurity on fuel restructuring and cladding attack were investigated in detail. Si impurity, along with Am, Pu and O were transported by spherical pores and cylindrical tubular pores to the fuel center during fuel restructuring of the Np-Am-MOX fuel, where a eutectic reaction of fuel and Si-rich inclusions occurred. After fuel restructuring of the Np-Am-MOX fuel, Si-rich inclusions without fuel constituents were agglomerated at fuel crack openings where shallow attacks on the inner wall of the cladding were seen. Such shallow attacks on the inner wall of the cladding were likewise observed near the location of fuel cracks in long-term steady-state irradiated MOX fuels. Evidence of these shallow attacks on the inner wall of the cladding remained after fuel restructuring in normal MOX fuel. However, grain boundary corrosion of the cladding inner wall at the opening of the fuel cracks was selective and was marked in MOX fuel at higher oxygen potential by the release of reactive fission products such as Cs and Te in comparison with other regions of cladding wall.  相似文献

L ‐Azetidine‐2‐carboxylic acid (AZC) is a toxic four‐membered ring analogue of L ‐proline that is transported into cells by proline transporters. AZC and L ‐proline in the cells are competitively incorporated into nascent proteins. When AZC is present in a minimum medium, misfolded proteins are synthesized in the cells, thereby inhibiting cell growth. The MPR1 gene has been isolated from the budding yeast Saccharomyces cerevisiae Σ1278b as a multicopy suppressor of AZC‐induced growth inhibition. MPR1 encodes a novel acetyltransferase that detoxifies AZC via N‐acetylation. Since MPR1 is absent in the laboratory strain of S. cerevisiae S288C, it could be a positive selection marker that confers AZC resistance in the S288C background strains. To examine the usefulness of MPR1, we constructed some plasmid vectors that harboured MPR1 under the control of various promoters and introduced them into the S288C‐derived strains. The expression of MPR1 conferred AZC resistance that was largely dependent on the expression level of MPR1. In an additional experiment, the galactose‐inducible MPR1 and ppr1+, the fission yeast Schizosaccharomyces pombe homologue of MPR1, were used for gene disruption by homologous recombination, and here AZC‐resistant colonies were also successfully selected. Properties of green and sintered bodies prepared by high-speed centrifugal compaction process (HCP) using wet-jet milled slurries were investigated. At 30 vol.% solids loading, the relative packing density of compacted body prepared from wet-jet milled slurry was 67%. On the other hand, the density of compacted body prepared from ball-milled slurry was 56% at the same loadings. The difference in relative densities of the top and bottom of green bodies prepared from wet-jet milled slurry was decreased to one-third of the ones prepared from ball-milled slurry. Moreover, the linear shrinkage of the sintered bodies prepared from wet-jet milled slurries was almost constant at sintering temperatures above 1400 °C; it was found to be increasing for bodies prepared from ball-milled slurries with increasing sintering temperature. Furthermore, activation energy for grain-growth was estimated. The grain-growth of green bodies prepared from wet-jet milled slurries was promoted at lower-sintering temperatures.  相似文献

During in situ micro-indentation of passive iron in pH 8.4 borate solution, a couple of anodic current peaks emerged; the first peak during loading and the second peak during unloading. The current transients, associated with rupture and repair of the passive film, were influenced by the indentation conditions. For instance, the current peak height, the current peak area and the time required for complete repassivation are strongly dependent on indentation rate. A numerical model was proposed to correlate the current transient during loading with mechanical deformation of the passive surface. The comparison between the current transient measured experimentally and that estimated from the load transient suggested that the ruptured area of passive film was about 10% of the surface area deformed by the contact with the indenter.  相似文献

The Object-Blog service application automatically converts raw sensor data to environment-generated content (EGC), including texts, graphs, and figures. This conversion facilitates data searching and browsing. Generated content can serve several purposes, including memory aids, security, and communication media. In Object-Blog, personified objects automatically post entries to a weblog about sensor data obtained from sensors attached to the objects.
