The activation of transducin (T) by photoexcited rhodopsin (R*) is kinetically dissected within the framework of Michaelis-Menten enzymology, taking transducin as substrate of the enzyme R*. The light scattering "release" signal (Vuong, T.M., M. Chabre, and L. Stryer, 1984, Nature (Lond.). 311:659-661) was used to monitor the kinetics of transducin activation at 20 degrees C. In addition, the influence of nonuniform distributions of R* on these activation kinetics is also explored. Sinusoidal patterns of R* were created with interference fringes from two crossed laser beams. Two characteristic times were extracted from the Michaelis-Menten analysis: t(form), the diffusion-related time needed to form the enzyme-substrate R*-transducin is 0.25 +/- 0.1 ms, and T(cat), the time taken by R* to perform the chemistry of catalysis on transducin is 1.2 +/- 0.2 ms, in the absence of added guanosine diphosphate (GDP) and at saturating levels of guanosine triphosphate (GTP). With t(form) being but 20% of the total activation time t(form) + t(cat), transducin activation by R* is not limited by lateral diffusion. This is further borne out by the observation that uniform and sinusoidal patterns of R* elicited release signals of indistinguishable kinetics. When (GDP) = (GTP) = 500 microM, t(cat) is lengthened twofold. As the in vivo GDP and GTP levels are comparable, the exchange of nucleotides may well be the rate-limiting process. 相似文献
Atomic Energy of Canada Limited and Ontario Hydro are conducting a research program to assess the environmental impact and safety of the concept of disposal of nuclear fuel waste in an underground vault in plutonic rock of the Canadian Shield. The Vault Sealing Program, one of the components of the Nuclear Fuel Waste Management Program, is concerned with the development of materials specifications and emplacement procedures for backfilling the vault. Backfilling materials would surround the nuclear waste containers and fill all vault openings to minimize leaching and movement of radionuclides. This paper presents the procedure followed to select candidate backfill materials, the integration of mathematical modelling studies and physical testing for the definition of materials specifications, and the principal elements of the recommended handling and emplacement systems. 相似文献
A comparison was made between three API grade pipeline steels (X60, X80 and the X100 grade) from the point of view of their susceptibility to hydrogen embrittlement. The main aim was to determine whether the development of higher strength materials led to greater susceptibility to hydrogen embrittlement. This was achieved by straining at 2.8 × 10−5 s−1 after cathodic charging. The results showed that there is a distinct susceptibility to loss of ductility after charging and this tends to increase with the strength level of the steel at a charging current density above 0.44 mA mm−2. All three steels exhibited fine cracks parallel to the major rolling direction after charging and an increasing amount of brittleness on the fracture surface. 相似文献
Radio frequency identification (RFID) is a powerful technology that enables wireless information storage and control in an economical way. These properties have generated a wide range of applications in different areas. Due to economic and technological constrains, RFID devices are seriously limited, having small or even tiny computational capabilities. This issue is particularly challenging from the security point of view. Security protocols in RFID environments have to deal with strong computational limitations, and classical protocols cannot be used in this context. There have been several attempts to overcome these limitations in the form of new lightweight security protocols designed to be used in very constrained (sometimes called ultra‐lightweight) RFID environments. One of these proposals is the David–Prasad ultra‐lightweight authentication protocol. This protocol was successfully attacked using a cryptanalysis technique named Tango attack. The capacity of the attack depends on a set of boolean approximations. In this paper, we present an enhanced version of the Tango attack, named Genetic Tango attack, that uses Genetic Programming to design those approximations, easing the generation of automatic cryptanalysis and improving its power compared to a manually designed attack. A key component of a hydrogen fuel cell is a catalyst to dissociate dihydrogen to hydrogen atoms. In the present study, the adsorption of hydrogen on Pt/C fuel cell catalysts has been investigated by inelastic neutron scattering spectroscopy.
Monitoring a clean Pt(50%)/C catalyst with low energy neutron spectroscopy, after exposure to dihydrogen at 20 K, as it was heated to room temperature, showed three distinct temperature regimes: (i) a decrease in intensity from 10 to 60 K, (ii) a rise to a maximum between 60 and 120 K and then (iii) a slow fall-off towards room temperature. We assign the three regions as: (i) desorption of physisorbed dihydrogen, (ii) dissociation of dihydrogen to give an adsorbed layer and (iii) damping of the response by an increasing Debye–Waller factor.
The vibrational INS spectra of a series of Pt/C catalysts prepared under varying conditions were similar indicating that the same types of site are common to all the catalysts, although the relative proportions of each site are sample dependent. Features at 520, 950 and part of the intensity at 1300 cm−1 are assigned to hydrogen on (1 1 1) faces, in good agreement with single crystal data. The mode at 640 cm−1 is assigned as the doubly degenerate asymmetric stretch of Pt(1 0 0) faces with the symmetric stretch near 550 cm−1.
