Cell-matrix interactions have critical roles in regeneration, development and disease. The work presented here demonstrates that encapsulated human mesenchymal stem cells (hMSCs) can be induced to differentiate down osteogenic and adipogenic pathways by controlling their three-dimensional environment using tethered small-molecule chemical functional groups. Hydrogels were formed using sufficiently low concentrations of tether molecules to maintain constant physical characteristics, encapsulation of hMSCs in three dimensions prevented changes in cell morphology, and hMSCs were shown to differentiate in normal growth media, indicating that the small-molecule functional groups induced differentiation. To our knowledge, this is the first example where synthetic matrices are shown to control induction of multiple hMSC lineages purely through interactions with small-molecule chemical functional groups tethered to the hydrogel material. Strategies using simple chemistry to control complex biological processes would be particularly powerful as they could make production of therapeutic materials simpler, cheaper and more easily controlled.  相似文献

Evaluated the impact of spoken language on listeners by examining the effects of prior head injury, speech register, and active vs passive listening among 56 head-injured (HI) and 55 non-HI undergraduates. Ss rated the degree to which they liked the speaker and also the extent to which groups of negative and positive adjectives described the speaker. There were no differences between HI and non-HI Ss in how much they liked speakers using the normal register, but non-HI Ss liked the motherese speakers more than did HI Ss. Raters liked motherese speakers less when addressed directly than when they were not being directly addressed. Hereditary predisposition to thrombosis due to activated protein C resistance (APCR) has been attributed to a missense mutation in the factor V gene at nucleotide 1691 (G to A), causing replacement of arginine at codon 506 with glutamine. Using an RFLP-PCR assay to detect this mutation, we measured a prevalence of 3.3% in healthy Caucasians and 1.25% in healthy African-Americans. In addition, we evaluated a total of 90 consecutive specimens submitted to the coagulation laboratory at the Medical College of Virginia for the presence of this mutation. We compared our results for 78 of these specimens with the values measured by a modified partial thromboplastin assay, the COATEST. Twelve of the 90 samples could not be tested using the COATEST because the patients were undergoing anticoagulant therapy. One of the latter 12 specimens was positive by the RFLP-PCR test. Using the genetic test as the definitive assay and the cutoff value established for distinguishing between normal and abnormal results by the COATEST, the COATEST had a sensitivity of 50% and specificity of 93% for the detection of factor V mutation. Analysis of the 90 samples stratified by ethnic groups revealed a frequency of mutation of 13.3% for Caucasians and 6.88% for African-Americans, although with the present sample size, the difference was not statistically significant. Although the COATEST is technically simpler to perform than the genetic test for diagnosing the presence of the factor V mutation, its use for this purpose is limited due to low sensitivity. Thus where this disorder is clinically suspected, submission of the specimen directly for genetic testing by RFLP-PCR or equivalent assay should be considered.  相似文献

Honey bees readily associate an odor with sucrose reinforcement, and the response generalizes to other odors as a function of structural similarity to the conditioned odor. Recent studies have shown that a portion of odor memory is consolidated in the antennal lobes (AL), where first-order synaptic processing of sensory information takes place. The AL and/or the sensory afferents that project into them show staining patterns for the enzyme nitric oxide synthase, which catalyzes the release of the gaseous transmitter nitric oxide (NO). The results show that pharmacological blockade of NO release impairs olfactory discrimination only when release is blocked before conditioning. Blockade of GABAergic transmission disrupts discrimination of similar but not dissimilar odorants, and does so when the block occurs before condition or before testing. Multiwalled carbon nanotubes have been produced by ethylene catalytic chemical vapor deposition and used to fabricate thick and dense freestanding films (“buckypapers”) by membrane filtering. Field emission properties of buckypapers have been locally studied by means of high vacuum atomic force microscopy with a standard metallic cantilever used as anode to collect electrons emitted from the sample. Buckypapers showed an interesting linear dependence in the Fowler–Nordheim plots demonstrating their suitability as emitters. By precisely tuning the tip-sample distance in the submicron region we found out that the field enhancement factor is not affected by distance variations up to 2 μm. Finally, the study of current stability showed that the field emission current with intensity of about 3.3 × 10?5 A remains remarkably stable (within 5% fluctuations) for several hours.  相似文献

