OBJECTIVE: To examine the changes in neutrophil deformability, aggregation, and adherence in response to stimulation with proinflammatory cytokines and bacterial toxins. DESIGN: Prospective, randomized trial. SETTING: Research laboratory. SUBJECTS: Neutrophils isolated from healthy volunteers. INTERVENTIONS: Neutrophils were exposed to tumor necrosis factor (TNF)-alpha, interleukin (IL)-1beta, IL-8, their combination, endotoxin (LPS), lipoteichoic acid (LTA), and staphyloccocal enterotoxin B (SEB). Neutrophil deformability was measured as percent neutrophils filtered through 5-microm diameter filters. Aggregation was measured using a platelet aggregometer. Adherence was determined by examining the binding of neutrophils to albumin-coated latex beads. MEASUREMENTS AND MAIN RESULTS: Exposure to TNF-alpha and IL-1beta led to significant decreases in neutrophil filterability, which was attenuated by cytochalasin D pretreatment. LPS and LTA also decreased deformability, suggesting that these toxins directly stimulated neutrophils independent of cytokines. IL-8 and SEB did not significantly affect neutrophil deformability. TNF-alpha and LPS were associated with significant neutrophil aggregation, which was inhibited by pretreatment with anti-CD18 antibodies. Neutrophil aggregation was not affected by IL-1beta, LTA, or SEB. TNF-alpha, IL-8, and LPS increased neutrophil adherence, which also was attenuated by pretreatment with anti-CD18 antibodies. IL-1beta, LTA, and SEB did not significantly affect neutrophil adherence. CONCLUSIONS: Cytokines and bacterial toxins differ in their effects on neutrophil deformability, aggregation, and adherence. Of the cytokines examined, TNF-alpha appears to have the greatest direct effects on neutrophil rheology. Similarly, endotoxin appears to have greater direct effects on neutrophil rheology than the Gram-positive bacterial toxins, LTA, and staphylococcal enterotoxins. 相似文献

The finite strip method based on the higher-order plate theory is developed for determining the natural frequencies of laminated plates. This method can accurately predict the through thickness effect of transverse shear deformation. Furthermore, only a few degress of freedom are required in the finite strip method. Some numerical results for various span-to-thickness ratios, material properties and stack sequences are presented for illustrative purposes. A new method of fabricating a-Si:H TFT with etching-stop structure has been proposed. Only one plasma-enhanced chemical vapor deposition is required in this new method and a PH3/H2 plasma treatment during the deposition has been used to form the TFT contact and thus saved another plasma deposition. With this method, a TFT of 500 Å active layer has been fabricated successfully. The drain current and saturation mobility of this device is 2.4×10-7 A and 0.1 cm2/V sec, respectively, which is comparable to the conventional fabricating method. The plasma treatment will also form an additional leakage path on the TFT top surface and increase the TFT subthreshold slope. However, a current of less than 1 pA at VG=-2.4 V can still be obtained. Smokeless tobacco use is a significant health risk for carcinoma of the oropharynx, and has a high prevalence in the southern United States. To evaluate the potential demand for a smokeless tobacco cessation program, we surveyed patients attending the General Medicine Clinic of a Veterans Affairs Medical Center. The prevalence of smokeless tobacco use was 5.8%, which did not justify creation of a clinic-based cessation program. Since we observed adults using smokeless tobacco as a means to reduce or stop their smoking, smoking cessation programs should include counseling against smokeless tobacco use. 相似文献

Morphology and geometry of melted zones, cooling rates, microstructure and microhardness in the laser-glazed Fe-4%C-10%Sn alloy have been investigated. The computer simulation on the basis of the moving gaussian source model was used successfully to predict the maximum width and depth of the melted zone and the cooling rate. The microstructure from the surface to the bottom of the laser-melted zone is a non-crystalline phase, dendritic grains and a microcrystalline zone successively. Values of the averaged-spacing of the non-crystalline phase are 0.2056 and 0.1219nm, respectively; twinned martensites, having an axial ratioc/a of 1.128, existed in dendritic grains, and carbides of Fe3 C at the interdendritic regions; the microcrystalline zone was composed of -Fe and a new bet (a=0.415 nm,c=0.955 nm) phase. The different microstructure in the melted zone can be explained by the results of the heat flow calculation. A fine eutectic structure (-Fe + Fe3C) was observed in heat-affected zones. Microhardness of the eutectic structure can be predicted by the empirical relation of fracture stress to the interlamellar spacing of pearlite. 相似文献

A new bioglass ceramic with composition of Na2O 12%, CaO 28%, P2O5 10% and SiO2 50% with a high bending strength (120–140 MPa) and compressive strength (600–750 MPa) was studied. The crystallized phases of-Ca2P2P7 and Na2Ca3Si6O16 were determined by X-ray diffraction. Optical microscopy of the material revealed that a very uniform crystal size of about 30 m was obtained with high nucleation frequency. The nucleation and crystallization processes were also investigated.
