AbstractIn the context of diseases of affluence, western diets have in the past years mainly been studied on their fat and sugar content and lack of dietary fiber. Yet, the more general aspect of food processing has recently sparked scientific interest as well. In addition, the gut microbiota have been put forward as an important link between diet, obesity and non-communicable diseases (NCD). Western dietary patterns, containing large amounts of processed foods might create an imbalance in the gut system by affecting gut bacteria and their metabolism. Here we discuss what has been already published regarding the relationship between several recently researched features of processed foods and the etiology of obesity and NCD. The addressed features concern micronutrient and energy density, several types of food additives and the generation of advanced glycation end products by thermal treatment during food processing. Overall, literature indicates that all discussed aspects can be linked to western ailments and that they can have a potential negative impact on human microbiota. Therefore, we propose that the thesis that a distressed gut microbiota is a mechanism that might explain how food processing features could harm human health is gaining empirical evidence. Future research will need to address the question whether the alteration of the gut microbiota is a direct or an indirect (via the host) effect. Zinc represents an essential microelement for several biochemical mechanisms. The body's inability to store zinc necessarily requires a constant dietary supply to avoid alteration of physiological functions. The aim of the present study was to investigate the effect of dietary enrichment with zinc on chemical-nutritional and aromatic properties of milk and cheese. Thirty commercial dairy cows, balanced for parity, milk production, and days in milk, were randomly assigned to 2 groups. The control group was fed with a conventional complete diet (22 kg of dry matter/animal per day), whereas the experimental group received a daily zinc supplementation of 60 mg per kg of dry complete feed. During the experimental period, the milk yield was monitored and samples of milk and caciotta cheese were collected to obtain information about the chemical-nutritional composition and aromatic profile. Dietary zinc integration did not influence milk yield and composition, but induced a marked reduction of somatic cell count and improved the oxidative stability of ripened caciotta cheese. In both milk and cheese, the experimental group samples were characterized by a lower concentration of saturated fatty acids and an increase in oleic acid, vaccenic acid, and rumenic acid. The aromatic profile of dairy products was also positively affected by dietary zinc intake, with an increase in concentration of carboxylic acids, aldehydes, and esters. The present results suggest a positive role of zinc in improving animal health and nutraceutical properties of milk and corresponding cheese. Taking into account the analysis of volatile compounds, zinc dietary supplementation of dairy cows should contribute to the production of cheeses with interesting organoleptic properties, although more studies are necessary to confirm the consumer acceptability of these changes. 相似文献

A flow cytometric method (RAPID-B™) with detection sensitivity of one viable cell of Escherichia coli serotype O157:H7 in fresh spinach (Spinacia oleracea) was developed and evaluated. The major impediment to achieving this performance was mistaking autofluorescing spinach particles for tagged target cells. Following a 5 h non-selective enrichment, artificially inoculated samples were photobleached, using phloxine B as a photosensitizer. Samples were centrifuged at high speed to concentrate target cells, then gradient centrifuged to separate them from matrix debris. In external laboratory experiments, RAPID-B and the reference method both correctly detected E. coli O157:H7 at inoculations of ca. 15 cells. In a follow-up study, after 4 cell inoculations of positives and 6 h enrichment, RAPID-B correctly identified 92% of 25 samples. The RAPID-B method limit of detection (LOD) was one cell in 25 g. It proved superior to the reference method (which incorporated real time-PCR, selective enrichment, and culture plating elements) in accuracy and speed. 相似文献

The determination of polycyclic aromatic hydrocarbons (PAHs) in human milk is relevant in assessing the dose to mothers and infants. Since measurements for the United States are lacking, we analyzed 13 PAHs in human milk from 12 nonsmoking American mothers. Analytes were extracted from milk by using a combination of centrifugation, liquid-liquid, and solid-phase extraction techniques. Following method validation with spiked bovine milk, PAH concentrations in human milk were determined by isotope dilution gas chromatography/mass spectrometry. Phenanthrene, fluorene, fluoranthene, and pyrene were found at median concentrations of 11.8, 3.0, 1.6, and 1.1 ng/g milk fat, respectively, whereas 9 additional analytes including benzo[a]pyrene were not detected (<0.04 ng/g). Calculated average daily doses from ingestion (high: 75.8, phenanthrene; low: 6.1 ng/kg/day pyrene) were compared to doses from air pollution inhalation. Milk ingestion accounted for 47.9-81.4% of the total dose of individual PAHs to the infant. However, calculated total average daily doses of phenanthrene and pyrene from ingestion and inhalation combined (12.2 to 110.3 ng/kg-day, respectively) were 3 orders of magnitude lower than documented doses known to cause health effects. The dissolution of UO(2) in a continuously stirred tank reactor (CSTR) in the presence of Ca(2+) and Zn(2+) was investigated under experimental conditions relevant to contaminated groundwater systems. Complementary experiments were performed to investigate the effect of adsorption and precipitation reactions on UO(2) dissolution. The experiments were performed under anoxic and oxic conditions. Zn(2+) had a much greater inhibitory effect on UO(2) dissolution than did Ca(2+). This inhibition was most substantial under oxic conditions, where the experimental rate of UO(2) dissolution was 7 times lower in the presence of Ca(2+) and 1450 times lower in the presence of Zn(2+) than in water free of divalent cations. EXAFS and solution chemistry analyses of UO(2) solids recovered from a Ca experiment suggest that a Ca-U(VI) phase precipitated. The Zn carbonate hydrozincite [Zn(5)(CO(3))(2)(OH)(6)] or a structurally similar phase precipitated on the UO(2) solids recovered from experiments performed in the presence of Zn. These precipitated Ca and Zn phases can coat the UO(2) surface, inhibiting the oxidative dissolution of UO(2). Interactions with divalent groundwater cations have implications for the longevity of UO(2) and the mobilization of U(VI) from these solids in remediated subsurface environments, waste disposal sites, and natural uranium ores. 相似文献

Common methods for assessment of surface checking in decorative plywood panels rely on manual handling and visual inspection of specimens, a laborious procedure practically limiting the number of materials and variables that may be considered within one project. In this study, a new automated optical method for detection and measurement of checks has been developed. This method was based on the digital image correlation principle, which allowed identification of checks as small as 0.2 mm wide and 1 mm long. Continuous measurement allowed reliable check counts, and measurement of check dimensions as they develop during exposure to drying conditions. A check severity index has been proposed. The method has been validated in exposure tests conducted in harsh but realistic conditions, to increase the likelihood of checking and reduce the test duration to 4 h. In addition, an innovative test setup allowed near simultaneous monitoring of check development in up to 48 panel specimens sized 30?×?30 cm. The efficiency of the method allows studies to examine an unprecedented number of treatments and replicates. 相似文献