The prevalence and genetic diversity of Listeria monocytogenes in raw broiler legs at the retail level in Estonia were studied. A total of 240 raw broiler legs (120 from Estonia and 120 of foreign origin, which had been imported to Estonia from Denmark, Finland, Hungary, Sweden, and the United States) from 12 retail stores in the two largest cities in Estonia (Tallinn and Tartu) were investigated from January to December 2002. Of these, 70% were positive for L. monocytogenes. The prevalence of L. monocytogenes in broiler legs of Estonian origin (88%) was significantly higher than in broiler legs of foreign origin (53%) (P < 0.001). Altogether, 169 (106 Estonian and 63 imported) L. monocytogenes isolates were characterized by pulsed-field gel electrophoresis (PFGE) typing after treatment with the restriction enzyme AscI. The isolates showed a wide genetic diversity, with 35 different PFGE types obtained. Of these, 11 PFGE types came only from isolates of broiler legs of Estonian origin, 4 of Danish origin, 2 of Finnish origin, and 4 of Hungarian origin. Fourteen PFGE types came from isolates of broiler legs that originated from various countries. The strains that shared the same PFGE types from isolates of Estonian origin were recovered from broiler legs that came from different stores over the course of several months. Seventy-one L. monocytogenes isolates, including all PFGE types, were serotyped, and three serotypes (1/2a, 1/2b, and 4b) were obtained. Serotype 1/2a accounted for 96% of the isolates.  相似文献

A tannins-rich chestnut extract was used to enhance the antioxidant and antibacterial properties of chitosan-based film materials. The favourable mechanical properties of the novel material enabled its application in the preparation of the sachets that were used for packing and storage of filled fresh pasta. The fresh pasta ageing progressed in conventional refrigerated storage conditions of 8 °C with 60 ± 2% relative humidity in the absence of light for 60 days. The rapid moisture mobility between a starchy food and sachets during the first 9 days of storage induced retrogradation of the fresh pasta, whereby total phenolic content show dependency on moisture throughout the shelf life. Active components within the sachet prevented microbial growth on the food surface during the entire 60 days.  相似文献

One hundred percent fruit juice (FJ) contains bioactive compounds with antioxidant activity. As such, this fruit form has the potential to improve antioxidant status and mediate outcomes influenced by redox status. A systematic review of the literature published between 1995 and 2013 was conducted using PubMed database to evaluate associations between intake of 100% FJ and markers of antioxidant/oxidant status and blood lipid levels in healthy, free-living adults ≥18 years. Data extraction and analysis was conducted according to the Academy of Nutrition and Dietetics Evidence Analysis Process. Limited evidence from ten clinical trials meeting inclusion/exclusion criteria suggests potential improvements in a variety of antioxidant or oxidants biomarkers postconsumption of 100% FJ. Weak evidence from five studies suggests that one or more blood lipid measures may be positively influenced by consumption of 100% FJ. Heterogeneity in study methodology including biomarkers, 100% FJ type, dosage, and intervention duration precludes the ability to make evidence-based recommendations regarding a specific dose-duration-juice effect. Key characteristics in study designs were identified which must either be controlled or statistically adjusted for in future investigations in order to obtain a more accurate understanding of the complex relationship between metabolic outcomes and consumption of 100% FJ in context of a healthy dietary pattern.  相似文献

Of 504 tonsil samples from slaughtered fattening pigs, 5.6 % were positive for Listeria monocytogenes by culture after enrichment. The 28 L. monocytogenes isolates were analyzed to gain insights into genetic relationships and virulence-associated traits. Of the 28 isolates, 57 % belonged to serotype 1/2a (genetic lineage II), 25 % to serotype 4b (genetic lineage I) and 18 % to serotype 1/2b (genetic lineage I). These serotypes are commonly associated with human listeriosis cases. Multilocus sequence typing assigned the 28 isolates to 16 clonal complexes (CCs) and three singletons, including one new sequence type (ST768). Several of these CCs were also found in strains from human infections. Sequence analysis of the whole internalin A gene (inlA) showed that all but one isolate (CC6/serotype 4b) encoded full-length proteins. Clinical strains from human patients commonly harbor full-length inlA. On the other hand, genes for benzalkonium chloride tolerance were not found and all but one isolate lacked genes for the stress survival islet (SSI-1). Thus, tonsils of slaughtered fattening pigs can be colonized with L. monocytogenes of public health impact. To counteract this threat during slaughter, prevention of contamination of carcasses and the environment is of major importance, in particular adherence to good slaughter hygiene practice. With regard to pig tonsils, special attention must be given to the handling and contamination of head meat and pig tongues.  相似文献