We assign the bending mode of the on-top site to the feature at 470 cm−1. The Pt–H stretch mode was observed at 2079 cm−1. This is a significant result: this is the first time that hydrogen on the on-top sites has been observed on nanosized platinum particles supported on high surface area carbon black. The width of the INS peak is surprisingly large and may give additional information on the type and relative proportions of the crystallographic faces present on the catalyst particles. 相似文献
The sorption equilibrium of dissolved copper by spherical partially-coagulated gels of calcium alginate was investigated in this work. The gels were formed by dispensing a viscous algin (food grade sodium alginate from kelp) solution with a multi-tip dispenser into 0.05 M CaCl2 solution in a loop fluidized bed reactor. The resultant semi-rigid spherical gels were then transferred to another reactor operated batch wise to absorb dissolved copper at low concentrations (10-40 ppm). When the concentration of the inert neutral salt NaNO3, added to the reactor fluid was 0.01 M, the amount of copper absorbed was found to be substantially higher than that at 0.1 MNaN03. The conventional Langmuir's model based on the concentration of copper in solution yielded different values of conditional stability constant at different ionic strengths in the reactor fluid. However, by defining the copper-binding stability constant on the basis of copper activity in the gel phase with the competition from calcium for metal binding sites taken into account, a unique copper-binding stability constant and a unique calcium-binding stability constant were obtained. We present an interactive approach for decompositing bitmap drawings and studio photographs into opaque and semi‐transparent vector layers. Semi‐transparent layers are especially challenging to extract, since they require the inversion of the non‐linear compositing equation. We make this problem tractable by exploiting the parametric nature of vector gradients, jointly separating and vectorising semi‐transparent regions. Specifically, we constrain the foreground colours to vary according to linear or radial parametric gradients, restricting the number of unknowns and allowing our system to efficiently solve for an editable semi‐transparent foreground. We propose a progressive workflow, where the user successively selects a semi‐transparent or opaque region in the bitmap, which our algorithm separates as a foreground vector gradient and a background bitmap layer. The user can choose to decompose the background further or vectorise it as an opaque layer. The resulting layered vector representation allows a variety of edits, such as modifying the shape of highlights, adding texture to an object or changing its diffuse colour. 相似文献
The application of Internet-enabled devices in the real world for the development of Smart Cities, environmental monitoring, bus tracking, and parking requires scalability, extensibility, and integration of emerging resources to reach a suitable ecosystem for data acquisition and interaction with citizens. Internet of things needs to offer efficient support for global communications and access to services and information. It needs to enable homogeneous and seamless machine-to-machine communication for different solutions and applications. This work presents an homogeneous and suitable mechanism for global resource discovery, device access for deployed smart objects in different scenarios, and sensors and devices from end users (participative sensing). The integration of legacy and sensors already available from smart buildings and smart objects is presented. For this purpose, a resolution infrastructure called “digcovery” is defined for maximizing efficiency and sustainability of deployments. Digcovery architecture offers the framework to allow users to register/include their own sensors into a common infrastructure and access/discover the available resources through mobile digcovery. Mobile digcovery exploits the context-awareness, geo-location, and identification technologies available in mobile platforms such as smartphones to discover, interact, and access the resources through its ElasticSearch engine. 相似文献
The 96 million gallon per day Yuma desalting plant is part of a complex project intended to solve a long standing water quality problem between the United States and Mexico. Designed to treat up to 129,000 acre-feet of 3200 mg/l irrigation return flows, the plant will deliver 283 mg/l product water to maintain a 115 mg/l differential between water delivered to U.S. users and to Mexico .
Design of the plant was preceded by an extensive research program to determine pretreatment requirements, membrane performance, plant capacity and equipment and instrumentation performance. Two pretreatment systems were tested. Partial lime softening followed by multi-media filtration was chosen for the final design. Nine membrane systems (both reverse osmosis and electrodialysis) were tested. Two spiral wound reverse osmosis systems were chosen as most efficient from physical and economic viewpoint, and contracts for site preparation and construction of the intakes and sedimentation basin is scheduled for award summer 1979 with construction of the main plant and installation of desalting equipment scheduled for award summer of 1980.
Based on January 1979 prices, total cost of the desalting complex is approximately $190 million (including interest during construction). The investment cost per daily gallon of installed capacity is approximately $2. per 1000 gallons of desalted water (capital amortization plus operation and maintenance) is $0.77. 相似文献
The deformation of the dispersed phase in polystyrene/high density polyethylene (PS/HDPE) blends produced by ribbon extrusion was studied numerically and experimentally. A mathematical model for the deformation of the dispersed phase in ribbon extrusion processing of polymer blends was developed assuming uniaxial deformation of the ribbon and the equilibrium shapes of the dispersed particles with a pressure balance over a drop. Simulated morphologies as function of the post‐extrusion parameters were obtained and compared with experiments. The analysis of the ribbon extrusion process showed that parameters such as draw ratio (DR) and ribbon‐water contact length (X) significantly influence the ribbon dimensions, the extensional stress, and the stretching force. The results also showed that deformation and coalescence of the dispersed phase in the ribbon extrusion processing of polymer blends increase at higher DR and/or lower X values. The comparison between the model and the experimental morphologies of PS/HDPE produced a good agreement. 相似文献
